[Clinical and pathologic analysis of orbital pseudotumor].

OBJECTIVE: To investigate the relationship between the histopathology and the imaging analysis of orbital pseudotumor to judge the histopathological types of these tumors before treatment according to imaging analysis, in order to search for different therapeutic approaches for different pathologic types and to predicate prognosis. METHODS: Fifty-six cases (58 eyes) of orbital pseudotumor that had been operated in Tongren hospital were studied. The clinical manifestations of these patients were reviewed. Changes in the imaging analysis and histopathologic examination were observed and their relationship was studied. Methods of treatment and their prognosis were also analyzed. RESULTS: Historically, the lymphocytic infiltrated type of orbital pseudotumor composed of large amounts of chronic inflammatory cells, only a small amount of collagen was found, so the imaging features of the lymphocytic infiltrated type presented as low or none reflecting areas with a clear posterior demarcation on B scan, and the signals were low or equal on T(1)WI and high on T(2)WI, which could be enhanced by contrast. Oppositely, the fibrotic type was composed of large amounts of fibrotic tissue, and few chronic inflammatory cells were found, so that only the anterior border of the lesion could be identified and the posterior demarcation was blur on the B scan, and the signals were low or equal on T(1)WI and low on T(2)WI, which could not be enhanced or could be enhanced only slightly by changing the contrast. The mixed type presented as an admixture of chronic inflammatory cells and fibric hyperplasia, and the imaging of the mixed type was midway between the lymphocytic infiltrated type and the fibrotic type. Forty-two patients of lymphocytic and mixed types which were identified by histopathology and imaging analysis received a local and systemic corticosteroid therapy. The effect was unstable, and recurrence occurred easily. Therefore, operations were performed on these patients, thirteen of them were treated with low-dose radiotherapy (2 000 - 3 000 cGy) two weeks after operation. Fifteen cases recurred after the operation and 4 cases recurred after the radiotherapy. The fibrotic type of orbital pseudotumor was identified in 12 cases in which no effect could be observed after conservative therapy and operations were performed subsequently. Two cases recurred and were operated again. Exenteration was performed in 2 cases of 56 cases. The cure rate of the composite treatment in the present series was 68.5%. CONCLUSIONS: Different types of orbital pseudotumor present different imaging exhibition, and imaging features are closely related with histopathological changes. Lesions can be localized by CT scan, and also can be localized by B scan and MRI. While it is difficult to obtain a cure result in the orbital pseudotumor, the cure rate can be improved by composite treatment.

[Transformation of sulfur forms during coal pyrolysis and partial gasification in a fixed bed reactor].

The development of various process to the pre-desulfurization of coal was drawn more attention. In present study, the transformation of sulfur forms of three different ranks high sulfur coals during coal pyrolysis and partial gasification were investigated in a fixed bed reactor. The sulfur and carbon content analysis of original coal and coal char produced were determined by LECO SC-444 and wet chemical analysis according to Sugawara's method. The results showed that half of inorganic sulfur and partial of organic sulfur were removed during coal pyrolysis. And the sulfur removal was much more than carbon during pyrolysis process; and the sulfur in the coal char, especially the sulfide sulfur was removed completely during partial gasification process for both Datong coal and Xishan coal, the degree of sulfide sulfur removal could be increased with increasing temperature. At the same time, the results of Yima coal showed that the effect of fixed-sulfur by alkaline metals increased when the temperature was higher than 700 degrees C, which attribute to the increase of the fixed sulfur reaction rate and the decrease of mass-transfer limitation.

[Gene prediction and function research of SARS-CoV(BJ01)].

Through reading the articles, this study points out the shortage of gene prediction and function research about SARS-CoV, and predict it again for developing effective drugs and future vaccines. Using twelve gene prediction methods to predict coronavirus known genes, we select four better methods including Heuristic models, Gene Identification, ZCURVE_CoV and ORF FINDER to predict SARS-CoV(BJ01), and use ATGpr for analyzing probability of initiation codon and Kozak rule, search transcription regulating sequence(TRS) in order to improve the accuracy of predicted genes. Twenty-one probable new genes with more than 50 amino acids have been obtained excluding 13 ORFs which are similar to the genes of NCBI and relative articles. For predicted proteins, we use ProtParam to analyse physical and chemical features; SignalP to analyse signal peptide; BLAST, FASTA to search similar sequences; TMPred, TMHMM, PFAM and HMMTOP to analyse domain and motif in order to improve reliability of gene function prediction. At the same time, we separate the 21 ORFs into four classes using codition of four gene prediction methods, match score, match expection and match length between predicted gene and Coronavirus known gene. In the end, we discuss the results and analyse the reasons.

