Four cases by high voltage electricity are reported. Particular interest is taken in describing initial conservative treatment of injuries where the entrance of the current has been through the head, followed by surgical excision and flap surgery. This procedure had an uneventful postoperative course. Other methods comprising an earlier excision and revitalizing flap coverage over the necrotic bone thereby avoiding later bone grafting are being discussed and may be preferable. A decrease in high voltage injuries has been noticed during the last ten-year period compared to earlier data (5.5% down to 1.4%). This may well be partially explained by improved precautionary measures, but children are still prominent among the injured. All the children were injured while playing around railroad areas.
High voltage electronmicroscopy was applied to the study of presynaptic nerve terminals and specifically the arrangement and staining properties of synaptic vesicles in and around the active zone. The zinc iodideosmium tetroxide technique was used for the impregnation of neuropil derived from the spinal cord of anesthetized (40 mg/kg pentobarbital sodium) and unanesthetized rats. Under optimal conditions of section thickness and tilt angles stereoscopic views of a paracrystalline lattice arrangement of synaptic vesicles could be ascertained. The ZIO affinity was significantly higher in synaptic vesicles of the anesthetized as compared with the unanesthetized preparations.
High-voltage transmission electron microscopy and cryo-ultramicrotomy together with scanning electron microscopy and some conventional transmission electron microscopy of ultrathin sections have been applied to the mucous surfaces of bovine olfactory and respiratory epithelia. Distal segments of olfactory cilia tend to run in parallel and could be followed over distances up to about 30 micrometer using high-voltage electron microscopy. This technique and scanning electron microscopy showed that on average 12--13 of such cilia could be observed per nerve ending. After correction for obscured cilia this number becomes about 17. High-voltage micrographs and micrographs made from sections prepared with a cryo-ultramicrotome showed the presence of electron-lucent pockets inside the olfactory mucus. The latter technique also showed that the mucus itself is not fibrous, but rather a continuum varying in electron density. The mucus layer contains various granular structures. Ciliary and microvillar membranes appear thicker with cryo-ultramicrotomy than when the sections are prepared with conventional techniques. The cores of the axonemal microtubules in olfactory as well as in respiratory cilia are darkly stained with this technique. Vesicles present inside the nerve endings are also darkly stained. Dimensions and some other numerical values of interest in olfaction are presented.
Keratinocytes and melanocytes cultured from guinea-pig epidermis were studied with scanning, transmission and high voltage electron microscopy to characterize the surface and internal morphology. Keratinocytes exhibited contact-inhibition and a range of surface structures consistent with cell-cycle dependent changes. Stereoscopic analysis of high voltage electron micrographs indicated regular oval nuclei with nucleoli at different depths, while thin sections revealed local channels in the nuclei. Secondary cultures differed from primary cultures in the disorder of the microfilaments, in the failure to form desmosomes, and in the failure of melanocytes to persist in culture. The beaded surface of melanocytes was indicative of underlying melanosomes that were seen in high voltage micrographs. Melanocytes were rounded with moderate ruffles or were dendritic with ruffles on the termini. These findings are discussed in relation to the observational techniques and in relation to modes of locomotion of and pigment transfer to epidermal cells.
The ultrastructure of the normal median eminence of the male rhesus monkey (Macaca mulatta) is described using high-voltage electron microscopy. Surface specializations of ependymal cells lining the infundibular recess included cilia, apical extrusions, and microvilli. Supraependymal cells were predominantly macrophage-like, but examples of lymphocytic types were also seen. Tanycytes had long, branching, basal processes filled with numerous microtubules, some lipid droplets, and granules. The zona interna was composed of large unmyelinated neurosecretory fibers. A few myelinated fibers were also seen, but their character as neurosecretory fibers could not be established. The zona externa was composed of densely-packed profiles of neuro secretory fibers of small diameter, was well-vascularized and contained the terminations of tanycytes. Perivascular glial cells, vesiculated elements, pituicytes, and cellular elements common to connective tissue were observed. The intricate relationships between both the cellular and fibrous elements of the median eminence can be appreciated with the capability of high-voltage electron microscopy to discern ultrastructure in sections 10 times thicker than those used for low-voltage electron microscopy. The median eminence of this primate species has an ultrastructural organization similar to that described for most other species.
