Dendritic and cancer cells in the breast tumors--an immunohistochemical study: short communication.

Normal and dysplastic mammary glands express immunocompetent S-100 protein positive dendritic cells (DCs), which are located in a regular pattern, in the suprabasal cell layer of the ducts and alveolar nodules. The epithelial cells, however, are S-100 protein negative. Since some breast cancers also express the S-100 protein, our aim was to check the diagnostic and prognostic value of the S-100 protein distribution combined with the tumor grade and expression of synaptophysin (Syn), chromogranin A (Chg A), c-erbB-2 oncoprotein and p53 protein in infiltrating and metastatic breast tumors. Applying immunohistochemical methods, we show in paraffin- or frozen breast tissue sections that in some cases of the infiltrating breast carcinomas, S-100 protein positive cells do not appear, whereas in other cases, either S-100 protein positive DCs are closely associated with cancer cells, or the cancer cells themselves stain positive to S-100 protein. However, we found no correlation between the S-100 protein expression and other investigated parameters.

Nitric oxide synthase type 1 expression in human lung cancer and its relation to p53.

BACKGROUND: The nitric oxide synthases (NOS) have been observed in human tumour cell lines as well as in solid tumours. Neuronal isoform of NOS (NOS1) was particularly abundant in low-differentiated gynaecological, breast and central nerve system tumours, suggesting that it may characterize poorly differentiated tumours. So far, little is known about expression of the neuronal NOS isoform in non-small cell lung cancer. Our aim was to compare NOS1 expression in non-small lung carcinomas with respect to tumor staging and p53 protein expression. MATERIAL AND METHODS: Thirty-two cases of non-small lung carcinomas of all grades of malignancy and ten control lung specimens with neoplastic lesions were examined. Paraffin-embedded tissues were stained with hematoxylin and eosin, or with antibodies to NOS1 and p53, and evaluated under light microscope. RESULTS: No statistical correlation between expression of p53, NOS1 and degree od tumour differentiation was observed. CONCLUSION: Expression of NOS1 can not serve as a marker for highly malignant tumours in the non-small cell lung carcinomas.

[Fibronectin in the skin of patients with allergic dermatitis].

In the skin of healthy subjects linear deposits were found of fibronectin at the dermoepidermal junction and in the walls of blood vessels, and reticular deposits in the papillary and reticular layers of the skin. In the epidermis of 12 out of 51 patients with allergic contact dermatitis fibronectin deposits were found in relation to Langerhans cells and keratinocytes. This was not observed in the epidermis in healthy subjects. The authors suggest that fibronectin on the surface of Langerhans cells and keratinocytes may play a role in antigen presentation.

[Ultrastructural image of the Langerhans cells in the skin of patients with allergic contact dermatitis].

The alterations in the ultrastructural feature of Langerhans cells in patients with allergic contact eczema were characterized by significant increase in number of Birbeck granules accompanied by the increase in volume of smooth- and rough-surfaced endoplasmic reticulum. These changes may be regarded as a morphological expression of the increase of the synthetic and secretory activity of the Langerhans cells. In the epidermis the Langerhans cells associated with immediately adjacent precursor cells (the so called indeterminate cells), lymphoid cells and keratinocytes form the morphologically visible and probably functional units.

Influence of oral glucose load upon blood glucose level in relation to the time of day.

Blood glucose levels were estimated at different times of day in fasted rats and after 30, 60,90 and 120 min, since oral glucose load. Circadian variations in basal glucose levels and in the levels after glucose load were observed with the highest values noted between 11 a.m. and 7 p.m., and the lowest ones about midnight. These variations were most prominent when the measurements were performed 60 min after glucose load. Circadian variation in glucose tolerance was also revealed with the best tolerance at about midnight while the worst one was noted at noon and in the afternoon.

Heparin and nitric oxide treatment in experimental acute pancreatitis in rats.

The aim of this study was to investigate the impact of L-arginine (nitric oxide synthase substrate), L-NG-nitro-L-arginine (nitric oxide synthase inhibitor), and heparin on the pancreas microcirculation, serum IL-6 level and microscopic alterations of the pancreas in acute pancreatitis in rats. Acute pancreatitis was induced by 4 i.p. injections of cerulein (15mg/kg). Microcirculatory values were measured by means of laser Doppler flowmetry 5 h after the first cerulein injection. Remarkable histopathological changes in the pancreas, including parenchymal necrosis, an elevation of serum IL-6 level, and a significant drop of pancreatic capillary perfusion was observed in rats with nitric oxide synthase inhibition. L-arginine improved the pancreatic microcirculation but worsened the microscopic alterations within the pancreas. Heparin had a beneficial effect on the microcirculatory values, serum IL-6 concentration, and morphologic changes. Authors conclude that inhibition of nitric oxide synthase aggravates acute pancreatitis. L-arginine treatment improves pancreatic perfusion but potentiates morphological alterations. Heparin, improving the microcirculation and inflammatory changes within the pancreatic gland, may be considered as a promising therapeutic agent in acute pancreatitis.
