Molecular basis for the extensibility of elastin.

Elastin is a cross-linked protein in the extracellular matrix that provides elasticity for many tissues. Its soluble precursor (tropoelastin) has two major types of alternating domains: (1) hydrophilic cross-linked domains rich in Lys and Ala and (2) hydrophobic domains (responsible for elasticity) rich in Val, Pro, and Gly, which often occur in repeats of VPGVG or VGGVG. Since native elastin is large and insoluble, many studies have focused on elastin-based peptides in an effort to elucidate its structure-function relationship. This review focuses on the molecular basis of elastin's conformational properties and associated elasticity. From both experimental and simulation approaches, elastin can be described as a two-phase model consisting of dynamic hydrophobic domains in water. The hydrophobic domain of elastin is best described as a compact amorphous structure with distorted beta-strands, fluctuating turns, buried hydrophobic residues, and main-chain polar atoms that from hydrogen bonds with water. Water plays a critical role in determining elastin's conformational behavior, making elastin extremely dynamic in its relaxed state and providing an important source of elasticity.

Identification of differentially expressed genes in human uterine leiomyomas using differential display.

UNLABELLED: In searching of differentially expressed genes in human uterine leiomyomas, differential display was used with twelve pairs of primers to compare human uterine leiomyomas with matched myometrium. False positives were eliminated by reverse Northern analysis. Positives were confirmed by Northern blot analysis. RESULTS: Four of 69 cDNA fragments (3 up-regulated named L1, L2 and L3 and 1 down-regulated named M1 in leiomyoma) were confirmed by Northern analysis. Sequence comparison and Northern analysis proved that L1 is exactly the human ribosomal protein S19. It was present ubiquitously in 13 tissues tested but in various levels and even in different size. L1 was highly expressed in parotidean cystadenocarcinoma, pancreatic cancer and breast cancer examined. No mutations have been found in human uterine leiomyomas (n=6). CONCLUSIONS: hRPS19 overexpression might be a universal signal in rapid cell growth tissues.

Identification of up-regulated genes in human uterine leiomyoma by suppression subtractive hybridization.

In searching for differentially expressed genes in human uterine leiomyomas (ULs), suppression subtractive hybridization was used to construct an UL up-regulated library, which turned out to represent 88 genes. After two rounds of screening by reverse Northern analysis, twenty genes were proved to be up-regulated, including seventeen known genes and three genes with unknown function. All these genes were firstly associated with UL. Three genes with notable difference were selected for Northern confirmation. Our results proved the authenticity of the twenty genes. One gene named Phospholipase A2 (PLA2) showed up-regulation in 4/6 of the patients and investigation of tissue distribution indicated that it had obvious expression in prostate, testis, liver, heart and skeletal muscle.

Differential expression and localization of Mn and Fe superoxide dismutases in the heterocystous cyanobacterium Anabaena sp. strain PCC 7120.

Superoxide dismutases (Sods) play very important roles in preventing oxidative damages in aerobic organisms. The nitrogen-fixing heterocystous cyanobacterium Anabaena sp. strain PCC 7120 has two Sod-encoding genes: a sodB, encoding a soluble iron-containing Sod (FeSod), and a sodA, encoding a manganese-containing Sod (MnSod). The FeSod was purified and characterized. A recombinant FeSod was also obtained by overproduction in Escherichia coli. Immunoblot study of the FeSod during induction of heterocyst differentiation showed that the cells produced six- to eightfold more FeSod 8 h after a shift from a nitrogen-replete condition to a nitrogen-depleted condition. However, the amount of FeSod protein in filaments with mature heterocysts was the same as that in filaments grown with combined nitrogen. Superoxide anion-generating chemicals such as methyl viologen did not induce upregulation of the sodB gene expression. The predicted preprotein of the sodA gene has a leader peptide and a motif for membrane attachment at the N terminus of the mature protein. Activity staining after gel electrophoresis of the purified thylakoid membranes showed that most of the MnSod in Anabaena sp. strain PCC 7120 was located on thylakoids toward the lumenal side. Expression of the sodA gene in E. coli shows that the leader peptide was required for its activity and the membrane localization of the MnSod. Northern hybridization detected one 0.82-kb transcript of sodA. The sodA gene was upregulated by methyl viologen, whereas its amount was unchanged during heterocyst differentiation. Immunoblotting and activity staining showed that isolated heterocysts contained a lower but still significant amount of FeSod, suggesting that its function is required in heterocysts. No MnSod was observed in isolated heterocysts. These results show that the two different Sod proteins have differentiated roles in Anabaena sp. strain PCC 7120.

[Toxicity assessment of soil contaminated by heavy metals using algae growth inhibition test].