A procedure using preparative free-flow high voltage electrophoresis is described for the fractionation of murine spleen and bone marrow cells so as to obtain cell subpopulations that are either enriched in or depleted of "natural killer" (NK) cells and "mitogen-induced cellular cytotoxicity" (MICC) effector cells. A nearly three fold enrichment in the NK and MICC activities of spleen cells was achieved. The enrichment in these cells could be further increased if the phagocytic cells were removed prior to electrophoresis. When bone marrow cells were fractionated a two and a half fold increase of NK activity, and a one and a half fold enrichment of MICC activity was achieved. In both cases, other fractions were nearly devoid of NK and MICC activity. The cell recovery after electrophoresis averages 70% of the cells applied, and at least 90% of these cells were viable. MICC and NK effector cells could not be separated to a useful extent electrophoretically but were found to be separable using Sephadex C-10 gel filtration columns. The MICC but not the NK cells were retained on these columns.
Nickel-based layered cathodes are promising candidates for high-performance, high-energy lithium-ion batteries, yet their high-voltage application is jointly limited by synthesis-inherited structural defects and an unstable lattice oxygen framework. Here, we show that both limitations can be overcome by decoupled synthesis pathway (DSP) via La/Nb oxalate functionalization of the Ni0.6Co0.1Mn0.3(OH)2 precursor. Unlike the conventional coupled synthesis pathway (CSP) where precursor dehydration and Li2CO3 decomposition overlap in temperature, the DSP introduces a low‑temperature decomposition of La/Nb oxalates at 200°C, which effectively avoids localized contact between the precursor and Li2CO3 and shifts Li2CO3-related reactions to high temperatures. This allows sequential precursor dehydroxylation, rock‑salt (RS) intermediate formation, and layered‑phase transformation over a broad temperature window. The resulting LiNi0.6Co0.1Mn0.3O2 cathode with La/Nb functionalization (NCM-LN) features a uniform surface LaNiO3 perovskite heterostructure and a Nb‑doped layered bulk with suppressed RS and spinel defects. Consequently, under 4.5 V operation (vs. Li+/Li), NCM-LN exhibits homogeneous Li+ (de)intercalation, and a stabilized oxygen framework. In graphite||NCM-LN full cells, NCM-LN retains 80.1% of its capacity after 2000 cycles at 1C, substantially outperforming the pristine cathode. This decoupling strategy is broadly effective across various Ni‑based systems, providing a generalizable route toward high‑energy, long‑life cathode materials.
High-voltage paper electrophoresis of small samples of serum and urine at pH 6.0 resolves basic and acidic amino acids and separates them from the neutral amino acids. For separation and identification of the neutral amino acids, the appropriate area of the electrophoretogram is cut out, sewn onto a second sheet of paper, and rerun at pH 1.9. By this method, amino acids are rapidly resolved. It is suited for use with special procedures such as oxidation of biological fluid with performic acid and specific staining for confirmation of amino acid identification.
External high-voltage radiotherapy of carcinoma of the prostate and the field of regional lymphatic spread leads to regression of the tumour in over half the cases. The definite response to radiotherapy does not seem to be related either to the degree of histological differentiation or to the palpable extent of the tumour in the pelvis and is not always seen until at least 6 months from the end of treatment. Radiotherapy has a good palliative effect. Oestrogens need not be administered during irradiation and subsequently as long as no disease activity is detectable.
