Streptomycin slightly inhibited lactic and malic dehydrogenases of Bacillus subtilis, and inhibited isocitric dehydrogenase to about 60%. The formation of lactic dehydrogenase, glutamic dehydrogenase, alpha-alanine dehydrogenase, and succinic dehydrogenase was stimulated by the antibiotic at a concentration causing 50% inhibition of bacterial growth. Streptomycin had practically no influence on the formation of malic dehydrogenase, but the antibiotic produced 48% inhibition of the synthesis of isocitric dehydrogenase.

This report describes treatment combining Ultrapulse CO(2) laser and Q-switched frequency-doubled neodymium:yttrium-aluminum-garnet (Nd-YAG) laser for a number of congenital nevocellular naevi (CNN). The Ultrapulse CO(2) laser removes the superficial component, which contains most of the color, preserving the integrity of the reticular dermis. At a later stage, any residual color is targeted using the Nd-YAG laser. To date, a total of 10 histologically proven CNNs have been treated at the Laser Suite. The median age of the patients was 18 years (range, 13-24 years). All the patients were white females. The patients accepted for treatment had relatively small lesions (mean surface area, 352 mm(2); range, 50-1,092 mm(2)), but the lesions were either cosmetically sensitive or located in an anatomically critical area in which excision could result in a scar that would be unpredictable. The median follow-up period was 24 months. The overall results have been satisfactory, with reduction of color and no significant scarring. The minor textural and pigmentary changes were acceptable to the patients.
To study coagulation of human ejaculates, fresh semen samples were classified into coagulated (CG), fully-gelified (FG), partly-gelified (PG) and liquid (LQ) groups. The groups were identified physically, morphologically and biochemically. Physically and biochemically, there was no essential difference between the CG and FG ejaculates, but they differed markedly from those of the PG and LQ samples. Coagulation did not occur in ejaculates showing decreased vesicular activities. PG and LQ ejaculates demonstrated significantly higher prostatic activities from those of the CG and FG samples.
Liquefaction of human semen in vitro was studied by scanning electron microscopy, determinations of total protein, free amino acid and phosphate and by correlation analysis. During liquefaction of the coagulum, its fibers were transformed into globular bodies that fused to form the homogeneous liquefied semen. It was found that the parts of the ejaculate taking more time to liquefy possessed higher concentration of protein than those of the parts liquefied earlier, a relation also appeared to exist with the whole ejaculates. Release rate of phosphate during macroscopic lysis and free reducing sugar level of ejaculates demonstrated significant negative correlations with liquefaction time which could be shortened using reductants.
The predominant acidic coagulum protein of freshly ejaculated human semen under reducing sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) was purified for the first time from washed seminal coagulum keeping urea throughout the entire procedure. Washed seminal coagulum was isolated from fresh semen sample in acetate buffer at pH 4.5 at 0 degrees C, solubilized rapidly in deionized 4 M urea followed by reduction and carboxymethylation of disulfide bonds. By CM-Sephadex and Sephacryl S-300 high-resolution chromatography in presence of urea at room temperature, the predominant acidic coagulum protein was highly purified (99.1%) with a yield of 2.7% of the coagulum protein. The molecular weight of the purified protein in reducing SDS-PAGE was 162 kD.
Treatment of severe Pseudomonas infections often calls for multidrug therapy. Combinations of aminoglycosides, beta-lactams and/or quinolones are generally administered, the effect of which may be additive, synergistic or indifferent. The present in vitro study was designed to find out the exact outcome of the postantibiotic effect (PAE). The minimal inhibitory concentrations and the PAE of three antibiotics (amikacin, ceftazidime and ciprofloxacin) were determined both singly and in combination. Ceftazidime alone exhibited a negative PAE but a synergistic effect was observed for the combination of ceftazidime with amikacin. The combination of ceftazidime with ciprofloxacin and ciprofloxacin with amikacin however gave an indifferent PAE. This synergistic PAE of amikacin with ceftazidime has a significant effect on designing optimal dosage regimens.
Dysplasia of the fibrous sheath (DFS) is characterized by male infertility, asthenozoospermia, and morphologically abnormal flagella that possess a severely malformed fibrous sheath. In many cases, DFS is familial, suggesting a genetic component. Human AKAP4 and AKAP3 are structural proteins of the fibrous sheath that also function to anchor protein kinase A to this structure via the regulatory subunit of the kinase. We hypothesized that defects in either AKAP4 or AKAP3 might cause DFS. No quantitative or qualitative differences between patients with DFS and normal controls were detected when sperm proteins were analyzed by either silver staining or immunoblot analysis using antibodies raised against AKAP4 and AKAP3. Additionally, AKAP4 and AKAP3 from DFS sperm retained the ability to bind the regulatory subunit of protein kinase A. Localization at the light and electron microscopic levels showed that AKAP3 and AKAP4 localized correctly to the FS of the amorphous flagellum in DFS sperm. Partial sequence analysis of the AKAP4 and AKAP3 genes in patients with DFS did not identify any significant alterations in potential AKAP4/AKAP3 binding regions, suggesting that the two proteins interact normally in DFS sperm. Our results did not find evidence to support the hypothesis that mutations in either gene are responsible for DFS in humans.
