Neuroprotective effect of cytoflavin during compression injury of the spinal cord.

Cytoflavin normalized energy metabolism, decreased the intensity of lipid peroxidation, and reactivated the antioxidant system in the spinal cord of rats with compression injury at the level of Th10-Th11. The neuroprotective effect of the test preparation manifested in normalization of hindlimb motor function and decrease in mortality rate of animals with spinal cord injury. Neuroprotective activity of cytoflavin was higher than that of Cerebrolysin.

Effects of cytoflavin and neuronol on morphological changes in the brain and survival of rats with ischemic disturbances in cerebral blood flow.

Cytoflavin and neuronol produce vasoactive and neuroprotective effects in rats with cerebral ischemia. Vasoactive activity of neuronol was higher than that of cytoflavin. These differences were most pronounced at the level of microcirculation. Test preparations were equally potent in producing the neuroprotective effect. Cytoflavin and neuronol markedly decreased the mortality rate of animals. Over the first 6 h of ischemia the relative effectiveness of cytoflavin was higher than that of neuronol. However, neuronol exceeded cytoflavin in the relative effectiveness during the follow-up period (days 1-21).

Direct antiviral effect of cycloferon (10-carboxymethyl-9-acridanone) against adenovirus type 6 in vitro.

Adenoviruses represent a broad group of human pathogens that currently have no specific and safe drugs for treatment. We demonstrated direct (non IFN-mediated) antiviral activity of cycloferon (10-carboxymethyl-9-acridanone, CMA), a potent interferon inducer, against adenovirus type 6 (Ad6) in Hep-2 cells. Virus production and details of morphogenesis were studied by ELISA with antibodies to the Ad6 hexon protein, and transmission electron microscopy, respectively. Immunoenzyme assay revealed that CMA does not inhibit viral protein synthesis but instead strongly reduces the ability of the virus to generate infectious progeny virus in a dose dependent manner. Ultrastructural study shows that CMA alters the structure of intranuclear virus-specific inclusions. We suggest that CMA suppresses the late stages of viral cycle in the infected cell.

Effect of neuronol on lifespan and development of spontaneous tumors in SAMP-1 mice with genetically accelerated aging.

Treatment of female SAMP-1 mice with Neuronol (drug containing succinic acid) given with drinking water starting from the age of 2 months during the whole life prolonged the lifespan and markedly reduced mortality of animals aged 1.5-2 years. Neuronol inhibited the development of spontaneous tumors, primarily lymphomas, and significantly prolonged lifespan in mice with tumors. Long-term treatment with Neuronol had no pathological side effects. Our experiments demonstrated geroprotective and anticarcinogenic activity of Neuronol and safety of its long-term use.

Inhibitory effect of synthetic interferon inductor Cycloferone on 1,2-dimethylhydrazine-induced colon carcinogenesis in rats.

The effect of a synthetic interferon inductor Cycloferone on colon carcinogenesis was firstly studied in rats. Seventy-five 2-month-old outbred female LIO rats were subdivided into three groups and were weekly exposed to 15 s.c. injections of 1,2-dimethylhydrazine (DMH) at a single dose of 7 mg/kg body wt. From the day of the fist injection of DMH rats from group 2 were given weekly i.p. injections of Cycloferone (62.5 mg/kg) until the end of the experiment. DMH-treated rats (group 3) were exposed to weekly i.p. injections of Cycloferone (62.5 mg/kg) starting in the week after the last injection of the carcinogen. Rats from group 1 were exposed to DMH and treated weekly with 0.2 ml i.p. of normal saline. Additional groups of rats were treated weekly with Cycloferone (62.5 mg/kg) or with 0.2 ml of saline. The experiment was ended 6 months after the first injection of DMH. In DMH-treated rats (groups 1, 2 and 3) colon adenocarcinomas developed in 87, 61 and 59%, respectively. The number of colon tumors per tumor-bearing rat was 2.5, 1.9 and 1.3 in groups 1, 2 and 3, respectively. Treatment with Cycloferone significantly inhibits carcinogenesis in ascending and descending colon. The incidence of tumors of the rectum was decreased in the group 2 as compared with the group 1. There were no cases of tumors of rectum in rats from group 3. The treatment with Cycloferone alone as well as with normal saline failed to induce any tumors in rats. Thus, our results demonstrated inhibitory effect of Cycloferone on colon carcinogenesis induced by DMH in rats.

[The use of cycloferon in the therapy of experimental herpetic keratitis].

The cycloferon efficacy was investigated in the treatment of experimental herpesvirus kerato-conjunctivitis in rabbits. The model was demonstrated to reflect the main aspects of herpesvirus eye lesions in humans. Cycloferon application similarly to that of known interferon inducer poludan has been shown to enhance processes of inflammation and subsequent regeneration of eye tissues as well as to decrease mortality of animals due to the generalization of infection.

[Intracellular localization of cycloferon, its binding with DNA and stimulation of cytokines expression after exposure to cycloferon].

