Geotrichum candidum link 1809: hydrolases activity and own method of digestive tract strains biotyping.

Hydrolase activity of (enzymograms, biotypes) in Geotrichum candidum, one of the poorly described pathogenic fungi, was studied 81 strains were isolated from oral cavity and faeces of patients with gastrointestinal tract disorders. Axenic strains were differentiated with API 20C Aux and API ZYM tests. Then, enzymograms and biotypes were determined for all strains based on the activity of 19 hydrolases. High variability of enzymograms (17 different types) was found. The highest activity was noted in case of: e2 - alkaline phosphatase, e6 - leucine arylamidase, e11 - acid phosphatase. E5 - lipase, e7 - valine arylamidase, e12 - naphtol-AS-BI-phosphohydrolase and e17 - beta-glucosidase were used for biotyping procedures. Our own system of biotyping of 81 strains of G. candidum was based on the mathematical binominal distribution formula (1 : 4 : 6 : 4 : 1) - all "+"; one "-", three "+"; two "two "+"; three "-", one "+"; all "-". We have found: A (11.1 +/- 3.5%), BI (6.17 +/- 2.67%), B2 (1.23 +/- 1.22%), B4 (4.94 +/- 2.41%), C, (1.23 +/-1.22%), C3 (63.0 +/- 5.4%), D2 (9.88 +/-3.31%), D3 (2.47 +/- 1.72%). Among all strains from 8 various biotypes of G. candidum.

[Biochemical characteristics of fungi isolated from ontocenoses of digestive tract in children].

Fermentation of carbohydrates and assimilation of carbon compounds were evaluated. The utilization of 18 specific carbon compounds was estimated in the investigation of each strains for differentiation of the species. From among 125 strains of fungi the following were found: Candida albicans (112 strain), C. famata (2), C. glabrata (1), C. guilliermondii (1), C. kefir (4), C. tropicalis (2). The activity of 19 hydrolases was investigated using API ZYM. Biotyping of Candida strains was done according to the Williamson classification (1986), modified by Kurnatowska (1998). All strains were isolated from the oral cavity, rectum, as well as from the materials collected during endoscopy in children.

[Fungal invasions in the digestive tract in children].

The aim of presented study were fungal invasions of the oral cavity and gastrointestinal (GI) tract in children suspected of the inflammation of gastric and duodenal mucosa and of absorption disturbance. The fungal strains (125) were identified using API 20C and API 20C AUX (bio Me'rieux); they were as follows: Candida albicans, C. famata, C. glabrata, C. guillermondii, C. kefyr and C. tropicalis.

Abdominal Pain↗

[The sensitivity to ketoconazole of Candida strains isolated from the digestive tract ontocenoses in children].

The study included 52 children. During endoscopy the contents of oesophagus, stomach and duodenum were collected with catheters, and additionally washings from the oral cavity and anal swabs or samples of faeces. Materials were inoculated on Sabouraud's media. The axenic strains of fungi were identified using own methods and others tests (API 20C, API 20C AUX). The activity of ketoconazole (K) was measured against 100 strains of Candida: C. albicans (91 strains), C. kefyr (4), C. famata (2), C. tropicalis (1), C. guilliermondii (1), C. glabrata (1). The strains susceptibility to K (RO4 1400 Janssen) was estimated with the own system based on the dose-response curves; the minimal inhibitory concentration (MIC) values were canculated. The low values of MIC indicate high susceptibility to K of examined Candida strains.

[Prevalence of Trichomonas vaginalis Donné in women of Lódź population in 1955-1999 years].

Search for Trichomonas vaginalis was carried out in direct (in 0.85% NaCl and 0.1% safranine) and fixed stained microscopical preparations ( Wright or Giemsa methods, after fixation with 70% methyl alcohol) and cultures ( Pavlova, Johanson-Trussel-John, Roiron-Ratner, Simic media), from materials of the vagina, urethra and cervix uteri. In the years 1955-1999 a total of about 12000 women were examined. The prevalence of T. vaginalis in women population of Lódź region was high (from 26.6% still 78.0%); on the last ten years in about half of the sample examined women were infected of T. vaginalis.

[Hydrolytic activity of Trichomonas vaginalis Donné strains].

