Comparison of intra- and extrasynaptosomal monoamine oxidase-A and -B activities in the striatum and frontal cortex of two mice strains with different sensitivities to the neurotoxic actions of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine.

Conditions for the assay of the intra- and extrasynaptosomal rates of deamination of dopamine and noradrenaline by the two forms of monoamine oxidase (MAO) have been determined in striatal and frontal cortical homogenates, respectively, from C57 BL/6 mice. The activities obtained were compared with the corresponding activities found for NMRI mice, a strain less sensitive to the neurotoxic effects of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) than the C57 BL/6 strain. In both strains, the intra- and extrasynaptosomal deamination of dopamine in the striatal homogenates was brought about predominantly by MAO-A. No significant differences between the two strains were found for the intra- or extrasynaptosomal MAO-A or -B activities towards dopamine in striatal homogenates. On the other hand, the striatal dopamine concentrations were higher in the C57 BL/6 mice than in the NMRI mice. The concentrations of the dopamine metabolites DOPAC and HVA were similarly higher, suggesting that the rate of turnover of dopamine is the same for the two strains. In frontal cortical homogenates, MAO-A predominated in the deamination of noradrenaline both intra- and extrasynaptosomally. The extrasynaptosomal rates of deamination of noradrenaline were similar in the two mice strains, whereas the intrasynaptosomal MAO-A activity was significantly higher for the C57 BL/6 mice. These results concur with and extend to the noradrenergic system the conclusion previously made by Jossan et al. (1987) for the dopaminergic system that although MAO-B activity is necessary for expression of MPTP neurotoxicity, it is not the rate-limiting step for the development of the neurotoxic effects.

Proximal closing wedge osteotomy and adductor tenotomy for treatment of hallux valgus.

After 2 to 4 years, 25 patients (27 feet) who had a proximal closing wedge osteotomy of the first metatarsal and an adductor tenotomy were reviewed. A total of 20 patients (22 of 27 feet) were completely satisfied; 5 patients not completely satisfied had metatarsalgia because of dorsal displacement of the first metatarsal head. Radiographic measurements showed a narrowing of the forefoot rather than a large change in the intermetatarsal angle. The recovery period was long, an average of 11 weeks. Pin inflammation occurred in 5 patients and incisional neuromas in 2 patients. The risk of these complications must be taken into consideration when using this operation.

Prognostic value of systolic ankle and toe blood pressure levels in outcome of diabetic foot ulcer.

The prognostic value of distal blood pressure measurements has been studied in 314 consecutive diabetic patients with foot ulcers. Systolic toe blood pressure was measured with a strain-gauge technique, and ankle pressure was measured with strain-gauge or Doppler techniques. Wound healing was defined as intact skin for at least 6 mo. One hundred ninety-seven patients healed primarily, 77 had amputations, and 40 died before healing had occurred. In 294 of 300 patients, it was possible to measure either ankle or toe pressure. Fourteen patients were not available for pressure measurements. Of these, 10 patients healed primarily, and 4 died before healing occurred. Both ankle and toe pressures were higher (P less than .001) among patients who healed without amputation compared with those who underwent amputation or died before healing. No differences were seen in ankle or toe pressure levels among those who had amputations or died. No patient healed primarily with an ankle pressure less than 40 mmHg. An upper limit above which amputation was not required could not be defined. Primary healing was achieved in 139 of 164 patients (85%) with a toe pressure level greater than 45 mmHg, whereas 43 of 117 patients (36%; P less than .001) healed without amputation when toe pressure was less than or equal to 45 mmHg. In conclusion, a combination of ankle and toe pressure measurements is a useful tool to predict primary healing in diabetic foot ulcers.

Fluorescein angiography for predicting healing of foot ulcers.

Fluorescein angiography (FA) was performed on 83 patients (68 diabetics) with foot ulcer or gangrene. Densitometric measurements were made on the FA images, and different FA parameters were defined. These parameters, as well as systolic ankle and toe blood pressures, were evaluated for predicting the future course, i.e., whether healing would occur or whether major amputation below or above the knee had to be performed. The toe slope (i.e., the rate of increase of fluorescence on the big toe during the first 10 seconds after its appearance on the toe) predicted healing correctly in 0.83 and major amputation in 0.88. The ankle and toe pressures had only slightly lower predictive value. The combination of ankle pressure and toe slope predicted healing correctly in 0.91 and major amputation in 0.88. When ankle pressure cannot be measured, FA is the method of choice. Further, FA provides information on regional blood flow unobtainable by any other method.

