Reflectometric blood glucose determination in the neonatological intensive care: haematocrit dependence.

The dependence upon haematocrit of the readings of different methods of reflectometric blood glucose determination ( Eyetone /Dextrostix: Reflomat / Reflotest Hypoglycaemie , Reflomat / Reflotest Glucose, Glucometer /Dextrostix) was studied in blood samples adjusted to haematocrit values between 0.47 and 0.90. In general the reflectometric methods measure lower blood glucose concentration if the haematocrit value is enhanced. There is a clear-cut correlation between the rise in haematocrit and the decrease in blood glucose reading. The critical haematocrit level to reach this effect in the Eyetone and Glucometer is 0.60, and in the Reflomat it is 0.80.

Cell-mediated immunoreactivity in acute human trichinellosis.

Twenty patients were examined for studying specific cellular immunoreactivity in human trichinellosis. Serial leukocyte migration inhibition tests were carried out using trichinella larvae as antigen. The mean migration indices of the patients differed significantly from that of the healthy controls. As the most distinct immunoreactivity was found during the acute phase of the disease, it is suggested that the cellular immune response may be of some importance in the development of symptoms. It is, however, stressed that the diagnostic value of the LMI method is less than that of the serological tests in individual cases.

Leukocyte migration inhibition (LMI) by a specific antigen in human toxoplasmosis.

The diagnostic value of the leukocyte migration inhibition test was examined by comparing it to other specific laboratory methods i.e. complement fixation, indirect haemagglutination and skin test in 10 patients with acquired lymphoglandular toxoplasmosis. In the acute phase the serological tests gave more reliable results than did the LMI test. In subacute and chronic cases, however, there was a good correlation between dermal reactivity and LMI test. LMI is believed to represent a specific test for the detection of cellular hypersensitivity in human toxoplasmosis and so it can be used as a non-invasive method instead of dermal tests.

Endogenous formation of N-nitrosomorpholine from a new fungicide.

Trimorphamide [N-(1-formamido-2,2,2-trichloroethyl)morpholine], with an acute LD50 of 990 mg/kg, is a systemic fungicide under development. To examine its possible nitrosation in vivo, CFY rats were treated i.g. with 326 mg/kg trimorphamide, suspended in 40 mg/kg sodium nitrite. after 1 hour, another 40 mg/kg sodium nitrite was given i.g. and the animals were killed an hour later. The nitroso derivative was extracted by dichloromethane from the gastric content, after steam distillation. The samples were analysed by gas chromatography (GC) and mass spectrometry (MS). N-Nitrosomorpholine (NMOR) was identified by comparison with authentic samples, both by GC and MS, using retention times and electron impact mass spectra. Quantification was carried out by GC-MS-specific ion monitoring, using the m/e 116 peak. The yield of NMOR ws found to be 0.65%. By the use of the Ames test, NMOR formed in vivo proved to be strongly mutagenic for the strains TA 1535 and TA 100, both in presence and absence of S-9 fraction. Tumour-bearing animals, treated by sodium nitrite plus trimorphamide, are being analysed.

Mutagenicity of N-nitroso pyridinol carbamate with the Salmonella/mammalian microsome test.

Pyridinol carbamate was nitrosated to the N,N1-dinitroso derivative and the structure was proved by spectroscopic methods, including chemical ionization mass spectroscopy. By the Ames test, the dinitroso derivative showed a significant dose-dependent mutagenic response with Salmonella typhimurium strains TA 1535 and TA 100; the response became more pronounced in the presence of microsomes. As the dosage of N-nitroso pyridinol carbamate increased, the number of revertant colonies also increased. Pyridinol carbamate was not mutagenic.
