Acute experimental allergic encephalomyelitis in radiation bone marrow chimeras between high and low susceptible strains of mice.

Acute experimental allergic encephalomyelitis (EAE) is an autoimmune disease involving the central nervous system (CNS) that can be elicited in susceptible strains of mice by the subcutaneous inoculation of mouse spinal cord homogenate (MSCH) in conjunction with complete Freund's adjuvant. In order to localize the physiological compartment conveying susceptibility to mice for EAE induction, hematopoietic radiation chimeras were prepared between the highly responsive SJL and low responder B10.S strains. Upon challenge with SJL MSCH preparations, high incidence of clinical disease was exhibited by B10.S----SJL chimeras but not by SJL----B10.S mice, suggesting that non-bone-marrow-derived factors were influencing development of disease. The incidence of histological lesions in the CNS was high for virtually all experimental and control groups except normal B10.S and B10.S----B10.S reconstituted mice. In contrast, challenge with B10.S MSCH induced a high clinical incidence of EAE in both B10.S----SJL and SJL----B10.S chimeras, indicating a possible interstrain difference in the immunogenicity of relevant CNS antigens.

Hypophysiotropic hormone testing in a patient with hypothalamic hypopituitarism.

A 59-year-old woman with metastatic breast carcinoma presented with weight loss, vomiting, and polyuria. Basal endocrine testing revealed low levels of thyroxine, cortisol, and gonadotropins, and the presence of diabetes insipidus. Direct stimulation of the pituitary with hypophysiotropic hormones indicated intact pituitary reserve. Insulin-induced hypoglycemia, however, failed to increase plasma cortisol or growth hormone levels significantly. On computed tomographic scanning, a lesion was found in the area of the hypothalamus. Thus, a functional abnormality of the hypothalamic-pituitary axis causing clinically significant hypothalamic hypopituitarism was not clearly apparent following administration of hypothalamic releasing factors but was demonstrable with indirect stimulation via insulin-induced hypoglycemia. Insulin-induced hypoglycemia remains an important diagnostic test in the evaluation of hypopituitarism.

Heat-sensitivity and the immune response of a methylcholanthrene-induced tumor.

Our results indicate that this methylcholanthrene (MCA)-induced tumor is immunogenic and that its heat-sensitivity is linked to the immune response of the host. Stimulation or reduction of the latter caused a corresponding change in the former. A therapeutic heat-dose failed to protect a majority of the treated mice against a second tumor challenge.

The NCI Drug Information System. 2. DIS Pre-Registry.

The Pre-Registry Module of the Drug Information System (DIS) is a staging area through which all new compounds are passed prior to acquisition and testing. Several methods are available for the entry of structures into the Pre-Registry; all involve built-in data validation. Newly entered structures are examined by computer programs for structural novelty and potential for anticancer activity. For those compounds that proceed to acquisition, the various acquisition steps, such as letter writing and record updating, are performed automatically. When a sample is obtained, the entire Pre-Registry record is updated and moved forward into the permanent DIS chemistry files.

The NCI Drug Information System. 3. The DIS Chemistry Module.

The Chemistry Module of the Drug Information System (DIS) handles a database of 400,000 structures. New or modified records are created in this database on a daily basis and are merged into the file promptly. The Chemistry database is searchable in a wide variety of ways and provides novel methods for both input and output of chemical structures.

TSH receptor antibody induction of thyroglobulin release from human thyroid cell monolayers.

We investigated the influence of TSH receptor antibody (TRA), as detected by inhibition of 125I-bTSH binding to detergent solubilized porcine TSH receptors, on in-vitro thyroglobulin (hTg) production using normal thyroid cells in monolayer. Secretion of hTg into the culture medium was analysed by a non-competitive enzyme immunosorbent (ELISA) technique utilizing two murine monoclonal antibodies. Basal hTg release (mean +/- SD 124 +/- 27 ng/10(5) cells/6 d, n = 5) was stimulated by bTSH (10, 10(2), 10(3) microU/ml) in a dose related manner (mean +/- SD 191 +/- 24, 587 +/- 80, 695 +/- 66 ng/10(5) cells/6 d, respectively). IgG (2 mg/ml) from seven patients with hyperthyroid Graves' disease, and known titres of TRA, similarly enhanced production of hTg, in a dose and time-dependent manner, when compared to control IgG. The degree of induction varied from a 140-230% increase in total hTg release over a 6-day incubation period. There was a direct correlation between the degree of 125I-bTSH binding inhibitory activity and the hTg response (r = 0.9, P less than 0.01). These data demonstrate that TSH receptor antibodies enhance hTg release from human thyroid cell monolayers and allow an assessment to be made of antibody-activated post receptor mechanisms.

Core criteria for diagnosing borderline patients.