Monoamine oxidase (MAO) in pancreatic islets of the mouse: some characteristics and the effect of chemical sympathectomy.

We have previously observed that chemical sympathectomy in the guinea pig induced an increase in the intensity of serotonin fluorescence in the insulin cells of the pancreatic islets. In the present study we have studied some basal characteristics of mouse islet monoamine oxidase (MAO) and the effect of chemical sympathectomy induced by 6-hydroxydopamine on the activity of this enzyme. Estimated Km-values for MAO activity with different substrates were as follows: serotonin (5-HT; 2 x 10(-4) M), dopamine (DA; 4 x 10(-4) M), and 2-phenylethylamine (PEA; 4 x 10(-6) M). The highest specific activity was displayed with PEA and DA as substrates, whereas it was about 10 times lower with 5-HT as substrate. Two days after the destruction of islet adrenergic nerves by 6-hydroxydopamine we observed a decrease of approximately 30% of the MAO activity with serotonin as substrate and after seven days the MAO activity with PEA as substrate had decreased by 18%. By contrast the MAO activity with DA as substrate was slightly increased (+ 12%) at two days after sympathectomy. There is a need for a further characterization of islet MAO activity(ies) and for further studies on its putative role in insulin secretory processes through its regulation of the islet monoamine stores.

Influence of age on effects induced by intermittent ethanol treatment on the ethanol drinking pattern and related neurochemical changes in the rat.

Male rats were treated with one ethanol (2.0 g/kg i.p.) or saline injections once a week for 50 weeks. During this treatment period the rats had in addition access to ethanol (10% in drinking fluid) as a choice against water for 24 h prior to the injection. During the following evaluation period, animals had a continuous choice between ethanol and water and the concentration of the ethanol solution increased every 3rd week from 5 to 10, 15 and 25%, with 10% as a reference tested between the other concentrations. The animals were killed after an abstinence of 4 weeks, whereupon the concentrations of noradrenaline (NA), dopamine (DA), serotonin (5-HT) and 5-hydroxyindole acetic acid (5-HIAA) were determined in the frontal cortex. In the remaining cerebral cortex, activity of monoamine oxidase, reuptake of NA and stimulated inositol phospholipid (PI) breakdown was also determined. Muscarinic binding sites were determined in the striatum. During treatment, saline injected rats had a constant voluntary 24 h ethanol intake. There was a decrease in the corresponding intake in the animals given the ethanol injections. The diminishing of the intake was more marked in rats starting treatment at an age of 19.4 weeks when compared to rats starting at an age of 5.4 weeks. In the evaluation period the ethanol intake was fairly constant for all groups. However, the regressions between intake of the reference concentration when plotted against the different tested concentrations were most marked in the group where ethanol injections started at an early age. In the total material there were significant F-values when concentrations of NA, 5-HIAA, 5-HT/5-HIAA in the cortex and muscarinic binding sites in the striatum were tested. Age could not be excluded as a contributing factor, but for muscarinic binding sites in the striatum, concentrations of DA and 5-HIAA in the cortex, and potassium stimulated PI breakdown in the cortex significant regressions with voluntary ethanol intake as dependent variable could be established. Since these intakes are stable, a causal relation with dependence may be involved.

The PCA unicompartmental knee. A 1-4-year comparison of fixation with or without cement.

