Distribution of nickel and cytogenetic changes in poisoned rats.

The effect of dose and duration of exposure on the relative distribution of nickel in the vital organs and bone of albino rats was investigated. The rate of accumulation of the metal, in general, was highest in myocardium and spleen followed by kidney, bone and other tissues. The sub-cellular fractionation of myocardium revealed higher metal localization in nuclear and mitochondrial fractions than in the microsomal and cytosolic fractions. However, the uptake of metal by various tissues and its sub-cellular distribution was regulated by the dose and duration of exposure. No significant chromosomal damage was observed in either the bone marrow or the spermatogonial cells in the animals during early nickel intoxication.

Biochemical and morphological changes in some organs of rats in nickel intoxication.

Biochemical and histological alterations in liver, kidney, testis and myocardium of albino rats were investigated at various intervals after daily administration of nickel sulphate. No significant changes were observed during the initial periods of the treatment, but longer exposures produced marked enzymatic and histopathological alterations in all the four organs examined. The severity of the histological lesions was found to be directly related to the duration of the treatment.

Effect of nickel sulphate on male rats.

Cytopathological and histopathological changes in skin, liver, kidney and testis of rats due to nickel sulphate have been studied experimentally. Skin of nickel sulphate painted rats showed hyperkeratinization, vacuolization, hydropic degeneration of basal layer and atrophy of epidermis. The testis showed degeneration and oedema of seminiferous tubules, while liver showed areas of focal necrosis, congestion and dilatation of sinusoids.

Comparative toxicity of trivalent and hexavalent chromium to rabbits. II. Morphological changes in some organs.

Toxicity of trivalent and hexavalent chromium compounds was investigated in experimental rabbits to ascertain health hazards among industrial workers of miners occupationally exposed to such chemicals. Brain, kidney and myocardium showed a tendency to accumulate chromium irrespective of its valency state; the morphological changes were more marked in animals exposed to hexavalent chromium. However, no definite co-relation could be observed between the concentration of the metal and the degree of histological changes in these organs.

Studies on cardiolipin biosynthesis in Mycobacterium smegmatis.

Supplementation of a growth medium with 5% glucose has been found to stimulate the formation of cardiolipin and phosphatidylethanolamine five- and threefold, respectively, in Mycobacterium smegmatis. The presence of both cytidine diphosphate diglyceride and phosphatidylglycerol pathways of biosynthesis of cardiolipin in cell-free extracts has been demonstrated. The enzymes were localized in the fractions which contained membranes. Isonicotinic acid hydrazide and streptomycin sulfate inhibited the formation of cardiolipin.

The effect of artificial ventilation on functional residual capacity and arterial oxygenation. II. Comparison of spontaneous respiration and artificial ventilation at similar arterial carbon dioxide tensions, tidal volumes and inspiratory gas flow rates.

We have compared cardiac output, gas exchange and pulmonary mechanics during spontaneous breathing and artificial ventilaton under conditions which kept PaCO2 within the normal range and maintained constant tidal volume and inspired gas flow rate. In dogs anaesthetized with pentobarbitone and ventilated with air, artificial ventilation increased VD/VT but did not reduce Q angstrom, FRC, or CL. PaO2 increased and A-aDO2 decreased during aritificial ventilation, perhaps because of a small increase in Q angstrom and a small decrease in oxygen consumption. It appears that many of the reported deleterious effects of artificial ventilation may be due to the use of other anaesthetic agents and patterns of ventilation, and to changes in PaCO2.

Effect of diazinon in male rats. Histopathological and biochemical studies.

Mild structural and functional changes were observed in liver and testes of rats after a single intraperitoneal administration of diazinon (21.6 mg/kg). Kidney, however, showed no pathological lesion. Attempts are made to correlate the pathological changes in these organs with the activity of succinic dehydrogenase, adenosine triphosphatase and alkaline phosphatase.

Thorium induced testicular changes in rats.

Daily intraperitoneal administration of thorium nitrate produced progressive morphological and biochemical alterations with the increase in thorium concentration in rat testis. The degenerative changes in the seminiferous tubules increased with the duration of treatment and after 90 days calcification occurred in about 25% of the tubules and in the connective tissue of the tunica albuginea. The activity of adenosine triphosphatase and alkaline phosphatase increased markedly as a result of thorium administration. An attempt has been made to interrelate histopathological and enzymatic changes and the metal concentration in the testicular tissue.