The hydrolytic activity of Trichomonas vaginalis Donné strains was investigated using API ZYM test (bioMérieux). Among 19 determined hydrolases were found alkaline and acid phosphatases; also in several strains N-acetyl-beta-glucosaminidase and a-glucosidase were demonstrated. The API ZYM test may be used for characterization of enzymatic activity of T. vaginalis strains, but adequate medium is necessary.

Serodiagnostics of Toxoplasma gondii invasions.

Diagnosis of T. gondii invasions is based mainly on serological tests, because of their relative simplicity and availability. In the present work we examined efficiency of immunoblot and two inhouse ELISA assays in anti-Toxoplasma antibody detection, comparing our results with those obtained using commercial ELISA kit of Organon Teknika. We found that inhouse ELISAs and immunoblot can be helpful in diagnosing toxoplasmosis.

[Prevalence of Toxoplasma gondii and analysis of specific immunoglobulins concentration in serum of women during the reproductive period in a sample of Włocławek population].

The aim of the study, from 1996 to 1999, was to investigate the serological course of prevalence Toxoplasma gondii in 985 women of procreation period. In non - pregnant women the incidence of Toxoplasma gondii specific IgG was significantly higher (59.0 +/- 1.9%) than in pregnant women (43.4 +/- 2.8%). Specific IgM was less frequent (from 1.47 +/- 1.02% to 1.76 +/- 0.66%).

[Multifocal fungal infections in pregnant women].

The aim of the study was to estimate the prevalence of fungi in monofocal and multifocal infections in pregnant women. 251 pregnant women were examined for presence of fungi in vagina, oral cavity and rectum. Fungal strains were recovered in 259 out of 753 samples (49.2% of all pregnant women). Presence of fungi in vagina was associated with the invasion of oral cavity and/or rectum in 98% of all women. Monofocal vulvovaginal mycosis was found only in 1.9% of the cases. Bifocal infections depended on the ontocenoses: vagina - oral cavity (32.1%); vagina--rectum (7.5%); oral cavity--rectum (13.2%). Trifocal infections (vagina - oral cavity--rectum) occurred in 58.5% of all examined women.

[Proteolytic activity of Candida albicans strains isolated from women's genital organs with clinical diagnosis of mycosis].

The aim of the presented study was to assess the in vitro activity of aspartate proteinase of strains isolated from women's genital organs with clinical diagnosis of mycosis. We have evaluated the activity of proteinase of 75 Candida albicans strains using Staib method with Rózga modification, in agar medium with human albumin at pH 4.6. The proteolysis zone ranged from 4.0 to 11.0 mm. The values of proteolysis zones in human albumin of C. albicans were similar to left-oblique normal distribution. High and extremely high proteolytic activity is characteristic for more than 50% of strains.

[Susceptibility to itraconazole and miconazole of fungi strains isolated from the vaginal ontocenosis].

The activity of itraconazole (RO 51211, molecular weight 705,64) and miconazole (RO 14889, molecular weight 479,15) obtained from Janssen Pharmaceutica NV, against 100 fungi strains, was studied. These strains were isolated from vaginal materials of patients with recognized candidosis. The axenic fungal strains were assessed by own methods and unified tests (bioMérieux: API 20 C, API 20 C AUX). The minimal inhibitory concentration (MIC) values calculated from dose-response curves on agar diffusion-test (Kadłubowski's method), were also examined by analysing the variation (mean x +/- standard error, median Me, mode Mo, Min-Max). The drug MIC values for all strains were characterized by a wide range of variation itraconazole 0.0001 - 5.30 mg/L, miconazole 0.0045 - 4.70 mg/L; the shape of MIC values variation polygons was similar to normal distribution.

[Some aspects of fungi multifocal invasions connected with the genital organs in women].

