Chemotherapeutic activity and histopathologic feature of BCNU + 5-FU effect on Lewis lung carcinoma.

In this study we investigated the effects of BCNU + 5-FU on the intramuscularly implanted Lewis lung carcinoma and on its spontaneous lung metastases. Chemotherapeutic agents were given i. v. either alone and in combination seven days after transplantation of 2.5 x 10(5) viable tumor cells, when the average weight of the primary tumor was 500 mg. The analysis of the following parameters: T/C, T--C, of %-metastases reduction, indicates that the simultaneous administration of BCNU + + 5-FU induces a more relevant response on the Lewis lung carcinoma than single drug therapy, especially evident at level of lung metastases.

In vitro and in vivo selection of two Lewis lung carcinoma cell lines.

Two tumor cell lines adapted to grow in vitro were originated from an explant of lung metastases of Lewis lung carcinoma. These lines differ in their malignancy when reinoculated into syngeneic animals; nevertheless, they do not show any difference for their in vitro clonogenic ability. From these lines 2 in vivo sublines of 3LL carcinoma were developed. The TD50 of the 2 in vivo sublines are different, and both the values obtained are lower than that of the original line. These results are interpreted as a selection of more malignant tumor cell lines.

Cyclophosphamide and iphosphamide against Lewis lung carcinoma: evaluation of toxic and therapeutic effects.

In the present report, investigations have been carried out to evaluate toxic and therapeutic effects of cyclophosphamide vs its isomer iphosphamide. Cytostatic action of the 2 drugs was assayed on the murine Lewis lung carcinoma (3LL). It has been observed that iphosphamide is less toxic as compared to cyclophosphamide (LD50 IP LD50 CP = 1.5); on the other hand, to reach the same therapeutic effectiveness on 3LL, an iphosphamide dose 1.6 - 2 times higher than that of its parent compound is necessary.

Lactate oxidation by skeletal muscle during sustained contraction in vivo.

Estimation of lactate oxidation in vivo was carried out by a tracer technique in the muscles of the lower hindleg of the dog during tetani of different duration. The fractional (%) rate of lactate oxidation increased markedly, compared with that of the resting muscle, after the first 2 min of stimulation. Lactate oxidation afforded a large contribution to the total oxygen consumption. Fatigue appeared to play a role in limiting lactate oxidation in the later phase of sustained contraction.

Observations on the actin content of the rabbit myofibril.

1. On extraction of whole muscle by the procedure of Hasselbach & Schneider (1951), the amount of actin that passes into solution seems to account for little more than 10% of the protein content of the myofibrils. 2. Extraction of isolated myofibrils with suitable media that allow identification and estimation of dissolved proteins seems to give about the same yield of actin (10-13% of the total). 3. A comparatively large residue of myofibrillar components remains after extraction. The amount of actin present in the residue can be only hypothetical.