Scenedesmus obliquus growth inhibition test was carried out to assess the toxicity of soil contaminated by heavy metals. The results showed that the growth rate of Scenedesmus obliquus was positively related to the concentration of heavy metals added to soil, and it was decreased with the increasing concentration of heavy metals. Two parameters were chosen and the sensitivities were compared. It was found that the cell growth rate was more sensitive than OD growth rate. The orders of general detection limit by EC50 of different metals were Pb > Cu > Zn > Cd by using cell growth rate as the test index and Pb > Cu > Cd > Zn by using OD as the test index. This study also indicated that under the complex pollution condition, the toxicity of heavy metals in soil was stronger than that in the case of soil contaminated by single metal.

[Local resection of ciliary body tumors with vitreoretinal surgery].

OBJECTIVE: To investigate the effects, safety and indication of local resection of ciliary body tumors with vitreoretinal surgery. METHODS: Twelve patients with ciliary body tumors, including 5 males and 7 females with the age range 13 - 48 years (average 32 years) were treated with local resection. Ultrasound biomicroscopy (UBM) and ultrasonography confirmed the tumors, and the surgery was performed under hypotensive general anesthesia or local anesthesia. The range of the diameters of tumors was 5 - 20 mm and of the thickness was 4 - 12 mm. Retinal detachment was detected in 2 eyes. Local lamellar sclerocyclectomy or sclerocyclochoroidectomy with vitreoretinal surgery was performed for the resection. The surgery included lensectomy and vitrectomy using perfluorocarbon liquid, endolaser and gases or silicone oil tamponade and also scleral allograft if necessary. The resected tumors were diagnosed by pathologic examinations. RESULTS: There were no tumor recurrence and metastasis during follow-up 7 - 38 months (average 14.2 months). In the last follow-up date, the visual acuities were less than 0.05 in 1 case, 0.05 - 0.2 in 3 cases and 0.3 - 1.0 in 8 cases. The pathologic diagnoses included malignant melanoma in 5 cases, melanocytoma in 2 cases, adenoma of non-pigmented ciliary epithelium in 3 cases, neurofibroma in 1 case and glioma in 1 case. CONCLUSION: Local resection of ciliary tumor not only can conserve the eyeball, but also part of vision, and the accurate diagnosis can also be made from the resected tissues. With vitreoretinal surgery, it can avoid complications and improve the success rate. It is a safe and effective method for benign or malignant tumors of the ciliary body.

[Identification of blue grained wheat and its irradiation-mutated offsprings by genomic in situ hybridization (GISH)].

Transferring useful chromosome(s), chromosomal fragment(s) or genes from related species into common wheat (Triticum aestivum L.) is an effective method for improving wheat cultivars, and DNA:DNA in situ hybridization has been proven to be an effective method for directly demonstrating alien chromosome number and distribution in plant. In this study, genomic in situ hybridization (GISH) was used to identify the chromosomal constitutions of the blue-grained wheat Blue-58 originated from distant hybridization between Triticum aestivum L. and Agropyron elongatum (Host) Beauv (2n = 10x = 70) and its irradiation-mutated offsprings. The results showed that, as previously reported, Blue-58 (2n = 42) was a substitution line in which a pair of 4D chromosomes of common wheat was substituted by a pair of 4E chromosomes from Agropyron elongatum. Mutation line LW004 might be a homozygous reciprocal translocation line in which two pair of chromosomes might be involved in the translocation event and it showed high efficiency in utilizing soil phosphorus in our field screening experiments. Mutation line LW43-3-4 was a blue monosomic line with 40 wheat chromosomes plus one 4E chromosome from Agropyron elongatum in its genome and its seeds showed light-blue color. Some other chromosome rearrangements and structure changes were also detected among the mutated offsprings through GISH, such as telomonosomic line (40 W + 1't4E) and a line with a chromosomal constitution of 39 W + 1'4E + 1't4E. Our results indicated very clearly that it was the chromosome derived from Agropyron elongatum carried gene(s) responsible for the blue pigmentation in wheat grains and it was also illustrated from our results that the combination of effective methods for generating mutations at chromosomal level and convenient detection of these mutations played pivotal roles in creating materials for chromosome engineering and in breeding by means of chromosome engineering.

[Expression, purification of the recombinant protein fragment of the monkey epididymis expressed gene ESc-615 and preparation of polyclonal antisera to it].

ESc-615 is a gene which was specifically expressed in epididymis, newly discovered in our laboratory. To elucidate the function of ESc-6 15 in sperm maturation and fertilization at the protein level, its C-terminal 310 amino ac id residues fragment was expressed in E.coli and used to immunize rabbits to obtain polyclonal antiserum with titer of 100 000. The antisera could detect th e antigen as low as 3 ng. A 63 kD protein in rat epididymis protein extracts could be detected in Western blot by using this particular antiserum, in accordance with the size of Esc-615 homologue polypeptide deduced from its cDNA. Immunostaining results showed that Esc-615 was a secreted protein and could bind sperm s, which was similar with other carboxylesterases in male reproductive system; t he positive staining was eliminated after antigen absorption. All these results showed that this antiserum was not only effective but also highly specific, providing a useful tool in further functional studies of Esc-615.

[Prostaglandin E1 versus sildenafil in the management of erectile dysfunction].

