Increased vascular collagen biosynthesis by hypertension and reversal by antihypertensive drugs.

Collagen synthesis was increased in aortas, mesenteric arteries, cerebral microvessels, pial artery, basilar artery and decreased in the heart of rats made hypertensive with deoxycorticosterone acetate-salt or the spontaneously hypertensive animal. The markers of collagen biosynthesis that were elevated were prolyl hydroxylase, prolyl hydroxylase-related antigen, total collagen content and the incorporation of labelled proline into total protein and into collagen. The antihypertensive drugs reserpine and chlorthiazide could both prevent the increase or reduce the increase in collagen synthesis.

Localization of prolyl hydroxylase by the immunoperoxidase method in cardiovascular tissues of hypertensive rats.

An immunohistochemical method and light microscopy were utilized to determine localization of prolyl hydroxylase in the cardiovascular tissues of hypertensive rats. The blood vessels and renal glomerulus from these animals demonstrated an enhanced immunoreaction for prolyl hydroxylase. As the most prominent staining was observed in medial smooth muscle cells of blood vessels, these are probably the major collagen-producing cells in arteriosclerotic lesions induced by hypertension. In the glomerulus, endothelial cells and probably mesangial cells are mainly responsible for the collagen formation. An intense immunoreaction was also found in the fibroblasts which proliferated in the area of myocardial fibrosis in hypertensive rats, while there was no specific staining in myocardial cells in either hypertensive or normotensive rats.

Glucose metabolism in spontaneously hypertensive rats.

Serum glucose levels in SHR (especially SHR selected for a greater hyperglycemic response) tended to be higher at 'fasting' and were significantly higher after peroral glucose administration than those in normotensive WK. However, plasma immunoreactive insulin was not decreased in SHR as compared with WK. No significant acceleration of gluconeogenesis was observed in SHR.
