A comparative study of total protein, total and lipid associated serum sialic acid levels in patients with Behçet's disease and control groups.

The current investigation was designed to determine the total serum sialic acid (TSA), lipid associated sialic acid (LASA), total protein (TP), and TSA/TP values for 16 patients with active form of Behçet's disease, 12 patients with other uveitis types (pathologic controls) and 22 normal individuals. Data analysis indicated significant differences in mean TSA, LASA and TSA/TP values in patients with Behçet's disease (95.6 +/- 15.3 mg/dl, 29.8 +/- 7.4 mg/dl and 12.9 +/- 2.1 mg/g, respectively) and in pathologic controls (75.8 +/- 8.1 mg/dl, 26.6 +/- 0.3 mg/dl and 10.0 +/- 0.9 mg/g, respectively) when compared to normal controls (59.5 +/- 7.5 mg/dl, 19.2 +/- 3.2 mg/dl and 8.2 +/- 0.9 mg/g, respectively) (p < 0.01). No significant difference was observed in mean LASA values in patients with Behçet's disease when compared to pathologic controls (p > 0.05). No significant difference was observed in mean TP values in patients with Behçet's disease (7.4 +/- 0.7 g/dl), normal controls (7.3 +/- 0.4 g/dl) and pathologic controls (7.5 +/- 0.6 g/dl) (p > 0.05). A statistically significant difference was observed between the patients with Behçet's disease and pathologic controls with respect to TSA, TSA/TP values (p < 0.01). The results indicated that TSA and TSA/TP is most useful for differentiating Behçet's disease from other forms of uveitis.

Behcet Syndrome

Total protein, electrophoretic patterns, total amino acid and creatinine in the amniotic fluid of premature infants.

Total protein, total amino acid, creatinine and the 5 major protein fractions were determined in amniotic fluid samples from 42 normal and 12 premature neonates. Total protein, albumin, globulin, total amino acid, creatinine and A/G ratio were significantly lower in the 6 premature infants who developed the respiratory distress syndrome (RDS) than in the controls. The alpha 1, alpha 2 and gamma globulin levels were within the normal ranges. In the 4 premature stillborn babies, total protein and globulins were significantly higher than those of the controls, while the albumin, total amino acid and creatinine levels were normal. A/G ratio for this group was still lower than that for the former group and much lower than the control ratio. The remaining 2 premature alive infants showed a significant decrease in the alpha 1, alpha 2 globulins and creatinine levels. Slight increase in total amino acid, and significant increase in the beta globulin, while total protein, albumin, gamma globulin and A/G ratio were within the normal ranges.

Amino Acids

Amniotic fluid total proteins, electrophoretic patterns, total amino acids and creatinine in diabetic women.

Total proteins, total-acids, creatinine and the five major protein-fractions were determined in amniotic fluid of 9 diabetic mothers and 42 normal women. The mean value of the estimated patterns in the diabetic subgroup of living full-term infants was within normal, except a significant decrease in creatinine, albumin and gamma-globulin. A highly significant decrease in albumin, gamma-globulin and total amino acids in the amniotic fluid of those given stillbirth infants (SB), while a significant increase was found in both of total proteins and albumin in cases developed respiratory distress syndrome (RDS). In addition to a significant increase in creatinine, alpha 1-, alpha 2- and beta-globulins in both subgroups of SB and RDS. The A/G ratio of the 3 subgroups were significantly decreased especially those of SB.

[Protein fractions and total proteins in serum of rats on high fat diets].

This paper deals with the behaviour of serum protein fractions in rats fed 22 resp. 52 weeks a 50% or 3% fat diet using Millipore electrophoresis. The total serum protein concentration was also estimated. In rats on the high-fat diet the albumin fraction significantly dropped, while elevated levels of alpha 1- and gamma-globulin fraction were found. Serum protein content was slightly elevated in rats fed the diet 22 weeks, but this elevation was only significant after 52 weeks feeding. Within the same diet group there were no significant differences observed.

Alpha-Globulins

A simple stained protein assay (SPA) for rapid total protein determination applied to spinal fluids.

A rapid, simple technique measures small amounts of total protein applied on an agarose gel slab. Alternating current through the gel may reduce diffusion of the protein, which is chemically fixed. The intensity of the Coomassie-Blue-stained spots is a function of the amount of protein determined by comparison with simultaneously run standards. Densitometric and visual estimates of protein in spinal fluids by the stained protein assay correlated with Lowry values highly significantly (n = 28; P less than 0.001, Spearman test of rank correlation). As little as 0.2 mug protein was measured; this figure should be compared with 10 mug for the Lowry method. The reproducibility of the method was +/- 5%.

Simple method for correcting total protein in plasma for actual fibrinogen content.