This study compares 17 hospitalized borderline personality disorder (BPD) patients with 20 normal control subjects. Four criteria sets--DSM III, Grinker, Gunderson, and Kernberg--were combined and used with the semistructured DIAGNOSTIC INTERVIEW BORDERLINES (DIB) Scale. Findings indicate that among BPD patients the following were most prominent: 1) impulsive episodes 2) unstable relationships 3) chronic feelings of depressive emptiness/loneliness 4) acting out behavior and, somewhat less prevalent, 5) identity disturbance. BPD patients can be discriminated by different patterns of disturbance while sharing other features in common with them. While DSM III requires five of eight items for BPD diagnosis, from the above group significantly less than five adequately made a positive diagnosis in this study. BPD seems to identify a heterogeneous group of patients with behavioral disturbances without particular personality specificity which share certain core characteristics among which additional features may further subtype component members.

Heating pattern of a single-loop applicator buried in muscle-equivalent phantom material measured at S-band.

We measured the initial rate of energy deposition at S-band (2.455 GHz) for a single-turn grounded insulated coil buried in muscle-equivalent phantom material. Most of the electromagnetic field is confined within the horizontal dimensions of the coil, and 30% or more of the incident energy is deposited at depths up to 1 cm below the coil. We believe that this device, sheathed in biocompatible PTFE teflon, could be implanted in a tumor region and left in situ during the course of a fractionated hyperthermia protocol.

Cost-benefit analysis of neonatal intensive care for infants weighing less than 1,000 grams at birth.

Cost-benefit analysis was performed on the care of 247 infants weighing between 500 and 999 g at birth, admitted to Women and Infants Hospital of Rhode Island between January 1977 and December 1981. The neonatal mortality was 68%. Eighty-seven percent of the survivors were evaluated neurodevelopmentally for 1 to 5 years: 74% were normal or minimally impaired, 10% were moderately impaired, and 16% were severely handicapped. Using these data in conjunction with cost information obtained from the hospital and therapeutic care facilities for handicapped children, total lifetime costs for the care of these infants were estimated. In 1982 dollars, present values of costs ranged from $362,992 per survivor for those weighing between 600 and 699 g to $40,647 per survivor for those weighing between 900 and 999 g, resulting in an inverse correlation between cost per survivor and birth weight (P less than .001). We estimated present values of expected lifetime earnings per survivor, with a range of zero earnings for infants between 500 and 699 g, to $77,084 for those with birth weight of 900 to 999 g. It is concluded that from the standpoint of cost-benefit analysis as was used for this study population, neonatal intensive care may not be justifiable for infants weighing less than 900 g at birth.

Spontaneous interstitial nephritis in kdkd mice. I. An experimental model of autoimmune renal disease.

Mice of the kdkd strain predictably develop a spontaneous tubulointerstitial nephritis after 8 wk of life. In this report we have examined several aspects of the nephritogenic immune response that seemed potentially relevant to the expression of this progressively destructive renal lesion. Of particular interest is that by direct immunofluorescence we were unable to demonstrate the presence of antibodies to determinants in the tubulointerstitium. Serum and kidney eluates from nephritic mice, furthermore, did not stain any renal structures in normal kidney. We did observe, however, that disease could be transferred through kdkd----CBA/Ca bone marrow chimeras, and prevented, in the reverse direction, by CBA/Ca----kdkd chimeras. The development of the interstitial lesion was markedly inhibited by thymectomy with T cell depletion, but disease could not be adoptively transferred with cells or serum from nephritic mice. The interstitial lesions also did not appear in (kdkd X CBA/Ca)F1 hybrids, and the development of disease in kdkd mice could be inhibited by treatment with adoptively transferred T cells from CBA/Ca mice. With these new findings we now hypothesize that susceptibility to the expression of interstitial nephritis in kdkd mice involves the cellular limb of the immune system, and may be related, in part, to alterations in regulatory T cell function.

Several heat-sensitive properties of the MULv-induced YAC lymphoma.

A therapeutic heat dose of 44.0 +/- 0.1 degrees C/20 min caused either temporary or permanent inhibition of tumor growth in 86% (25/29) of the CAF1/J female mice inoculated in the footpad with MULv-induced YAC lymphoma and heated when the tumor was well established. Heating before the tumor is well established is less effective because the heat-sensitivity of the tumor increases as it grows in the host. The transplantability of this tumor is more effectively impaired when heated tumor tissue is left in situ for 24 h post-heating than if it is excised immediately after heating and implanted in host mice.

Delay in onset of heat-sensitivity in a methylcholanthrene-induced tumor.

A delay in onset of heat-sensitivity was demonstrated for methylcholanthrene-induced tumors implanted in the footpad of BALB/cJ or CAF1/J female mice. A heat dose of 46.0 +/- 0.1 degrees C/10 min resulted in long-term cure for a majority of the mice heated on day 7 post-implantation but cured only 4% of those heated on day zero and non-heated on day 1 post-implantation. By comparison, all untreated controls grew tumors which never spontaneously regressed and were dead within 3 months post-implantation.