The aim of presented study was analysis of fungal invasions of the genital organs, oral cavity and digestive tract in 799 women. Fungal strains were identified using API 20C and API 20C AUX (bioMérieux); they were as follows: Candida albicans, C. krusei, C. tropicalis, C. guilliermondii, C. kefyr, C. glabrata, C. famata, C. parapsilosis, Rhodotorula rubra, Geotrichum candidum. The number of fungal strains isolated from the oral cavity ontocenosis was the highest (41.8 +/- 2.4%). Fungal strains isolated from the vagina were less numerous (34.6 +/- 2.3%), and from the rectum the least numerous (23.6 +/- 2.1%). Multifocal invasions (limited to one ontocenosis) were present in about 50% of women; bifocal were less frequent, and this difference was statistically significant (p < 0.01): fungi in the vagina and oral cavity were detected in 28.6 +/- 3.0%, in the oral cavity and rectum in 19.0 +/- 2.6%, and in the vagina and rectum in 13.0 +/- 2.2%. Trifocal invasions were more frequent (39.4 +/- 3.2%). There was a visible trend of increasing prevalence of fungi in genital organs over 10 year period from 17.9 +/- 1.4% to 34.5 +/- 1.7%.

Search for the pathogenic fungi in amniotic fluid.

Intra-amniotic infection with Candida is rare but considers an etiological factor for preterm rupture of membranes (PROM). The aim of the study was the investigation of the mycotic invasion of the amniotic cavity in pregnant women with PROM and with intact fetal memranes. 170 women were included in two groups: I (n = 150)--women with intact membranes; II (n = 20)--women with PROM. Samples of amniotic fluid were cultured on Sabouraud's medium. Fungal strains were isolated also from vagina, oral cavity and rectum of women with PROM. Intraamniotic infections were absent both in women with PROM and with intact membranes. Candida strains were isolated from vagina, oral cavity, and rectum in the group of women with PROM. Coexisting of multifocal Candida infections in pregnant women with PROM indicates that in those cases mycologic diagnosics should be recomended.

[Proteolytic activity of Candida strains--parameter of their identity or similarity].

Proteolytic activity of 238 strains of Candida was investigated by the Staib method in Rózga modification, permitting for quantitative evaluation of the result (agar medium with albumin). Analyzing size of proteolysis zones--obtained for strains isolated from selected ontocenoses of the same women--a linear correlation coefficient was used; for confirmation of identity of tested strains a conformity coefficient was used. Isolated strains were identified as C. albicans, C. guilliermondii, C. krusei, C. pseudotropicalis, C. tropicalis and other species (singular). It was found that the curve of distribution of proteolysis zone sizes of all strains is normal and that statistically significant correlation between size of proteolysis zone of strains isolated from vagina and oral cavity of the same women does exist.

[Characteristics of fungi and attempts of their elimination from the oral cavity in children treated with orthodontic appliances].

Microbiological evaluation of oral cavity was performed in 112 children, aged 6-18 years, with retrogenic defects (group I) and other defects (group II). The material for microbiological investigation consisted of contents of oral cavity cultured on the Sabouraud broth. Isolated abacterial strains of fungi were differentiated according to their morphological and biochemical properties. Among isolated fungi, independently of the bite defects and age of children, highest percentage of strains of Candida albicans was noted. Remaining species represented genus Candida (C. glabrata, C. guilliermondii, C. krusei, C. parapsilosis, C. pseudotropicalis, C. zeylanoides), Geotrichum (G. candidum), Rhodotorula (R. rubra) and Trichosporon (T. cutaneum). Fungi were present in oral cavity in 45% (age 6-8 years) and 36.4% (age 9-18 years) of group I patients. In group II these percentages were respectively 15.2 and 58.7. In control groups occurrence of fungi was 3-4 times lower. Despite application of disinfectants and treatment with nystatin, frequency of isolation of fungi did not change.

[Comparison of antitoxoplasma antibody level using various serologic tests in women in consecutive trimesters of pregnancy].

A total of 540 women, in two clinical groups, differing by information obtained during the interview, were investigated. Several observations were made. It was found that values of titers in indirect immunofluorescnece test (OIF) in consecutive trimesters of pregnancy in the same women are not changing in 50%, in about 10% differ by some to 10 times, and in remaining patients move by one grade of serum dilution with a tendency to increase. This seroconversion regards about 17% and from negative to positive in 10%. High convergence of results obtained by OIF and ELISA tests for IgG antibodies was detected. The level of specific antibodies determined by ELISA test can be analyzed basing on parameters of distribution of quantitative feature and coefficients of linear correlation. Secondary specific antibodies against Toxoplasma of the IgM class may appear in every trimester of pregnancy with positive antibodies of IgG class which in some cases remain unchanged for several months regardless of treatment with anti-toxoplasmal drugs.
