By using microanalytic Hydroxyproline (HP)-determination method (Woessner and Stegemann) the content of collagen in 50 normal and pathologicaltered placentae of the second part of pregnancy was measured. A positive linear correlation is demonstrated between HP-concentration and age of pregnancy in normal placentae. Most of the pathologic altered placentae have a lower content of collagen. HP-content measured in main stem and terminal villi of further 35 placentae showed the essential influence of the main stem villi on total placental collagen concentration. In the second part of gestation the HP-concentrations of the main stem villi rise on a factor about 5-6, in the terminal villi they remain constant. The values measured within both groups of villi show a great dispersion. The quantitative results were compared with the histology of the placentae.
Uncalibrated flow in the vertebral arteries was registered with a bidirectional CW Doppler ultrsound apparatus. It was applied to 20 patients, results being compared with aortic arch angiograms in which the extracranial part of all four cerebral vessels was seen. In 14 the vertebral arteries had a symmetrical flow and lumen, a similar symmetrical result being obtained with the Doppler. Hypoplasia of one vertebral artery was present in two patients the distinct asymmetry being recorded by the ultrasound. Three patients with central stenosis in one subclavian artery and subclavian "steal" were found to have reverse flow by the Doppler method in the vertebral artery on the side of the stenosis. Endarterectomy was performed on the affected subclavian artery in two of these patients: post-operative vertebral ultrasound recordings demonstrated a return to physiological flow direction in both vertebral arteries.
A study group of 623 employed Swiss women aged 30-49 years showing objective evidence of intake of phenacetin-containing analgesics, and a control group of 621 comparable women showing no such intake, were observed for 4 years (1969-72) for laboratory evidence of urorenal disorders. In both study and control groups morbidity was low. There was no difference between the study and control groups with respect to subsequent proteinuria, bacteriuria, and haematuria. The 4-year incidence of low urine specific gravity after overhight thirsting was significantly higher in the study group than in the control group (3-8% v. 0-8%) and the incidence of raised serum-creatinine was also significantly higher in the study group (2-9% v. 0-4%). However, when the study group was further subdivided into a sub-group showing evidence of high intake of phenacetincontaining analgesics and one showing low intake, only the high-intake subgroup had an incidence of raised serum-creatinine (5-4%) significantly higher than the control group (0-4%), whereas the low-intake subgroup had an incidence (0-4%) similar to the control group.
The solution conformations of adenosine, guanosine and inosine in liquid ND3 have been determined by NMR. Comparison of the Karplus analysis of the proton HR spectra of the ribose moiety obtained in this solvent with the data from aqueous solutions of A and I proves that the conformations of the nucleosides are very similar in both liquids. From the analysis of the vicinal coupling constants of the ring protons it has been deduced that the S state C(2')-endo is slightly preferred. The mole fraction in S approximates 0.6 for all three nucleosides. C-13 relaxation measurements have been applied in the determination of the correlation times for rotational diffusion. Only at temperatures below - 40 degrees C is the pseudo-rotation of the furanoside ring slowed down sufficiently for it not to contribute to the measured relaxation rates. From NOE studies and T1 measurements on the individual protons it is derived that the N, C(3')-endo, form of the ribose is correlated with an anti conformation of the base (Y approximately 210 degrees to 220 degrees) and the S, C(2')-endo, form of the ribose with a syn conformation of the base (Y approximately 30 degrees to 50 degrees). The glycosyl torsion angles derived for the two conformations of A, G, and I are equal within the limits of accuracy.
Radicals produced at temperatures between 77 K and 300 K by X-irradiation of L-histidine.2HC1 crystals have been analysed by electron spin resonance spectroscopy. The results are compared with previous findings with L-histidine and L-histidine.HC1.H2O crystals. In these three crystals we have identified three main species, which, depending on the molecular environment, are stabilized at different temperatures and produced in different concentrations. These findings are discussed in connection with the observations made on irradiated aqueous solutions of amino acids. It is concluded that our results corroborate the mechanism for the action of ionizing radiation on dry protein proposed by Platzman and Franck (1958),
Radicals produced by X-irradiation at 77 K of 1-methyluracil. HBr crystals have been analysed by electron spin resonance spectroscopy. The results are compared with those obtained on 1-methyluracil crystals. Four radicals have been identified, two of which are present only in 1-methyluracil. HBr crystals: the C6-addition radical and the pyrimidine--anion radical. Warming-up experiments have been performed in order to study the secondary radical reactions. The different mechanisms proposed for the radiolysis of DNA constituents in the solid state are discussed in connection with these findings.
Radicals produced by X-irradiation at 77 K and at 300 K of cytosine. HCl crystals have been analysed by electron spin resonance spectroscopy. Four radicals have been identified: the anion radical of the cytosine molecule, the radical resulting from H-addition at position C6, the radical resulting from H-addition at position O2, and finally a radical resulting from addition of a Cl- to nitrogen N3. Hückel molecular orbital calculations are presented, which support the hypothesis according to which in unsaturated pyrimidines the site of hydrogenation or protonation depends on the state of the molecule.