Totally, 93 knees were operated on with the PCA unicompartmental knee for gonarthrosis Stages I-III according to Ahlbäck. The clinical findings for 84 arthroplasties with 1-4 years' follow-up showed 82 excellent or good results. The mean postoperative flexion was 124 degrees. The position of the tibial component was respectively 86 degrees and 89 degrees in the AP and lateral views. The mechanical axis was slightly undercorrected. There were two failures, both with persistent pain on walking. One failure was among 43 uncemented knees, whereas the other was among the 41 cemented arthroplasties. Four uncemented arthroplasties had a first steps' problem but were satisfactory otherwise. There was no infection or clinical loosening. A reoperation was performed in 1 case, in which a symptomatic osteophyte was removed. When compared with our earlier Marmor series, the PCA unicompartmental arthroplasties were better. The cemented arthroplasties had a somewhat higher frequency of complete pain relief. An uncemented arthroplasty should only be considered in special situations.

Impaired performance of rats in the Morris swim-maize test late in abstinence following long-term sodium barbital treatment.

Rats were tested for place learning in the Morris swim maze on days 110-114 of abstinence following 48 weeks of treatment with sodium barbital. A retarded acquisition of the swim-maze task, that could not be ascribed to motor impairments, was found in the barbital-treated rats. There was a significant difference in brain weight, but there were no significant differences between the control and barbital-treated rats in the frontal cortical concentrations of noradrenaline (NA), dopamine (DA), 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA), nor in the intra- and extrasynaptosomal activities of cerebral cortical monoamine oxidase towards NA and 5-HT. Postsynaptically, neither the cerebral cortical inositol phospholipid breakdown responses to carbachol and NA (mediated by muscarinic and alpha 1-adrenergic receptors, respectively), nor the striatal and cortical densities of muscarinic receptors labelled by [3H]quinuclidinyl benzilate [( 3H]QNB) were found significantly to be altered in the barbital-treated rats. A strong correlation between the density of striatal and cortical [3H]QNB binding sites was seen for the barbital-treated (r = 0.91) but not for the control (r = -0.05) rats. It is suggested that the deficit in performance of the barbital-treated rats in the Morris maze may be related to a cholinergic dysfunction.

Intra- and extra-dopamine-synaptosomal localization of monoamine oxidase in striatal homogenates from four species.

MAO-A and MAO-B activities within and outside dopaminergic synaptosomes in homogenates of striatal tissue from pig, cat, rat and human brains have been studied by using a specific "low substrate concentration technique" with dopamine. It was found that within the synaptosomes, MAO-A activity predominated in all species. Outside the synaptosomes there were more pronounced differences and only in the rat did MAO-A predominate, while in the other species MAO-B predominated. When estimating MAO-A and -B activities with a conventional method the activity of MAO-B predominated in man, cat and pig. Thus, also in species where the MAO-B activity (as estimated in a conventional way) was dominating, the intrasynaptosomal deamination of dopamine was brought about mainly by MAO-A. The "low substrate concentration technique", more adequately reflects physiological conditions by taking into account the higher concentration of monoamine transmitter substrates within the monoamine neurons. With this technique it was found that in all species (with the possible exception of man) the oxidation rate was higher within than that outside the DA-synaptosomes. In man the unavoidable longer time between death and estimation of the enzyme activity may be the cause of the deviating result.

Glycogen and glycogen-hydrolysing lysosomal enzyme activity in mouse liver: effects of fasting, adrenoceptor antagonism and insulin-induced hypoglycaemia.

Whereas the phosphorolytic breakdown of liver glycogen is known to be of great physiological importance, the functional role of the hydrolytic glycogenolysis in the lysosomal system is less well understood. In the present study the effects of fasting, alpha- and beta-adrenoceptor antagonism and insulin-induced hypoglycaemia on liver lysosomal glycogen-hydrolysing enzyme activity were investigated in mice. In freely fed mice the glycogen-hydrolysing activity (acid amyloglucosidase) was only 50% of the maltose-hydrolysing activity (acid maltase). Starvation for 24 h reduced the acid amyloglucosidase activity by approximately 30% (P less than 0.001), whereas the activities of acid maltase, acid phosphatase and beta-glucuronidase appeared unaffected. N-acetyl-beta-D-glucosaminidase activity was moderately (20%; P less than 0.01) enhanced by fasting. Thus, liver lysosomal enzyme activities may change independently of each other during fasting. Further, during short-term hypoglycaemic conditions (45 min) induced by endogenous or exogenous insulin, the activity of liver acid amyloglucosidase was found to be moderately reduced (15-20%). Blockade of alpha- and beta-adrenoceptors by phentolamine and propranolol did not result in any apparent influence on acid amyloglucosidase activity except for the indirect effect exerted by the phentolamine-induced hypoglycaemia. A moderate negative correlation (r = -0.51; P less than 0.001) between total liver glycogen concentration and acid amyloglucosidase activity was observed in a series of 43 freely fed NMRI mice. Our data show that in mouse liver the acid maltase activity predominates over the acid amyloglucosidase activity.(ABSTRACT TRUNCATED AT 250 WORDS)

