Changes in vascular ionic composition at different stages of DOC-salt hypertension in the rat.

1. Sequential changes in the ionic composition of the aorta and skeletal muscle were followed during 1, 2 and 4-6 weeks in 30 rats given deoxycorticosterone (DOC) and salt supplemented with potassium chloride. Twenty-one rats, drinking water, were used as controls. 2. Twenty-five per cent of the test rats were hypertensive after 1 week, 60% after 2 weeks and 100% after 4-6 weeks. 3. Muscle potassium fell in all test rats by an average of 15%. In contrast, aortic potassium fell by 19% only in those rats which did not develop hypertension after 1 week. 4. Total and non-inulin sodium and water of the aorta were normal in rats which remained normotensive after 1 or 2 weeks and high in those which became hypertensive during the same period. 5. Total sodium and water content of the aorta were also high in rats which were hypertensive at 4-6 weeks. However, because of simultaneous expansion of the inulin space, non-inulin fractions were normal in this group. 6. Results suggest that vascular ionic changes participate in the pathogenesis of DOC-salt hypertension through more than a single mechanism.

Radical formation in single crystals of aromatic amines and radical transformation by light.

Single crystals of the four aromatic bioamine salts phenylethylamine hydrochloride, tyramine hydrochloride, tryptamine hydrochloride, and histamine dihydrochloride were grown in various states of deuteration. Free radicals were produced by exposure to X-rays between 77 and 300 K and investigated by electron spin resonance spectroscopy. Dissociation of atomic hydrogen from C beta of the aliphatic chain occurs in all compounds studied except tryptamine. However deamination as usually present in the analogous amino acids is not found. The C beta-radical is characterized by an anisotropic H alpha-splitting and two isotropic H beta-splittings. The latter splittings depend strongly on temperature in tyramine. In comparison to the analogous amino acids, radical formation in the aromatic residues is favoured. Among the seven different aromatic radicals found only one is identified in histamine but two in each of the three other bioamines. Two of these are characterized by hydrogen dissociation which occurs in phenylethylamine and tyramine. One hydrogen addition radical is found in each of the three compounds phenylethylamine, tyramine and histamine. In tryptamine two different addition radicals are detected. One of the two products can be converted into the other by visible light. The reverse process is induced by heat, thus permitting the switching of the radical site reversibly between two different structures.

Metabolic products of microorganisms. 200 Isolation and characterization of niphithricins A, B, and elaiophylin, antibiotics produced by Streptomyces violaceoniger.

Fermentations of Streptomyces violaceoniger TU 905 produce the antifungal antibiotics niphithricins A, B, elaiophylin and nigericin. The niphithricins have been characterized as new macrolide antibiotics, and the previously unknown structure of elaiophylin was determined to be a macrodiolide. The niphithricins were biologically active against Gram-positive bacteria and fungi. The mode of action is attributed to an alteration of the membrane permeability.

[Microaggregate formation in buffycoat-free packed red cells (author's transl)].

The amounts of microaggregates of 200 non-selected units of buffycoat-free PRC were determined gravimetrically using a cascade of nylon mesh screen filters with pore sizes of 200, 50, 20 and 10 micrometers. In PRC the total amounts of amorphous debris are on average 2 to 3 times less than in stored whole blood. Nevertheless the quality of the buffycoat-free PRC is subject to immense variation in view of removed debris in individual cases. Microaggregates retained on the 20 and 10 micrometer filter sieves are still considerable, their order of magnitude is comparable to that found on the 200 micrometer sieves, so that a microfiltration is also necessary under 40 micrometers. Taking into account the necessity of eliminating all factors which contribute to the cause or severity of an acute pulmonary failure even buffycoat-free PRC should be transfused through microfilters.

[Lichen myxedematosus and multiple myeloma type IgG/kappa].

Lichen myxedematosus and scleromyxedema are rare disorders with cutaneous deposition of mucin, without any disturbance of the thyroid function. They are regularly associated with the presence of abnormal proteins in the serum. These proteins have been now identified as paraproteins and are one of the main symptoms of these diseases. The association of a papular mucinosis with a classical plasmacytoma is exceptional. We report a patient in whom lichen myxedematosus is associated with a multiple myeloma of the type IgG/kappa. Cultured skin fibroblasts show a marked decrease of arylsulfatase A activity. An antineoplastic-steroid therapy has brought a remission of both plasmacytoma and skin lesions.