[Identification on an antagonistic rhizobacterium X3 from rhizosphere of cucumber].

Classical physiological and biochemical methods, Biolog, and 16S rDNA analysis were used to identify the strain X3 which could control cucumber seedling damping-off. Physiological and biochemical identification showed that it belonged to Pseudomanas aeruginosa, but Biolog identification showed it was P. spinosa. 16S rDNA determination and analysis was used for further identification, which showed that the 16S rDNA sequence of X3 shared 93.7% homologies with published sequence of P. aeruginosa from GenBank, and both sequences constituted a branch in Phylogenetic tree. Based on these results, it is considered that the strain X3 belongs to one strain of P. aeruginosa.

[Inhibition of intracellular Ca(2+)-chelating agent on Hela cell apoptosis induced by HSV-1].

AIM: To explore the variation of intracellular free-Ca (2+) concentration during Hela cell apoptosis induced by HSV-1 and inhibition of Ca(2+)-chelating agent on the apoptosis. METHODS: Apoptosis of the Hela cells infected by HSV-1 was observed under scanning electron microscope (SEM) and transmission electron microscope (TEM), and the variation of intracellular free-Ca (2+) concentration, labeled with fluorescent probe was observed under fluorescence-microscope at a series of timepoints. RESULTS: The Hela cells infected by HSV-1 showed typical apoptotic morphological changes. Free-Ca (2+) concentration increased and reached peak at the 12 hours after HSV-1 infection. Characteristic morphological features of apoptosis occurred at the 24 hours after infection. This apoptosis could be inhibited by intracellular Ca(2+)-chelating agent. CONCLUSION: Intracellular free-Ca(2+) might play an important role in Hela cell apoptosis induced by HSV-1, which provides useful clues for clinical treatment of associated diseases.

[Review on genetic diversity in Pinus].

The ways of probing genetic diversity of pines involve many aspects, such as morphology, chromosome, isozyme, DNA, etc. The phenotypic characteristics in pines vary widely and the differences of inheritability(h2) are obvious among characteristics and among species. Up to now, isozyme is still the most common means to measure genetic diversity of pines. Generally, there are high allozyme diversity within populations and low differentiation coefficient among populations, but differences exist between species in Pinus. The variations of chromosome among pines are very low and the karyotypes of pines are consentaneous, but the genomes of pines in cell nucleus are much larger than that of broadleaves. Diversity of pines are abundant at nucleus DNA level but are poor at plastid DNA level, such as ctDNA. There are many factors that will affect genetic diversity of pines, in which mating system and environment are two main factors. Finally,we reviewed the research on origin of Pinus and conservation strategy of genetic diversity, etc.

Generation of a consensus sequence from prevalent and incident HIV-1 infections in West Africa to guide AIDS vaccine development.

We considered several key issues regarding the development of a DNA-based human immunodeficiency virus type 1 (HIV-1) vaccine: (1) should the candidate vaccine construct be derived from incident or prevalent HIV-1 strains; and (2) should circulating plasma virus, archived HIV-1 provirus recovered from peripheral blood mononuclear cells, or both be included? To address these questions, we collected circulating HIV-1 strains from infected individuals residing in Abidjan, Côte d'Ivoire. From a panel of 23 strains, 22 were HIV-1 subtype A in gag, 19 of which phylogenetically clustered with the recombinant HIV-1, CRF02-AG strains from West Africa. The mosaic genome of CRF02-AG was confirmed by sequencing the protease gene. A consensus gag p24 protein sequence was generated and 147 of 148 codons were identical to CRF02-AG (IbNG). Regardless of the sequence origin (RNA, provirus, incident, or prevalent), the gag p24 consensus sequences were highly representative of these distinct virologic compartments. These data suggest that the consensus sequence generated from incident and prevalent infections may provide an appropriate sequence for a DNA vaccine and is largely representative of the major circulating viral strain.

KNa(3)In(9): a Zintl network phase built of layered indium icosahedra and zigzag chains. Synthesis, structure, bonding, and properties.