Two cases are reported in which reversible deep coma subsequent to high voltage electrical injury occurred in association with alpha frequencies in the EEG. The EEG pattern differs from the alpha rhythm of the normal awake patient by its diffuse distribution and unresponsiveness to a variety of stimulation. The term "alpha-pattern coma" is introduced to designate the occurrence of this pattern in comatose patients. The EEG in the cases described initially demonstrated activity in the alpha frequency which occurred in a generalized distribution and was unresponsive to stimulation. During the early stages of recovery an increased incidence of theta and delta activity was observed. The recovery records contained a low voltage alpha rhythm and responded to photic stimulation. The literature on this subject is reviewed and the possible modes of pathogenesis are discussed. The authors conclude that such examples of alpha-pattern coma are the result of diffuse cerebral damage and might be detected more frequently in comatose patients who survive if these patients are studied earlier in their clinical course with EEGs.
A high-voltage electron microscopic study of adrenal medullary cells from hypoglycemia-stressed rats revealed the existence of tubular channels which create a luminal continuity between the mitochondrial compartment and the catecholamine-storage vesicles. It is suggested that these channels allow for the transfer of materials such as high-energy nucleotides between the mitochondria and the catecholamine-storage vesicles without an intervening membrane.
Pulmonary changes following irradiation after post-operative high voltage therapy of the carcinoma of the breast is usually greater than following orthovoltage treatment. Among our own 635 patients, treated post-operatively with telecobalt including the chestwall, pneumonitis occurred in 35%. The data published in the literature and our own show the need for refined techniques of irradiation and individual planning.
Human liver alcohol dehydrogenase isozyme patterns were studied using prolonged high voltage starch-gel electrophoresis and gel-slab isoelectric focusing. Homo- and heterodimers of ADH2 locus were easily distinguished from each other. The gene frequencies of ADH2 2 and ADH2 3 in 46 random liver samples from Germany were found to be 0.044 and 0.424 respectively.
A method for the high-voltage electrophoresis of dihydrofolate reductase from Escherichia coli W 3110 is described. Dihydrofolate reductase catalyses the reduction of dihydrofolic acid to tetrahydrofolic acid. By addition of a tetrazolium salt, tetrahydrofolic acid reacts by formation of a violet water insoluble formazane which is an indicator for the enzyme. Besides several unspecific bands, two isoenzymes of the dihydrofolate reductase from Escherichia coli W 3110 are found which are specificially inhibited by the folate antagonists methotrexate and trimethoprime in a concentration of 0, 1muM, 1 muM respectively.
Reconstruction of the oral cavity from 17 0.5 mum thick serial sections observed with a high voltage electron microscope (JEM-1000, operated at 1000 kV) has enabled us to describe the in situ shape of the cavity, the orientation of the membranelles, oral ribs, cytostomal lip and forming food vacuole of Tetrahymena pyriformis. The study also showed that many different sets of microtubules encircle the oral cavity forming an interwoven, basket-like structure around the cavity thus providing it with considerable structural rigidity. By correlating results obtained from the reconstruction with results obtained from scanning electron microscopy and freeze-fracturing we have been able to elucidate the probable mechanism of how food particles are propelled into and through the oral cavity.
A high-voltage electron microscope was used to study the structure of the centroplast and axonemes in the centrohelidian Heliozoan Raphidiophrys ambigua. In Raphidiophrys the microtubules (Mts) of each axoneme are arranged so that they describe the apices of an array of hexagonally shaped prisms and all axonemes originate from a centroplast located near the centre of the cell. The centroplast was found to consist of 3 components: (1) a tripartite central disk, (2) a sphere of electron-opaque material, approximately 0.75 micron in diameter, which immediately surrounds the central disk, and (3) a slightly less electron-opaque centroplast shell, approximately 2-3 micron in diameter which consists primarily of hitherto undescribed components associated with the proximal 1-2 micron of each axonemal base. The new axonemal components described in this study include: (1) a 20-nm rod positioned in the centre of each hexagonal prism of Mts comprising the axonemal pattern, and (2) an electron-opaque linkage material which connects each Mt of the axonemal pattern to its 4 nearest neighbours. Both of these structures are associated with the base of each axoneme within the centroplast shell, both form crossbridges with axonemal Mts, and both terminate at the periphery of the centroplast shell once the complete axonemal Mt pattern is established. It is suggested that these structures are involved in forming and maintaining the Mt pattern seen in the axopodia of Raphidiophrys.
