Allotype polymorphism of serum globulins (Gp system) in pigs.

By alloimmunization with whole serum reagents were obtained which revealed two new allotypes, G8 and P30, in pigs. Evidence is presented that the allotypes G8, P30 and previously described G4 and G6 are controlled by three complex autosomal allelic genes forming a closed system. For this new allotype system we propose the designation Gp (globulin, pig). For alternative G4-G8 and G6-P30, forming pairs of mutually exclusive alloantigens, it is proposed to use a standard designation GpA-Gpa and GpB-Gpb respectively.

Blood serum immunoreactive gastrin level in infants with hypertrophic pyloric stenosis.

Blood serum immunoreactive gastrin level (IRG) was measured in infants with hypertrophic pyloric stenosis before and after corrective surgery and in a control group children of corresponding age. No significant difference in IRG level was found between the stenotic infants and the control group. In the stenotic infants IRG values were higher at the seventh day after than before the operation and significantly higher in those infants in whom gain of body weight was noted during that time as compared with the infants without gain of weight. These observations remain in agreement with the view that the main role of gastrin in infants is trophic action on the mucosa of the upper gastrointestinal tract.

Esterolytic activity of blood serum in infants with hypertrophic pyloric stenosis.

Esterolytic activity of blood serum was determined in infants with hypertrophic pyloric stenosis and in healthy control children. The substrates hydrolyzed mainly by arylesterase (EC. 3.1.1.2), i.e. p-nitrophenyl esters of acetic, propionic and butyric acid were used. It was found that in infants with hypertrophic pyloric stenosis the rate of p-nitrophenyl propionate hydrolysis was significantly higher than that of the acetate or butyrate. In control group children all three substrates were hydrolysed at similar rates, and these were significantly lower than in infants with hypertrophic pyloric stenosis.

Protein-cationic detergent interaction. Interaction of bovine serum albumin and other proteins with alkylpyridinium bromides studied by viscosity, gel filtration and spin-label methods.

Viscosity, gel filtration and spin-labelling methods have been used to study the influence of alkylpyridinium bromides on the conformation of bovine serum albumin and other proteins. Cationic detergents cause partial unfolding of the native protein molecules. The magnitude of these changes increases with increasing length of the detergent hydrocarbon chain. When cationic detergents are added to reduced and carboxymethylated bovine serum albumin the observed changes are opposite to those found in native protein.

Genetic Transformation as a tool for detection of Neisseria gonorrhoeae.

A rapid method for the detection of Neisseria gonorrhoeae, making use of the ability of deoxyribonucleic acid samples from clinically isolated strains of this organism to transform nutritional mutants of a particular strain of N. gonorrhoeae, has been described. In addition to using isolated cultures, transforming deoxyribonucleic acid can be obtained directly from the material that adheres to swabs of the cervix or the urethra. The time interval for transfer of swabs to the diagnostic laboratory is not a significant factor. It is not necessary to use pure cultures on primary isolation plates to obtain definitive results. Nongonorrhoeae neisserias, as well as a large variety of commonly encountered unrelated bacteria, do not react or interfere in the transformation assay when using one of the mutant strains under a standardized set of conditions. The entire assay can be completed in less than 24 h. It has also been shown that type T4 cells of the strain of N. gonorrhoeae employed in the present study are competent for genetic transformation, although type T4 cells are transformed at a significantly lower frequency than are type T2 cells of the same strain.