This phase was discovered following direct fusion of the elements in welded Nb tubes at 550 degrees C and equilibration at 300 degrees C for 1 week. Single-crystal X-ray diffraction analysis reveals that KNa(3)In(9) crystallizes in an orthorhombic system (Cmca, Z = 8, a = 9.960(1) A, b =16.564(2) A, c = 17.530(2) A, 23 degrees C). The structure contains a three-dimensional indium network built of layers of empty In(12) icosahedra that are each 12-bonded and interconnected by 4-bonded indium atoms that also form zigzag chains. All cations bridge between cluster faces or edges, and their mixed sizes appear critical to the stability of this particular structure, which does not occur in either binary system. Both empirical electron counting and EHTB band structure calculations on the macroanion indicate that the bonding in this structure is closed-shell, whereas resistivity and magnetic susceptibility measures show that the compound is a moderately poor metal.

Expression of hetN during heterocyst differentiation and its inhibition of hetR up-regulation in the cyanobacterium Anabaena sp. PCC 7120.

The hetN gene plays an important role in heterocyst differentiation and pattern formation. An immunoblotting study showed that the hetN gene in Anabaena sp. PCC 7120 was expressed in vegetative cells grown with combined nitrogen. After a switch to a medium without combined nitrogen, hetN expression first declined and was then followed by a rapid increase in its product, HetN, which was only present in mature heterocysts. HetN is located on both thylakoid membranes and plasma membranes as determined by immunoblotting using purified membranes. Overexpression of hetN completely prevented hetR up-regulation under nitrogen-deprivation conditions, suggesting that its role in pattern control may depend on its inhibition of hetR expression.

Differential localization of chicken FIP2 homologue, Ag-9C5, in secretory epithelial cells.

When hepatocytes polarize, a subset of cellular proteins specifically localizes to the apical cell surface forming the boundary of the bile canaliculus. We have isolated a cDNA encoding a protein recognized by a monoclonal antibody (9C5) that specifically stains the bile canaliculus. The encoded protein (Ag-9C5) is a cytoplasmic protein with three leucine zippers and a zinc finger at the C-terminus. Extensive amino acid sequence similarity indicates that Ag-9C5 is likely the chicken homologue of a human protein, FIP2, which interacts with huntingtin and Rab8. Epitope-tagged Ag-9C5 colocalizes with endogenous Ag-9C5 and other canaliculus marker antigens in transfected organ cultures. In Cos7 cells and MDCK cells Ag-9C5 forms punctate cytoplasmic structures. In intact tissues Ag-9C5 is highly concentrated at the apical surfaces of cells that secrete protein from the apical surfaces, but is found in a fine punctate cytoplasmic pattern in other polarized epithelia. Because this protein has a number of characteristics of proteins that act as scaffolds for assembly of protein complexes (e.g., the cytoplasmic domain of classical cadherins and the FERM superfamily of proteins), it appears that FIP2/Ag-9C5 may act as a scaffold for assembling a complex of proteins that are involved in targeting of some secretory vesicles to defined regions of the cell surface.

Characterization of the nuclear export signal of polypyrimidine tract-binding protein.

The polypyrimidine tract-binding protein (PTB) is a nuclear protein that regulates alternative splicing. In addition, it plays a role in the cytoplasm during infection by some viruses and functions as a positive effector of hepatitis B virus RNA export. Thus, it presumably contains a nuclear export signal (NES). Using a heterokaryon export assay in transfected cultured cells, we have shown that the N-terminal 25 amino acid residues of PTB function as an autonomous NES, with residues 11-16 being important for its activity. Unlike the heteronuclear ribonucleoprotein A1 NES, this NES is separable from the nuclear localization signal, which spans the entire N-terminal 60 residues of PTB. The PTB NES cannot be shown to bind to CAS or Crm1, cellular receptors known to export proteins from the nucleus, and it functions in the presence of leptomycin B, a specific inhibitor of Crm1-dependent export. PTB deleted of its NES, unlike wild type PTB, does not stimulate the export of hepatitis B virus RNA. Therefore, the PTB NES is a functionally important domain of this multifunctional protein that utilizes an unknown export receptor.

Stabilization of globular proteins via introduction of temperature-activated elastin-based switches.

To investigate whether swapping native turns of a globular protein with an elastin-based turn sequence (VPGVG) can increase its thermostability, we have performed molecular dynamics simulations of wild-type chymotrypsin inhibitor 2 (CI2) and variants containing elastin-based turns at 10 degrees C and 40 degrees C. Wild-type CI2 is more stable at 10 degrees C, while both of the variant forms are more stable at 40 degrees C. Detailed analyses indicate that the elastin-based turns do indeed contribute to the inverse temperature behavior of the modified proteins. Therefore, swapping a wild-type turn sequence with an elastin-based turn provides a novel way to both improve stability of target proteins at body temperature and to possibly introduce a temperature-sensitive switch.
