[Hemodynamic effects of the treatment of hypertension with the calcium channel blocker diltiazem].

The 1.5-benzotthazepine derivative diltiazem, which blocks slow calcium channels, is an effective hypotensive agent, used for the treatment of moderate hypertension. Treatment with diltiazem alone, its dose adjusted on an individual basis, controlled arterial blood pressure in 58% of the patients. With a 240 mg daily dose, blood pressure was controlled in 12 of 52 patients; when the daily dose was increased to 360 mg, blood pressure was controlled in another 18 patients. In the remaining 22 patients, blood pressure was only controlled after captopril was added in a daily dose of at least 50 mg. The drop of arterial blood pressure was associated with falling total peripheral resistance and end diastolic blood volume, and was accompanied by an increase in left-ventricular ejection fraction and myocardial oxygen want.

[Effect of clopheline on thrombocyte aggregation].

Data on clopheline effect on platelet aggregation, both spontaneous and ADP- or adrenaline-induced, in normal donors are reported. Clopheline is shown to slightly increase aggregation in normal subjects, exhibiting properties of a weak aggregation inductor. Where adrenaline is present in the aggregating system, clopheline undermines its platelet aggregation-inducing effect and, consequently, shows properties of a more competitive antagonist, as compared to adrenaline.

[Hemodynamic effect of a combination of prazosin with cardiac depressants in the treatment of hypertension].

Arterial blood pressure and central hemodynamic changes were assessed in 44 patients with essential hypertension, uncontrollable by prazosin alone, after propranolol (anaprilin), celiprolol, clonidine (clofelin) or verapamil were added to the treatment schedule. A selective beta 1-adrenoblocker, celiprolol, is shown to be the best option in prazosin-treated patients with reflex increment of heart rate and stroke volume. The prazosin-verapamil combination is also justified in cases of moderately increased pulse rate, while propranolol and clonidine produce no additional hypotensive effect when combined with prazosin.

[Pharmacokinetic bases for the drug-free prevention of drug-induced hepatitis in pulmonary tuberculosis patients].

The causes and frequency of occurrence of toxic hepatitides determined by isoniazid and rifampicin, were analysed in 551 patients with primary tuberculosis detected by clinical pharmacological methods. The intravenous mode of drug administration was shown to be indicated for prevention of isoniazid hepatitides in patients with a fast type of acetylation. Doses and methods of therapy with isoniazid and streptomycin leading to their high concentrations in the patients' body should be avoided to prevent liver affection with rifampicin. Since rifampicin hepatitides develop in persons with a slow type of acetylation, rifampicin and isoniazid with streptomycin should be injected with a 6-hour interval to prevent them.

[Effect of no-shpa, halidor, dibazol and euphylline on the fibrinogen content and fibrinolytic activity of blood].

With a single intravenous administration to albino rats of both sexes of nospanum and halidor in a dose of 2 mg/kg and dibazol--in 1 mg/kg produce activation of fibrinolysis, while euphylline in a dose of 12 mg/kg has no effect on this process. A protracted introduction by the subcuataneous route of halidor and nospanum in a dose of 10 mg/kg, of dibazol--in 5 mg/kg and of euphylline intramuscularly in a dose of 24 mg/kg depresses in animals the activity of the proteolytic process. It was found that in vitro nospanum, halidor and dibazol accelerate fibrinolysis, whereas the presence of euphylline in the blood plasma deccelerates this process.
