[Decomposition of aqueous sodium pentachlorophenolate (PCP-Na) by using TiO2 coating photocatalyst].

Titanium dioxide (TiO2) photocatalysts were coated on hollow glass beads by sodium silicate (Na2SiO3.9H2O) solution. The coated catalysts were characterized by BET, XRD. Their photoactivity were examined using photocatalytic degradation of sodium pentachlorophenolate(PCP-Na) and glucose solution. The impact of catalysts dosage, PCP-Na initial concentration, initial pH and oxygen concentration on reaction kinetics were also studied in detail. The experiments illustrated that the performance of C3-type catalysts was satisfactory. Conditions of the experiments were following: The initial concentration of PCP-Na and CODCr were 10 mg.L-1 and 400 mg.L-1, respectively, and illumination time was 2 hours and catalysts dosage was 2 g.L-1, the removal rates of CODCr and PCP-Na were above 65% and 92% respectively. Optimum dosage of C3-type catalysts was 2-2.5 g.L-1, DO was no less than 3 mg.L-1.

Mechanisms of mechanical heart valve cavitation: investigation using a tilting disk valve model.

BACKGROUND AND AIM OF THE STUDY: The induction of mechanical heart valve (MHV) cavitation was investigated using a 27 mm Medtronic Hall (MH27) tilting disk valve. METHODS: The MH27 valve was mounted in the mitral position of a simulating pulse flow system, and stroboscopic lighting used to visualize cavitation bubbles on the occluder inflow surface at the instant of valve closure. MHV cavitation was monitored using a digital camera with 0.04 mm/pixel resolution sufficient to render the tiny bubbles clearly visible on the computer monitor screen. RESULTS: Cavitation on MH27 valve was classified as five types according to the time, site and shape of the cavitation bubbles. Valve cavitation occurred at the instant of occluder impact with the valve seat at closing. The impact motion was subdivided into three temporal phases: (i) squeezing flow; (ii) elastic collision; and (iii) leaflet rebound. MHV cavitation caused by vortices was found to be initiated by the squeezing jet and/or by the transvalvular leakage jets. By using a tension wave which swept across the occluder surface immediately upon elastic impact, nuclei in the vortex core were expanded to form cavitation bubbles. CONCLUSION: Analysis of the shape and location of the cavitation bubbles permitted a better understanding of MHV cavitation mechanisms, based on the fluid dynamics of jet vortex and tension wave propagations.

[Composting process of municipal solid waste with high effective complex microbial community].

The effects of high effective complex microbial community (MECMC) in a combination composting process of municipal solid waste (MSW) and sludge were examined through inspecting biomass, temperature, organic matter, the carbon, nitrogen ration (C/N). Composting was performed under such conditions: The ratio of MSW:sewage:sawdust:composting of 45:25:15:15 (W/W) and organic matter of 60%, total N of 1.4%, total P of 0.69%, total K of 1.25%, original moisture of 58.5%, original ratio of C/N of 30, aration flow of 0.8 L/min.kg(volatile organic matter). The rates of adding HECMC to treatment 1, treatment 2 and treatment 3 were 2%, 3%, 5% respectively. Control experiment added 3% inactive HECMC. The day of MSW and sludge mature in control experiment, treatment 1, treatment 2 and treatment 3 were 30, 24, 18 and 12, respectivly. This indicated HECMC were effective to compose organic matter and speedup composting change into humus. There are many active bacteria in composting which are favorable for soil.

[Comparison of serological and DNA typings for HLA-AB of cord blood samples].

OBJECTIVE: To evaluate the practical value of serological method for HLA-AB typing of cord blood samples. METHODS: Serological typing results of 1800 cord blood samples were analyzed and 36 of them were compared with that of PCR-SSP (polymerase chain reaction-sequence specific primers). Lymphocytes in cord blood samples were isolated by T-immunomegnetic beads or Ficoll. RESULTS: T lymphocytes isolated by fluorobeads have better activity than that in mononuclear cells isolated by Ficoll. In order to get the more active cord blood cells, samples stored in vitro over 24 hours must be isolated with fluorobeads. Its serological reaction is lower as compared with that of peripheral mononuclear cells. Except 28 samples which had too many dead cells or few cells, there were 310 (17.5%) unconfirmed antigens among 1772 cases by serological typing. The main causes were low specificity of typing reagents, cross reaction, weak serological reaction, high background of dead cells and extra reaction. Confirmed by PCR-SSP, the serological typing had a wrong result rate of 13.88%. CONCLUSION: Although serological typing is rapid and easy, it is necessary to use genetic typing as a complement to it.

[Interference of human processed pseudogene and corresponding counter-measures in reverse transcriptase-polymerase chain reaction].

OBJECTIVE: This study aimed to set up an effective method to find the interference of processed pseudogene and overcome it in RT-PCR. METHODS: Using RT-PCR methods, the authors amplified the cytokeratin 19 cDNA or DNA sequence in human lung squamous cell carcinoma samples. RESULTS: They could find the influence of pseudogene through performing genomic DNA and nonreversed transcribed RNA controls. By digesting the extracted RNA sample with RNase-free DNase or regulating the annealing temperature based on mismatched primers, they could effectively eliminate the interference. CONCLUSION: The presence of processed pseudogene usually interferes in the experimental protocol in RT-PCR. This problem can be resolved with the method aforementioned.

Circadian and septadian variation in the occurrence of acute myocardial infarction in a Chinese population.

