Biological and mechanical behavior of autologous pericardium used for tricuspid valvular replacement in dogs.

The fate of autologous pericardium when used to replace a tricuspid valve leaflet has been investigated in dogs. Changes have been studied by means of macroscopic, mechanical, histological, scanning electron microscopy, and in vitro culture techniques. In each of nine experiments the valve failed and death occurred within the first 15 post-operative days. Retraction, thickening of the cusp, fibrinous deposition and/or tissue rupture were associated with a decrease of resistance to traction tests, decrease of elasticity and increase of viscosity of the implanted tissue. The early behavior of autologous pericardium was characterized by precocious inflammatory reaction on the valve leaflets, focal disruption of the collagenous structure of the leaflets, absence of fibroblastic cell infiltration as confirmed by in vitro culture of implanted pericardium and the lack of endothelial-like cell surface at the 10th-15th postoperative day.

[Oral contraceptive and coronary thrombosis. Two clinicopathological cases].

The cases reported are interesting from two points of view: they confirm the thrombotic mechanism of the arteria lesions (coronary amongst others) and the oral contraception pill was the only cardiovascular risk factor, smoking, hyperlipidaemia, hypertension and coronary atherosclerosis having been eliminated. The two young women (31 and 34 years old) presented with severe myocardial infarction and shock, leading to death within hours in one case and within weeks in the other (despite an attempt to resect the infarct and prolonged intraaortic balloon pumping). In both cases multiple thromboses were discovered: coronary, intra cardiac and in the collateral aortic circulation.

Distribution of adrenaline-synthesizing enzyme activity in the human brain.

A study of the distribution of phenylethanolamine-N-methyl-transferase (PNMT) activity in normal human brain is presented. After a preliminary dissection to separate brain tissue for formalin fixation and tissue designed for biochemical studies, the hemi-brain stem is cut in slices by hand and a cerebral hemisphere is cut on a cyromicrotome. "Punches" are made with an operating microscope. The dissection method was used to study the distribution of PNMT activity in 117 "punches" made on 21 slices obtained from 5 normal human brains. The caudo-rostral distribution of PNMT activity in C1 and C2 groups was found to be identical in each brain. The distribution of PNMT activity was found to be similar to that in the rat, but, in addition, important activity was found in the substantia nigra, internal pallidium and nucleus accumbens.

[Variation of the inflammatory response of an autologus implant function and viability: mixed and unilateral inflammatory response].

The behaviour of an autologous grafted implant depends upon its viability. In our study, pericardial implants were used as cardiac valves. Results showed that when using fresh live pericardium, an inflammatory response developed destroying it within 8 days. When implants cells were killed without alteration of their antigenicity--in order to prevent rejection--cellular growth and neovascularisation from implantation site occurred leading to a secondary live graft. Controlled killing of the implant seems to be an essential step in order to get live grafts.

[Giant cell myocarditis. Attempt at interpretation, apropos of 3 cases].

Three cases of giant-cell myocarditis were observed, and 63 comparable cases were published in the literature. On the basis of this material, the significance of this disease was studied, which was considered for long to be of "granulomatous" origin, while it seems to be a peculiar histopathologic type of myocardialgeneration. The cause for this necrosis remains unknown; in spite of as complete as possible investigations in one of the three cases (in which death was preceded for labile recurring lung infiltrates during eleven months) no cause could be demonstrated.
