[Expression of PRAME gene in multiple myeloma].

AIM: To determine clinical significance of PRAME gene expression in multiple myeloma (MM) and feasibility of its use as a marker of residual tumor clone. MATERIAL AND METHODS: 35 MM patients, of them 15 were newly diagnosed and 20 had resistance to previous therapy. PRAME was made if the patients received programmed therapy with high-dose chemotherapy (VD) and autologous transplantation of peripheral cell stem cells. 12 PRAME-positive patients were examined on the day +100, 5 patients--a year later. Monoclonal paraprotein was detected by electrophoresis and radial immunodiffusion of blood serum. Bone marrow affection was assessed at roentgenography and/or MRI. PRAME gene expression in bone marrow biopsy was measured by reverse transcription and PCR amplification. RESULTS: Activation of expression of PRAME gene in MM was found in 68.57% patients. It was higher in patients with MM duration more than 1 year and if they were treated before (85%) than in new cases (46.67%). Expression of PRAME tended to associate with activity of LDP of blood serum. After the above chemotherapy and autotransplantation transcript PRAME did not disappear in 8 of 12 cases. One year after the treatment, of 5 PRAME-positive patients 2 died, 1 had recurrence, 2 are in a compete clinicohematological remission. CONCLUSION: Frequent activation of transcription of the gene PRAME in MM, its assay can be used for monitoring of the disease course, assessment of remission completeness, detection of tumor cell contamination of preparations of autologous stem cells of peripheral blood.

[Assessment of the immunohematological status of children exposed to low doses of ionizing radiation in the early and late periods after the Chernobyl AES accident].

The qualitative and quantitative parameters of hemopoiesis and immunocompetent cells were studied in 365 children in the early "iodine" and long-term periods after the accident. In accordance with primary response, a group consisting of 28 subjects with so-called "radiation injury" was distinguished. The early period was marked by unusual qualitative and quantitative hematological changes which, however, did not lead to the development of the hematological syndrome characteristic of acute radiation injury. Certain deviations that corresponded to somatic pathology were detectable at the late periods after the accident.

[Clinical-immunological characteristics of the state of the thyroid gland in children exposed to ionizing radiation because of the Chernobyl AES accident].

Overall 806 children evacuated from the city of Pripyat were examined for the thyroid condition. The children who received a dose of more than 30 rad for the thyroid manifested primary response in the form of euthyroid hyperthyroxinemia, a high risk of the development in future of autoimmune diseases in the lack of hypothyrosis.

[Simultaneous flow cytometric analysis of cell cycle and subpopulations of immunocompetent cells in workers participating in the clean up of the Chernobyl Atomic Energy Station accident].

Surface phenotype and cellular cycle of nonstimulated peripheric blood mononuclear cells of 35 cleaner-worker with dose commitment 0.05-0.25 Gy and 12 control persons were studied by means of flow cytometry. Differences in cellular cycle were found, they needed further investigations. The details of the method promoting its reproducibility are described.

[Immunological aspects of the study of contingents of population exposed to ionizing radiation effects as the consequence of the Chernobyl AES accident].

The immune system was examined in those who participated in the liquidation of accident sequelae at the Chernobyl Atomic Power Station and the population exposed to ionizing radiation. Alteration in surface antigenic markers of basic regulatory subpopulations of immunocompetent cells and metabolic changes are caused by radiation and co-existent somatic diseases. Typing for HLA antigens and proteins with a genetically determined phenotype revealed characteristic features of their distribution for the general population.

[Clinico-immunocytological characteristics of leukemia in persons exposed to ionizing radiation effects due to the Chernobyl AES accident].

A total of 18 patients with acute leukemia from those exposed to radiation when they had liquidated the consequences of the accident at the Chernobyl Atomic Power Station were examined. Sixteen of them had acute myeloblast cell leukemia, one had acute lymphatic leukemia, and another one had acute promyelocytic leukemia. A complex of hematological, immunocytological, and cytochemical findings, as well as leukocytic and erythrocytic genetically determined antigen typing were used for diagnosis. Signs of severe vegetovascular dystonia was found to be followed by acute leukemia in all the patients. A relationship was established between the therapy resistance and the HLA antigen homozygosity in combination with genetically-determined low reactivity. There were no significant differences between the parameters in question and those in patients with acute leukemia who had been studied in the preaccident period.

[Morphometric analysis of immunocompetent cells].

The authors have developed the technique of morphometric analysis of human peripheral blood immunocompetent cells by means of Laborscal PSA-1, PSL-1 conductometric cytoanalyzer and DBK-010 computer-processing of the resultant histograms. The method was tried in examinations of more than 400 patients with various diseases and in studies of lymphocyte activation changes. Formulae for calculation of cellular diameters and volumes are offered, as is the program for histogram processing.

[The immunological monitoring of patients with acute radiation sickness 12 to 36 months after the accident at the Chernobyl Atomic Electric Power Station].

Immunoassays of the 1st and 2nd levels were investigated in patients with acute radiation disease 12-36 mos. after irradiation. A decrease in the level of helper/inducer T-lymphocytes, a tendency to a decrease in the number of suppressors and an increase in the number of theophylline-stimulated T-cells in patients with acute radiation disease of the 3rd degree were observed. Control of the immune status of this group of patients is recommended.

[Effect of splenin on the development of tuberculin shock and Bordet's phenomenon in guinea pigs].

Effect of preinjection of splenin to tuberculosis-infected guinea-pigs on the development of tuberculin shock and Bordet's phenomenon was examined. In animals exposed to tuberculin shock, splenin was found to increase blast transformation of lymphocytes in response to tuberculin, as well as the degree of the proliferative reactions in lymphoid organs. It was also demonstrated that administration of splenin led to the reduction of the complement titer and the foci of tuberculosis in the organs. In guinea-pigs with the induced Bordet's phenomenon, splenin exerted no beneficial immunomodulating effect, promoting the development of more severe tuberculous lesions in the internal organs.