[Investigation of the exposure to radiation of the embryo in the course of x-ray examination during pregnancy. 2nd communication: Estimation of the exposure to radiation using the area-dose or the tube-current-time product (author's transl)].

Thermoluminescence dosimetry of surface doses and doses near the uterus was performed during standardized X-ray examinations of the pelvis. Surface doses and the dose to the uterus were estimated from the available radiographic data referred to the tube and with the use of tissue/air ratios, and a comparison with the results of measurement was made. This comparison shows that the mean whole-body dose to the embryo can be estimated with a relative total risk of 20%, if all essential data of the exposure to radiation are present and if the position in depth of the embryo is known. Guiding data of the surface dose and of the dose to the uterus are given individually for fluoroscopic tests, a.-p. and lateral radiographs under standardized conditions of X-ray examination.

[Granulocytes in the oral cavity (author's transl)].

The counting of the granulocytes in the oral cavity ("salivary corpuscles", orogranulocytes") gives an indication to the efficiency of the granulocytopoiesis, because there exist some analogies to the emigration of granulocytes into the tissue. Two different counting methods of the orogranulocytes, their normal values (which are in correlation to the number of teeth), the influence of local factors (like inflammations in the oral cavity), the values of orogranulocytes in myeloproliferative disorders and in agranulocytosis and granulocytopenia respectively are demonstrated. Furthermore the contributions deal with the values of the orogranulocytes during the therapy with glucocorticoids. The methods described are also suitable for the estimation of the efficiency of granulozyte transfusions.

Morphology and morphogenesis of a new paramyxovirus (PMV 107).

The morphology of the virions and nucleocapsids of paramyxovirus 107 (PVM 107) and the replication of the virus were investigated by electron microscopy. The virions and nucleocapsids exhibited the same structural properties as other paramyxoviruses. Nuclecapsids were found in the nucleus and cytoplasm of infected bovine embryonic lung (BEL) cell cultures. A similar situation has been described for the morbilliviruses measles, SSPE, distemper and rinderpest. Alignment of nucleocapsids beneath the plasma membrane and budding of PMV 107 in the productive BEL cell infections were also similar to the morbillivirus-infected cells. In a line of monkey cells (CV1) persistently infected with PMV 107 only cytoplasmic nucleocapsids could be demonstrated. On the basis of its morphology and morphogenesis it is suggested that PMV 107 should be classified as a paramyxovirus. Since nucleocapsids could also be found in the nucleus of infected BEL cells the morphogenesis of PMV 107 closely resembles that of viruses of the morbillivirus group.

Aggregation of sponge cells. Isolation and characterization of an inhibitor of aggregation receptor from the cell surface.

From the cell membranes of the sponge Geodia cydonium a component was isolated and purified which inhibits the aggregation factor isolated from the same source; the component was termed anti-aggregation receptor. This molecule was characterized as a glycoprotein (54% neutral carbohydrate) and its molecular weight is in the range of 180,000 One biological site of the anti-aggregation receptor was determined to be D-galactose. Indirect evidence presented seems to indicate that this molecule is present in an active form in aggregation-deficient cells and absent in aggregation-susceptible cells.

[Levels of immunoglobulins and numbers of lymphocytes in the venous blood from the neighbourhood of oral carcinomas Relations to short-time-prognosis (author's transl)].

Lower levels of immunoglobulins A and G than in a cubital vein were found in the venous blood from the neighbourhood of non-metastatic oral carcinomas. The authors these results interprete as an increased connection of antibodies to the receptors of tumor-cells. It is evident that the receptors are saturated in metastatic carcinomas. Favourable shorttime-prognosis is often connected with an increased number of lymphocytes in blood of tumour- and cubital-vein.

In vivo measurement of glucose concentration with lasers.

Common polarimeters defined the concentration of an optically active sample by measuring the intensity of the light beam after it has passed through the analyser. Consequently, it can be stated that up to now, there have been the disadvantages of having a signal dependent on absolute light intensity. The new method that is described has a sensitivity that is in great measure independent of absolute light intensity, whereby only one light trace is necessary. The new principle uses no mechanical rotations. Instead, an electrical signal indicates the amount of optical rotation of the sample. The high sensitivity that can be reached is theoretically only limited by polarization noise. By going to the uttermost physical and electronic lengths, sensitivity values of more than 10(-5) degrees can be reached. Furthermore, the mechanical dimensions of the apparatus can be made very small by the application of a solid-state laser.
