Cytogenetic and immunohistochemical profile of myxoid liposarcoma.

Cytogenetic and immunohistochemical studies were performed in nine myxoid liposarcomas. The tumor karyotype was determined after short-term culture of cells in vitro. Immunohistochemical studies were performed on frozen tissue in five cases and on paraffin-embedded tissue in three cases. Chromosomal analysis demonstrated a balanced translocation t(12;16) (q13;p11) as the sole abnormality in four cases. Two cases showed an association with other abnormalities. Three tumors showed variants of the t(12;16) translocation involving other chromosomes. In all cases studied, the 12q13 breakpoint was involved in rearrangements. In the majority of cases, immunohistochemical studies demonstrated vimentin (9 of 9) and S-100 protein (8 of 9). Strong focal expression of desmin was observed in two tumors. Weak focal expression was observed in three tumors. Two tumors, which were both desmin positive, showed focal expression of MSA and alpha-SMA. Strong expression of CD36 was present in all four cases that were studied for this marker. CD34 was negative in tumor cells, but it highlighted an intricate capillary network in the tumor. Close relationship between the tumor cells and pericapillary pericytes was demonstrated with CD34 and alpha-SMA strains. The authors conclude that myxoid liposarcoma is characterized by a specific chromosomal rearrangement. Its immunohistochemical profile is wider than previously believed, including expression of muscle markers.

The effect of high fructose intake on renal morphology and renal function in rats.

The aim of this study was to investigate the effect of 2-week fructose feeding on plasma lipids renal function and morphology in the rats. Pair-fed rats were kept individually in metabolic cages for two weeks and given diet containing either 25% fructose or 25% starch (weight/weight). After that time under Nembutal anesthesia blood pressure was measured intra-arterially and blood was withdrawn for cholesterol, triglyceride, insulin, glucose and 24hrs urine was collected to estimate creatinine clearance. At the end of experiment kidneys were removed and fixed for microscopic examination. Creatinine clearance was higher by 15% (p < 0,01) in fructose fed rats and microscopic examination revealed moderate mesangial cells proliferation in animals of the same group. No statistical significant difference was observed with regard to lipids or glucose and insulin between two groups. Our study suggest that fructose feeding is associated with hyperfiltration and hyperplasia of mesangial cells in glomeruli.

p53 expression in breast cancer related to prognostic factors.

In this article the results of molecular marker p53 examinations were presented in relation to the following established breast cancer prognostic factors: age, histologic type, histologic grade, lymph node involvement, tumor size as well as estrogen a progesterone receptor status. Twenty one percent of these primary breast cancer specimens exhibited the overexpression of p53 protein. Significant associations were found between p53 overexpression and younger age, high histologic grade and low content of estrogen and progesterone receptors. Identification of p53-positive breast carcinomas potentially represents a clinically useful indicator of breast cancer aggressiveness.

[Level of estrogen receptors (ER) as a function of histological malignancy stages of tumors in patients with breast neoplasm].

The presence of estrogen receptors (ER) has been studied in 70 patients with primary operable breast cancer. The tumors were graded histologically according to Bloom and Richardson criteria. A significant correlation was found between these two prognostic factors: the better--differentiated tumors (I + II histological grade) had significantly higher level of ER than that in the poor--differentiated tumors. Estrogen receptor content of primary breast cancer as a function of tumor grade can be useful in selection of the most efficient treatment modalities for individual patients.

Ultrastructural changes in liver after the test exposition of laser irradiation.

The aim of this research was to verify characteristic morphological changes in the liver with regard to survival time after laser irradiation. The duration of irradiation was constant. For the purpose of irradiation we used a helium-neon laser. The value of the energy absorbed during the exposition amounted to about 2.5 J. Segments from the irradiated liver field of rats were taken directly after decapitation following different times of survival, and the changes were estimated in an ultrastructural microscope. Our results showed dependence of certain morphological changes (perceptible only by ultrastructural examination) on the time of survival after irradiation.

The effect of indomethacin on the ultrastructure of renomedullary interstitial cells of the rat.

The administration of the prostaglandin synthetase inhibitor indomethacin to the rat result in a significant increase of the number as well as the size and density of the lipid granules of renomedullary interstitial cells. The increased granularity is associated with the ultrastructural alterations of the organelles which are characteristic for cells with small secretory activity (a sparse rough and smooth endoplasmic reticulum and a small GOLGI complex). The findings indicate that there exists an inverse correlation between the number of the lipid granules of the interstitial cells and the secretory activity of these cells and seem to support the hypothesis that the interstitial cell lipid granules serve as storage site for renal prostaglandin precursors.

The ultrastructure of renomedullary interstitial cells in hypercalcemia induced by calcium gluconate.

Under hypercalcemia induced by calcium gluconate the degranulation of renomedullary interstitial cells accompanied with the increase in volume of the rough and smooth surfaced endoplasmic reticulum and enlargement of the Golgi apparatus were observed. The ultrastructural changes can be regarded as an expression of the increase of a synthetic and secretory activity of these cells. Because the changes of renomedullary cells observed in hypercalcemia induced by calcium gluconate are not really different from that observed in hypercalcemia induced by vitamin D3 (Roszkiewicz et al. 1979) it can suppose that these changes may be connected solely with a calcium serum elevation and that the renomedullary interstitial cells rather do not play any important role in vitamin D3 metabolism.

The ultrastructure of renomedullary interstitial cells in short duration hypercalcemia induced by vitamin D3.

Under short duration hypercalcemia induced by pharmacological doses of vitamin D3 significant ultrastructural changes were observed in the renomedullary interstitial cells of rats. The most striking alteration was the degranulation of these cells accompanied with the increase in volume of the rough and smooth-surfaced endoplasmic reticulum, enlargement of the Golgi apparatus and occurence of osmiophilic inclusions probably of lipid nature in mitochondria. The ultrastructural changes can be regarded as an expression of the increase of a synthetic and secretory activity of the renomedullary interstitial cells and they may be associated with an enhanced production of prostaglandins or other lipid hormonal substances than prostaglandins under condition of hypercalcemia.

Ultrastructure of the C cells forming adenoma-like nodules of the thyroid in experimental hypervitaminosis A.

The degranulation of the C cells, their hypertrophy and hyperplasia leading to the formation adenoma-like nodules were observed under conditions of prolonged hypervitaminosis A which causes bone damage without associated hypercalcemia. These changes which are probably the morphologic manifestation of the increased requirement of the body for calcitonin, connected with the damage of the bone tissue seem to indicate that calcitonin is an important factor essential for the protection of skeleton from its excessive mineralization.

Ultrastructure of cells of proper gastric glands and their stroma in hemorrhagic shock in the rat.

Ultrastructural changes in the cells of the proper gastric glands and their stroma are biphasic in the hemorrhagic shock. First phase: "paralysis" of the capillary vessels with an oedema of their stroma, an intracellular oedema with hydropic degeneration of the parietal cells and a degranulation of the argentaffin and argentaffin-like cells. Second phase: ischemia of the gastric mucosa with ultrastructural features of: a) the increased secretion of the parietal cells, b) the degranulation of the chief gastric cells, c) the increased secretion processes in the mucous neck cells. These findings suggest that at last 2 factors coincide in the pathogenesis of so-called stress gastric ulcerations: biphasic disturbances of blood circulation initially damage the gastric mucosa and are followed the digestive hyperactivity of the gastric juice finally resulting stress ulceration.