Using plasma instead of serum for routine chemistry analyses has many advantages. To overcome the disadvantage of inclusively measuring fibrinogen in the plasma total protein assay without changing the clinical significance of the total protein assay, we investigated the possibility of subtracting the actual amount of fibrinogen from the plasma total protein. The correlation between serum and plasma total protein was excellent (plasma total protein = 0.989 x serum total protein + 6.7 g/L; r = 0.969; n = 131; mean difference = 5.55 g/L; P < 0.001). When the plasma total protein was corrected for the actual amount of fibrinogen, the correlation with serum total protein was equally good but the intercept was practically eliminated (corrected plasma total protein = 1.009 x serum total protein + 0.25 g/L; r = 0.985; n = 131; mean difference = 0.78 g/L; P = 0.47). The mean concentration of fibrinogen was 2.5 g/L (range: 1.38-3.62 g/L; n = 404) for blood donors, 3.6 g/L (n = 2707) for patients from the outpatient department, 4.6 g/L (n = 2023) for patients admitted to the hospital, and 6.6 g/L (n = 219) for patients whose concentration of C-reactive protein was > 50 mg/L. We conclude that the plasma total protein result should be corrected for the actual amount of fibrinogen.

Blood Proteins

Detection of serum proteins in the electrophoretic patterns of total proteins of mycoplasma cells.

The contamination of mycoplasma cell preparations by serum proteins originating from culture medium was studied. A. laidlawii and M. arthritidis cells were grown in the presence of [14C]-aminoacids, and the cells were washed with 0-9% NaC1 by threefold centrifugation. Total proteins of the washed cells were analysed by SDS gel electrophoresis. Coomassie-stained electrophoretic patterns were compared with autoradiographs of the same gels. The stained electrophoretic pattern of washed A. laidlawii grown without serum was identical with autoradiographs of the same cells grown without or with serum. That of washed A. laidlawii grown with serum differed from the corresponding autoradiography by the presence of extra protein bands I, II, III, and IV with molecular weights of over 160,000, 80,000-87,000, 55,000 and 25,000, respectively. The same extra bands were found in stained electrophoretic patterns of washed: (a) A. laidlawii cells grown without serum and mixed with serum in the stationary phase, (b) M. arthritidis cells, as compared with their autoradiographs, (c) serum precipitate. The bands III and IV may be due to the heavy and light chains of gamma-globulin, the band II might belong to transferrin or to some component of complement. Acidification of serum to pH 5 brought about 100-fold rise of amount of serum precipitate, the number of bands in the electrophoretic pattern of the precipitate being also increased. Stained electrophoretic patterns of cells purified by twofold centrifugation in step sucrose density gradient (1-20-1-27 g./cm.3 for A. laidlawii, and 1-15-1-25 for M. arthritidis) contained no extra bands and matched completely with their autoradiographs. It was concluded that contamination of washed mycoplasma cells by serum proteins is mainly due to co-precipitation of aggregated serum proteins together with cells during centrifugation rather than to adsorption of serum proteins on the cell surface.

Acholeplasma laidlawii

Total protein patterns in human amniotic fluid.

Total protein patterns of amniotic fluid samples of different age groups, following polyacrylamide disc gel electrophoresis, showed a wide range of variation in early pregnancy, but no two cases had identical banding, indenticating their genetic variability. An assessment has been made here of the total protein, albumin and transferrin patterns from normal fluids of different age groups by gel electrophoresis, in order to observe the genetic variations and relationship with foetal age, if any.

Albumins

Variation of immunoglobulin G and total protein concentrations during lumbar cerebrospinal fluid collection.

In successive lumbar samples of cerebrospinal fluid (CSF), each consisting of 5 ml, immunoglobulin G (IgG) and total protein were determined. Variation was studied by comparing the values in the initial sample with values in succeeding samples. In 13 out of 17 patients, IgG was reduced in sample 2. There was no reduction in the following samples. The concentration of IgG in sample 2 was below 50% of the concentration in sample 1 in 2 patients and below 70% in 9 patients. IgG was also reduced if the concentration in each sample was given as percent of the total protein concentration. The total protein concentration was not significantly reduced during collection of 25 ml of CSF. IgG was increased in sample 2 in 3 out of the 4 patients with neurological diseases since childhood. In 1 patient there was a considerable rise in the total protein concentration after collection of the first 5 ml, without a corresponding rise of IgG concentration. In clinical practice, the portion to which the CSF sample belongs should be noted, and comparison of values from various patients should be made between equivalent samples.

Immunonephelometric assay for urinary total protein and albumin in mice.

A quantitative immunonephelometric assay for measuring total protein and albumin in mouse urine has been developed. Antisera to whole mouse sera or to mouse albumin was added to aliquots of urine and the antigen-antibody complexes formed were measured by laser nephelometry. Sample volumes were small (20 microliters) and the assay time was 90 minutes. The assays were sensitive to 10 mg/L and within-day and day-to-day precision studies demonstrated coefficients of variation of less than 15%. Average urinary total protein and albumin concentrations for random urine specimens in female C3H mice (age 80-150 days) were 42 and 24 mg/L respectively. Urinary total protein and albumin to creatinine ratios were also determined.

Albuminuria

Changes in serum total protein values occurring in clinical samples during shipment.

