Ankylosing spondylitis: klebsiella and HL-A B27.

A search for the presence of Klebsiella-Enterobacter spp. and Yersinia enterocolitica in urine and faeces of 63 patients with ankylosing spondylitis was conducted because these microorganisms have been demonstrated to cross-react immunologically with HL-A B27 positive lymphocytes. The patients were graded into three groups on the basis of disease activity. Klebsiella spp. were found in the faeces of 13 (93%) of the 14 patients with 'active' disease, 10 (48%) of the 21 patients with 'probably active' disease and in one (4%) of the 28 patients with 'inactive' disease. Positive cultures were also obtained in 47 (38%) of 124 controls. It is suggested that the presence of Klebsiella spp. in faecal cultures may be associated with 'active' disease in patients with ankylosing spondylitis.

Genetic control of the immune response to ferritin in F1 hybrid mice.

The immune response to the antigen horse spleen ferritin, has been investigated in ten inbred parental strains and seven different F1 hybrid strains of mice, using an antigen excess technique. The degree of dominance in an F1 hybrid system can be estimated by using the Fisher dominance index. The responses in F1 hybrid animals, obtained from crosses of high and low responder parents, varied from dominant to recessive but the overall mean dominance index for the ferritin immunogenetic system was found to be -0.0467 +/- 0.0083 (mean +/- s.e.), a value close to zero, which suggests a codominant mode of inheritance of IR-genes to ferritin and this is consistent with most published data in other F1 IR-gene systems.

Effect of haemolysis on plasms and serum immunoglobulin estimations.

Plasma IgG, IgM and IgA estimations were carried out in blood samples with varying degrees of haemolysis, produced by controlled ultrasonication. Extent of haemolysis was determined by plasma haemoglobin, plasma potassium and packed cell volume; and plasma IgG, IgM and IgA estimations showed a steady and significant fall with increasing haemolysis. A calculated correction is proposed which compensates for the dilutional effect of haemolysis, during estimations of plasma and serum immunoglobulins.

Genetic control of the immune response to ferritin in mice.

The immune response to the antigen horse spleen ferritin was tested in sixteen inbred strains of mice. Using a fixed antigen percentage bound isotope technique, it was found that the quantity of antibody produced was related to the genetic status of the responding animal. A continuous distribution of responses was obtained, which were found to be linked to H-2 loci and the 2 non-H-2 loci 'Tla' and 'Theta' and these responses are detectable following primary immunization. It is suggested that the continuous distribution of quantitative antibody responses to ferritin, is compatible either with a large number of IR-genes or with a cross tolerance hypothesis requiring no IR-genes at all.

Codominant inheritance in immunogenetic (IR-gene) systems.

Immunogenetic (IR-gene) systems consist of animals showing different quantitative antibody responses when immunized with similar doses of a given antigen. Strains of animals giving high and low antibody titres are described as high and low responders, respectively. The degree of dominance in F1 hybrid strains, obtained from a cross between high and low responder parents, can readily be calculated using the dominance index formula, which takes the value of +1 for complete dominance, -1 for complete recessivity and the value of zero for no dominance. In reviewing 1527 F1 animals, obtained from ninety-one immunogenetic systems, the degree of diminance (d) was found to be: +0-0076 +/- 0-1053 (mean +/- s.e.), which is close to a value of zero and this is consistent with codominant inheritance. It is suggested that in immunogenetic systems, both alleles are expressed as codominant genes.

Raised serum IgG levels in hypertension.

Serum IgG levels were significantly higher in 118 severely hypertensive patients compared with a group of 163 normotensive blood donors, matched for age and sex. Serum IgA and IgM were the same in both groups. Raised levels of serum IgG were found in patients who had never been treated for hypertension, as well as in those who were treated with methyldopa or other hypotensive drugs.It is suggested that the raised levels of serum IgG may be an index of vascular damage induced by hypertension.