To investigate whether circadian or any other temporal pattern(s) exist in the occurrence of acute myocardial infarction (AMI) in a Chinese population, we analyzed 428 patients with confirmed AMI for temporal patterns of AMI occurrence. The patients admitted to the Affiliated Hospital of Shandong Medical University during 1991-95 were from Jinan, the capital city of Shandong Province of China, which has a population of 2.5 million. The chi-square test for goodness of fit was used to test the difference among the frequencies of AMI occurrence in 4 equal intervals (01.00-07.00 h, 07.00-13.00 h, 13.00-19.00 h, 19.00-01.00 h) during the day and among those on 7 days during the week. The results showed that AMI occurrence exhibited significant circadian (p<0.001) and septadian (day of the week) (p=0.046) periodicity, with a peak at 01.00-07.00 h and a trough at 13.00-19.00 h during the day, and a peak on Saturday and a trough on Wednesday during the week. The peak to trough ratio of risk was 2.7 during the day and 2.1 during the week. It is concluded that there were circadian and septadian biorhythms in AMI occurrence in the Chinese population and that these were different from those observed in Western populations. Further investigation of the underlying mechanisms may shed further light on the trigger mechanisms of AMI and thus be helpful in the prevention and treatment of AMI.

[Simulation study on the blood volume feedback control].

To describe the relationship between blood volume and the cardiac function mathematically, a computer cardiovascular model, was developed including a systemic circulation, a heart and a pulmonary model and a blood volume feedback control. Simulation results indicate that the chronic heart failure could be compensated by increasing the blood volume to maintain the proper arterial pressure. Deterioration in heart failure causes an increase in blood volume, as well as increases in heart volume and cardiac energy consumption. These results are in agreement with the clinical data and results reported in the literature. The simulation also shows that for a predetermined ventricular contractility, an adequate increase in blood volume will increase the heart volume but decrease the cardiac energy consumption and lead the heart to work in an optimum condition.

Simple and sensitive method for the quantitative analysis of lometrexol in plasma using high-performance liquid chromatography with electrochemical detection.

Previously described methods for the determination of lometrexol in plasma used either fluorescence or ultraviolet detection. An alternative method for the determination of lometrexol utilizing electrochemical detection is described, having comparable sensitivity to fluorometric methods but not requiring pre-analytical oxidation. Following sample clean-up, separation is achieved on a phenyl column with a mobile-phase of 8% acetonitrile in 50 mM sodium acetate buffer, pH 4.0. The calibration curve in plasma is linear from 10 to 200 ng/ml with inter- and intra-day precision of 5.4 and 5.5%, respectively. The recovery of lometrexol from plasma is 81 +/- 1.5%, and the lower limit of detection is 5 ng/ml, using a signal-to-noise ratio of 3.

Prevention of tissue calcification on bioprosthetic heart valve by using epoxy compounds: a study of calcification tests in vitro and in vivo.

Calcification is the principal cause of the clinical failures of the bioprosthetic heart valves fabricated from glutaraldehyde pretreated porcine aortic valves or bovine pericardium. In this paper, we compared the calcification on various types of bovine pericardiums pretreated with two hydrophilic epoxy compounds adding GA post-treatment (EP 1 and EP 2), glutaraldehyde (GA)- and nontreated pericardium (Fresh), respectively, by in vitro and in vivo tests. Significant decrease of calcification was found by pretreatment with both epoxy compounds rather than with glutaraldehyde: 0.250 +/- 0.001 (Fresh), 0.276 +/- 0.058 (EP 1), 0.302 +/- 0.071 (EP 2), and 0.478 +/- 0.172 (GA) micrograms (Ca)/mg (dried tissue), respectively, after 20 days dipping in a simulating serum solution in vitro; 115.13 +/- 60.11 (Fresh), 129.84 +/- 51.08 (EP 1), 167.39 +/- 20.81 (EP 2), and 205.19 +/- 16.86 (GA) micrograms/mg, respectively, after 3 months subcutaneous implantation in rabbits. The in vitro method for evaluating calcification designed by us gave the similar order among four samples with that obtained by in vivo test. Because the bovine pericardium pretreated with the epoxy compounds adding GA post-treatment possesses the greater tenacity than that pretreated only with epoxy compounds or GA, meanwhile the calcification is also significantly decreased with this pretreatment, it may be expected that the bovine pericardium with this pretreatment will have the greater anticalcification and durability in dynamic stress.

Effect of pretreatment with epoxy compounds on the mechanical properties of bovine pericardial bioprosthetic materials.

Early failures of bovine pericardial heart valves are due to leaflet perforation, tearing and calcification. Since glutaraldehyde fixation has been shown to produce marked changes in leaflet mechanics and has been linked to development of calcification, bovine pericardium fixed with the four hydrophilic epoxy formulations and their mechanical properties are studied in this paper. We measured the thicknesses, shrinkage temperatures, stress relaxations and stress-strain curves of bovine pericardiums after different treatments with (1) non-treatment (fresh), (2) glutaraldehyde (GA), (3) epoxy compounds followed by the posttreatment with GA (EP 1#, EP 2#), and (4) epoxy compounds (EP 3# and EP 4#). Results of this study showed that the hydrophilic epoxy compounds are good crosslinking agents. There are no significant differences of shrinkage temperature and ultimate tensile stress among all tissue samples pretreated with GA, EP 1# and EP 2#. However, the stress relaxations of tissue-samples pretreated with epoxy compounds followed by the posttreatment with GA (EP 1# and EP 2#) are significantly slower than that pretreated with GA, and the strains at fracture of EP 1# and EP 2# are also significantly larger than that of GA or epoxy compounds. These facts show that the bovine pericardium pretreated with the epoxy compound followed by the posttreatment with GA (EP 1# and EP 2#) possesses greater tenacity and potential durability in dynamic stress.