Total protein and protein electrophoresis determinations were performed on paired serum samples before and after shipment of the samples via parcel post. After shipment, one serum from each pair of samples was analyzed at the same laboratory that had performed the initial analysis; the other sample from the pair was analyzed after its receipt by a referral laboratory. The results indicate that, due to some factor or a combination of factors occurring during shipment, statistically significant apparent increases were observed in 86 per cent of the total protein values and in 66 per cent of the albumin values. Experimentally, both mechanical agitation and bacterial contamination are capable of causing slight elevations similar in pattern to those observed in the shipped samples. The significance of these findings relative to clinical interpretation and to analytical quality control programs is discussed.

Bacteria

Some clotting factors in plasma during danazol therapy: free and total protein S, but not C4b-binding protein, are elevated by danazol therapy.

Anabolic steroids are known to increase the plasma concentrations of certain plasma proteins. In four patients given treatment with danazol, an attenuated androgen, the concentrations of heparin cofactor II, Hageman factor (factor XII), protein C, and both free and total protein S increased significantly when tested 39 to 103 days after the start of therapy. The titers of these proteins in samples obtained 21 days to 5 years after therapy was discontinued were similar to those before treatment, except for total protein S, the titer of which remained elevated. No significant changes in the titers of C4b binding protein or plasma plasmin inhibitory activity were found.

Assessment of five quantitative methods for determination of total proteins in urine.

1. We assessed five quantitative methods for the determination of urinary total proteins, three of them with precipitation followed by spectrophotomollowed by spectrophotometry (Tsuchyga/Benedict, Tsuchyga/Biuret and TCA/Biuret); one with gel-filtration (Sephadex G50/Biuret) and a Turbidimetric procedure (SSA). 2. Day-to-day precision was between 3.58 %CV (Tsuchyga/Benedict) and 11.46 %CV (Sephadex/Biuret). The Tsuchyga/Benedict method showed the closest values to controls quantitated by the Kjeldahl method and the lowest detection limit )17.3 mg of total proteins per liter). Recovery studies showed that a level of 100-200 mg/l the Tsuchyga/Benedict recovered over the 96% of the protein added. 3. Comparison studies of 66 24-hour urine samples showed that the best correlation was obtained between the Tsuchyga/Benedict and the Tsuchyga/Biuret methods (r = 0.996). 4. We conclude that the Tsuchyga/Benedict is a sensitive, precise and accurate procedure for the routine quantitation of urinary total proteins.

Biuret Reaction

The total protein content in the blood serum of vertebrates.

The total protein content in the blood serum of 416 species and subspecies of vertebrates is determined by the microbiuret method. Generally speaking the total protein content of the serum shows an increase with a higher evolutionary level. The difference between the mammals and the other groups is particularly large. This increase is probably attributable to a number of factors, for instance adaptation to living on land, development of the circulatory system, the development of adaptive immunity and the forming of specific transport proteins. A connection between albumin content and level of development can also be noticed.

Amphibians

Effect of excess iodine supplement on serum total proteins, electrophoretic patterns and body weight of Fayoumi chickens.

Total proteins and their major fractions were determined in the serum of 135 males and females of Fayoumi chickens, 65 were used as controls and 70 were treated with excess iodine in the form of KI. Serum proteins increased significantly with the increase in age till the 90th day in normals, then decreased gradually. Excess iodine supplement decreased body weight, serum total proteins, alpha2- and beta-globulins in most of males, while in females the decrease was pronounced till the 90 days old. Albumin increased in females than in males in normals, while in the treated groups it decreased in all females and in some of males. A/G ratio decreased with maturation in normals, but in the treated groups it increased in males and decreased in most of females than those of controls.

Animals

[Total protein and immunoglobulin content of human milk].

The content of total protein and immunoglobulins in the breast-milk has been determined by the biuret-method and radial immunodiffusion. The immunoglobulins IgG, IgA and IgM has been ascertained in all samples. The concentrations of total protein and of immunoglobulins are higher in the colostrum but in the breast-milk.

Colostrum

[Total proteins of the cerebrospinal fluid obtained by punction of the cisterna magna: normal values. Variations related to sex].

The total protein content of CSF collected from the cisterna magna (cisternal fluid) of 98 patients (58 males and 40 females) with no neurological diseases was determined by the colorimetric method devised by Lowry and co-workers. This method has been previously chosen based mainly on it's sensitivity, accuracy and low fluid consumption. The CSF samples were normal with regard to pressure, color, citology and it's content in glucose, chloride and urea. Complement fixation tests for syphilis and cisticercosis, as well as the globulin tests (Pandy, Nonne--Appelt and Takata-Ara) were negative. The average value and the normal range of cisternal fluid total protein was calculated for the mixed population (males and females). The mean protein value was 26.78 mg/100 ml, the lower and upper limits were respectively 13.20 and 40.36 mg/100 ml. These data are higher than those stated in some publications, and factors that could interfere in the different results mentioned in the literature are briefly discussed. Since the statistical analysis of the results showed that the mean total protein concentration in males (28.76 mg/100 ml) was higher than in females (23.91 mg/100 ml), normal limits for each sex were established: 16.96 to 47.13 mg/100 ml for males and 14.76 to 42.76 mg/100 ml for females.