The uptake of serotonin and dopamine by homogenates of frozen rat and human brain tissue.

A recently described procedure of freezing and thawing, which allows retention of metabolic and functional integrity, has been applied in the study of serotonin and dopamine uptake into frozen rat and post mortem human frozen tissue. The Km and Vmax for the serotonin uptake into human hypothalamus were estimated to be 0.12 microM and 0.03 nmol/g/min respectively. The Km and Vmax for the dopamine uptake into human putamen were estimated to be 0.28 microM and 0.13 nmol/g/min respectively. The results indicate that the freezing procedure does not affect the uptake sites for these transmitters. The storage time before freezing is however of importance for the Vmax value. The Km value for the uptake, on the other hand, seems to be rather resistant to storage time before freezing.

Intra- and extraneuronal monoamineoxidase-A and -B activities after central axotomy (hemisection) on rats.

Hemitransection of the left side of rat brain results in a selective increase (40%) in the activity of MAO-B in the left side striatum, as compared to the right, unoperated side. This increase is shown to be the result of an increase in the activity of extraneuronal MAO-B using a "low substrate concentration method" with dopamine as substrate. This result is compatible with the hypothesis, that in certain degenerative processes such as aging, Alzheimer's disease, Huntington's disease and axotomy there is a stimulated growth of extraneuronal cells, which are relatively rich in MAO-B activity.

The effect of deprenyl (selegiline) on intra- and extraneuronal dopamine oxidation.

It was found that in the rat striatum DA was oxidized extrasynaptosomally to 11% by MAO-A and to 3% by MAO-B. The corresponding intrasynaptosomal oxidations were 84% and 2%, respectively. Those figures were virtually unchanged even if the rat brain MAO-B was selectively inhibited to 87% by deprenyl. In the human brain extrasynaptosomal oxidation was 16% and 66%, respectively, by MAO-A and -B. Intrasynaptosomally the corresponding figures were 12% and 6%, respectively. Selective inhibition of human caudate MAO-B was calculated to give a total reduction of DA oxidation of 63%. The differences between man and rat are due to the proportionately greater oxidation of DA by MAO-B in man, which is a consequence of a higher ratio of concentration of MAO-A/-B in the rat.

Effect of ovariectomy on longitudinal bone growth in the rat.

The effect of ovariectomy on longitudinal bone growth was studied in growing rats. The operation was performed at ages 20, 40, and 60 days. Sham operations were made at age 40 days. At different postoperative intervals, the growth rate was determined with the tetracycline technique, and the width of the growth plate was registered. After a slight initial retardation, the growth rate after ovariectomy was significantly greater than in normal rats during a 40-60 day period. During the same period, the growth plate was wider. The increase in growth rate was greater and more rapid if ovariectomy was performed in older animals. The results indicate that ovariectomy increases longitudinal bone growth and that longitudinal bone growth and skeletal maturation depend less on ovarial function in young animals than that in more mature animals.

The Lund drill guide. An instrument for repair or reconstruction of the cruciate ligaments.

A new drill for cruciate ligament repair is described. It is a modification of a French drill guide (Landanger). The guide allows for the possibilities of free adjustment in all planes and good stability during the drilling procedure. The instrument has been used in 22 patients with anterior cruciate ligament injuries and five patients with injuries to the posterior cruciate ligament. With this instrument it is sufficient with only an anterior approach for a posterior cruciate ligament repair. It is concluded that the Lund cruciate drill guide is simple and manoeuvrable for surgery of the ligaments of the knee joint. In all cases correct drilling was possible and in no instance the instrument has failed.