Despite the wide usage of immunomodulating preparates including interferon inducers in medical practice little is known about their mechanism of action. We investigated some theoretical aspects of action of potent interferon inducer--cycloferon (10-carboxymethyl-9-acridanone), such as intracellular localization, ability to DNA binding and cytokine expression stimulation. This preparate has been found to be localized in nuclei of monocytic cells U-937, T- and B-lymphocytes and HeLa cells. In Hep-2 line cycloferon was bound to cells from non-adhesive subpopulation and was not detected in cells of monolayer. Human fibroblasts did not bind the substance. Interaction with double-stranded but not with single-stranded DNA occurred at pH lower than 4.7 regardless of the GC-contain. As shown by dot-hybridization cycloferon stimulated the transcription of interferon-alpha gene in U-937 cells 29-44-fold compared to the control but did not affect the transcription of tumor necrosis factor and interleukin-2 genes. Our data allow to propose that some specific receptor exists in cell with affinity to cycloferon.

[Effect of cycloferon on the evolution of cervical and vaginal cancer in mice].

7,12-dimethylbenz(a)anthracene (DMBA) was given by tampon intravaginally to 198 mice CBA (9 groups), twice a week, for 2 months, to induce cervical and vaginal tumors. Cycloferon (CF)--an interferon inductor--was injected in study group animals subcutaneously and/or introduced into the vagina by smear, in 2-month courses. CF was given during DMBA treatment or after it. Controls were injected saline solution, sc, and/or received placebo smears, i.v. The experiment was run for 6 months. Tumor incidence in controls was 35-70%. Carcinogenesis tended to slow down when CF was given during DMBA treatment. Neoplasia was found in 38-57%. More marked anticarcinogenic effect was registered in animals receiving CF on termination of DMBA application: tumor frequency dropped to 29-41%, with malignant tumor number significantly reduced.

[The use of cycloferon in experimental radiotherapy of tumors].

The biological activity of cycloferon (CF) used in interferon manufacture has been investigated for its therapeutic and prophylactic properties which may be used for radiotherapy of glioma. It was demonstrated in experiments on rats that use of CF is most effective at an early stage of irradiation due to decrease in tumor mass which does not occur in case of irradiation alone. When used at a later stage, CF caused certain hematological indices of immunity to rise. CF did not produce an antitumor effect of its own. It successfully competed with Leukomax when used to bring blood indices in irradiated tumor-bearing animals back to normal.

[Immunomodulator Cycloferon in the comprehensive treatment of patients with acute non-lymphoblastic leukemia].

The therapeutic benefit and action of the immunomodulator Cycloferon were evaluated in patients with acute non-lymphoblastic leukemia. When administered in conjunction with chemotherapy during first complete remission, it reduced both early-onset relapse frequency and degree of cytostatic-induced immune deficiency. The drug's action is mediated by changes taking place in cytokine and interferon synthesis.

[Effect of cycloferon on dissemination of Lewis lung carcinoma in mice and rhabdomyosarcoma ra-23 in rats].

Tumor growth and proliferative activity of tumor cells were suppressed and the number of pulmonary metastases in C57B16 mice decreased 3.3-fold following seven injections of cycloferon (100 mg/kg body) to induce interferon production. Injections were carried out 1-16 days after subcutaneous transplantation of Lewis lung carcinoma. After mice were immunized with ovine red blood cells, cycloferon administration raised thymus-dependent humoral immune response. After eight injections of cycloferon (50 mg/kg body) into rats, from day of intravenous transplantation of rhabdomyosarcoma RA-23 until day 20, no significant effect on metastasizing into the lung was recorded. However, single injection of cyclophosphamide 50 mg/kg inhibited metastasis formation. The highest suppressor effect was registered with combination cycloferon-cyclophophamide treatment: mean weight of metastasis decreased by half, as compared with treatment with cyclophosphamide alone. Both drugs caused karyotypical abnormalities to occur in metastatic cells. Tumor growth and spreading suppression after cycloferon should be attributed to cytotoxic antitumor action, cell proliferation inhibition and immunomodulating effect.

[The effect of cycloferon administration on the growth of transplantable tumors in rats and mice].

The influence of cycloferon and its combinations with cyclo-phosphamide on the growth of transplantable tumors was studied in rats with lymphosarcoma of Pliss and CBA and NMRI mice bearing Ehrlich's carcinoma and leukemia L1210. While cycloferon alone failed to significantly influence tumor growth, treatment with 100 mg/kg of the drug was followed by longer survival in mice with transplantable Ehrlich tumor. The highest inhibitory effect of combined cycloferon and cyclophosphamide was recorded on day 10 when tumor growth inhibition was as high as 90%. Mitotic activity of tumor was inhibited following multiple treatment with cycloferon, in combination with cyclophosphamide and cyclophosphamide alone. Survival longer than in control was registered only in animals with L1210 receiving cyclophosphamide alone or in combination with cycloferon. Cycloferon administration was shown to inhibit the growth of Pliss lymphosarcoma only within a short period after the start of the study. Combined treatment with cycloferon and cyclophosphamide produced the highest inhibition. The antitumor effect of cycloferon varied depending on tumor pattern, species and line of animals and dose and modality of administration.

[Protective effect of cycloferon in experimental influenza].

Protective effect of granulated cycloferon has been studied in albino mice infected with influenza. The drug induced interferon production and prolonged the life span, decreased the mortality, suppressed the virus reproduction in the lungs, and decreased the infective activity of the virus. Morphologically the drug decreased the intensity of viral lesions in the bronchiolar epithelium, impeded the infection dissemination, and stimulated the productive (cellular) component of local inflammatory reaction. No influence on the development of chronic lesions in the lungs was detected.
