Cholecystokinin-like bioactivity in rat cerebral cortex.

Cholecystokinin (CCK)-like bioactivity was studied in extracts of rat cerebral cortex by three different bioassays. Positive results were obtained by all procedures used. A fairly good quantitative correlation was found when testing the same extracts by two of the methods. A considerable amount of CCK-like activity was demonstrated in the various subcellular fractions (synaptic membranes, synaptic vesicles, and synaptic fragments) corresponding to synaptic structures. The results are in accordance with the findings of several authors who demonstrated CCK-like immunoreactivity in the brain, especially in the cortex. The use of three different bioassays in three species (rabbit, rat, dog) seems to prove that some bioactive molecular form of CCK exists in the brain, mainly in the cortical area. The findings that subcellular elements of the synaptic structures contain relatively high concentrations of CCK raises the question of the possible role of CCK in the neuronal transmission.

[Early gastric carcinoma detection by endoscopy and biopsy; clinical experiences (author's transl)].

This report presents the clinical features of 15 cases of early gastric cancer detected amont altogether 144 gastric cancers diagnosed with the aid of gastroscopy and directed biopsy in a series of 2953 patients. Since the clinical and laboratory data of the patients with early gastric cancer do not provide satisfactory diagnostic evidence, careful X-ray examination must be carried out in all cases with long-standing symptoms relating to the upper part of the abdomen, especially in patients over 40 years old. Gastroscopy is essential in the clarification of any suspected pathological signs indicated by the radiologists and, furthermore, in all cases in which no radiological diagnosis was reached to account for the patient's symptoms. During gastroscopy multiple biopsy specimens should be taken from all lesions which may mask any type of the early cancers. The importance of regular endoscopic examination of high-risk persons for gastric cancer is emphasized.

Effect of atropine on different phases of pancreatic secretion in conscious rats.

The effect of submaximal doses of intravenous atropine and their combination with topical anaesthesia of the intestine by oxethazaine-HCl was investigated in conscious rats. I. Basal water, bicarbonate and protein secretion were significantly augmented after diversion of pancreatic juice. On the basis of the protein secretory pattern of basal secretion, 2 stable stages have been recognized: 1. The most physiological basal stage during return of pancreatic juice. 2. The first, highly elevated plateau from the 4th to the 7th 30 min period after diversion. 3. The second delayed and less elevated plateau after 240 min diversion. II. Atropine greatly suppressed protein secretion during recirculation but only moderately after diversion of juice, and during the first plateau an atropine resistant peak appeared. Inhibition of water secretion was equal during all the stages. Atropine infusion resulted in a further decrease in pancreatic secretion also after topical anaesthesia of the intestine. It was concluded that atropine had a complex effect on pancreatic secretion: it possibly decreased the CCK release in the first period after diversion but not later, and decreased the duodenopancreatic reflexes and other factors of the cholinergic tone.

Exogenous and endogenous stimulations during different phases of pancreatic secretion in conscious rats.

The effects of stepwise increasing doses of intravenous caerulein, secretin, urecholine and intraduodenal oleic acid were investigated in conscious rats. a) Basal water, bicarbonate, and protein secretion significantly augmented after diversion of pancreatic juice. On the basis of protein secretory pattern of basal secretion 3 stable stages have been recognised: 1. The most physiological basal stage during return of pancreatic juice. 2. The first, highly elevated plateau from the 4th to 7th 30-min period after diversion of pancreatic juice. 3. The second, delayed and less elevated plateau after 270 min. b) The following patterns of stimulated secretion were observed: 1. Caerulein stimulated protein and bicarbonate secretion during return of juice and the first plateau but it failed to elicit a significant response during the second, delayed plateau. 2. Secretin and urecholine showed similar protein responses during return of pancreatic juice but after diversion, they stimulated water and bicarbonate secretion only. 3. During the second plateau oleic acid stimulation resulted in a significant increase in water and bicarbonate secretion but no significant increase occured in protein output. The delayed inhibition of protein secretion, previously described in our laboratory, was unchanged. 4. Supramaximal doses of exogenous stimuli caused an inhibition in pancreatic secretion.

The effect of gastroduodenal xylocaine on the basal pancreatic secretion in rats.

The effect of intragastric and intraduodenal xylocaine on the pancreatic secretion was investigated in conscious rats. A 1% xylocaine infusion strongly decreased pancreatic protein output and flow during recirculation of pancreatic juice into the duodenum, but it diminished only moderately the increase in the secretion seen after its diversion and after duodenal infusion of soybean trypsin inhibitor. Moderate inhibition appeared during prolonged diversion of pancreatic juice. The response to exogenous CCK + secretin was not modified. The results indicate that during recirculation of pancreatic juice, which is more physiologic than its diversion from the duodenum, the neural control may play a significant role.

Pancreatic amylase- and protein-concentrations associated with thioacetamide-induced cirrhosis of the liver in rats.

In cirrhosis of the liver induced in rats by administration of thioacetamide for 8 months, pancreatic amylase and protein concentration and the number of zymogen granules were found to exceed those of the controls. The finding is attributed to a deficient metabolism of enterohormones. The model seems to provide a suitable approach to the study of the hypersecretory state of the pancreas associated with cirrhosis of the liver.

Effect of alpha-pinene on the mixed function microsomal oxidase system in the rat.

The microsomal fraction of the liver was studied for protein and cytochrome P 450 contents as well as for aminopyrine N-demethylase and aniline hydroxylase activity and for its butylisocyanide spectrum under the effect of alpha-pinene treatment. A moderate increase was found in the amount of microsomal protein and cytochrome P 450, a marked increase in the metabolism of both aminopyrine (substrate of type I) and aniline (substrate of type II), together with an increase in the butylisocyanide-absorption maxima of both haemoproteins involved in the reaction. Thus, alpha-pinene is a broad-spectrum enzyme inductor of the phenobarbital type, with certain quantitative differences.