OBJECTIVES: To evaluate the outcome of treatment in patients with erectile dysfunction (ED) using sildenafil or intracavernosal injection of prostaglandin E1(PGE1). METHODS: 54 patients with ED were randomly classified into two groups and received either oral sildenafil (group A) or intracavernosal injection of PGE1(group B) for 4-9 months with an average of 6 months. RESULTS: The percentages of efficacy in the two groups were 80.0% and 83.3%, respectively. There was no statistical difference between group A and B (P > 0.05). Two of six patients who did not respond to sildenafal in group A achieved erections sufficient for sexual intercourse when the six patients received intracavernous injection of PGE1. None of the four patients who did not respond to intracavernous injection of PEG1 in group B achieved erection sufficient for sexual intercourse when they received oral sildenafil. CONCLUSIONS: Both oral sildenafil and intracavernous injection of PGE1 are effective for patients with ED of various etiologies. The patients who do not respond to sildenafil can receive intracavernous injection therapy. The satisfactory results can probably achieved.

[Telomerase expression in tarsal gland carcinoma].

OBJECTIVE: To investigate the roles of telomerase activity (TA) and human telomerase RNA (hTR) in tarsal gland carcinoma. METHODS: The telomerase repeated amplification protocol (TRAP) was used to demonstrate telomerase activity in 12 tarsal gland carcinomas. In situ hybridization (ISH) was used to demonstrate the expression of hTR in 55 cases of paraffin-embedded tarsal gland carcinoma, and the results were compared with the proliferative index determined by Mib-1 immunolabeling, histological patterns and recurrence of the tumor. RESULTS: The telomerase activity was revealed in 12 tarsal gland carcinomas with different degrees. The positive expression of hTR was 85.5% (47/55) and observed in tumor cells, but not in the adjacent tissues. The expression of hTR was correlated with the proliferative activity (as assessed by Mib-1 immunolabelling, r = 0.942, P < 0.001) and the differentiation of tarsal gland carcinoma (chi(2) = 17.621, P < 0.001), but no significant relationship with tumor recurrence. The level of hTR expression had a gradually raised tendency along with the decrease of differentiation of tarsal gland carcinoma. CONCLUSION: The results suggest that up-regulation of telomerase expression play some roles in tarsal gland carcinogenesis, and the expression of hTR be a useful marker for malignant degree of tarsal gland carcinoma.

[Effect of preoperative intra-arterial chemotherapy on apoptosis and p53 expression of gastric cancer].

BACKGROUND & OBJECTIVE: Regional chemotherapy is charactized by a high drug concentration in tumor tissues and slight side-effects. Some significant histopathological changes were observed in the patients with gastric carcinoma treated by selective intra-arterial infusion chemotherapy before operation. The aim of this study was to investigate the effect of preoperative intra-arterial chemotherapy on apoptosis and p53 expression in human gastric cancer and its clinical significance. METHODS: The apoptosis was examined by terminal uridine deoxynucleotide nick end labeling (TUNEL) and the expression of p53 was detected with immunohistochemistry in 33 patients with gastric carcinoma pre- and post-chemotherapy respectively. RESULTS: Apoptosis index(AI) was significantly higher in the post-chemotherapeutic patients than in the pre-chemotherapeutic ones (19.29 +/- 6.48 versus 9.24 +/- 5.03, P < 0.01). In contrast, the positive expression rate of p53 was significantly lower, in the post-chemotherapeutic patients, than in the pre-chemotherapeutic ones(30.30% versus 54.55%, P < 0.05). In the post-chemotherapeutic patients, the droped group of p53 expression were strongly associated with a better clinical response(P < 0.05). CONCLUSIONS: Preoperative intra-arterial chemotherapy could enhance the apoptosis of gastric cancer cell, decrease the level of p53 expression and keep the patients for a longer survival.

[The expression of Fas, FasL and Bcl-2 on RMA cells during the process of apoptosis induced by chemotherapeutic drugs].

The objective of the study is to explore the effect of Fas, FasL and Bcl-2 on the process of apoptosis induced by chemotherapeutic drugs through detecting the expression of Fas, FasL and Bcl-2 on murine lymphoma cell line RMA. Dexamethasone(DEX), etoposide (VP-16), arsenic trioxide As(2)O(3) and all trans-retinoic-acid (ATRA) were added to the RMA cells as well as to the cells preincubated with interleukin-2 (IL-2), interleukin-6 (IL-6) or granulocyte-macrophage colony-stimulating factor (GM-CSF), respectively. The effect on apoptosis was observed and the expression of Fas and FasL mRNA as well as the expression of Fas and Bcl-2 antigen were measured. DEX and VP-16 could promote apoptosis of RMA cells while upregulating the expression of Fas and FasL without affecting the expression of Bcl-2. ATRA downregulated the expression of Bcl-2 without any change of Fas and FasL, and no apoptosis of RMA cells induced by ATRA was observed. Although As(2)O(3) induced apoptosis of RMA cells, it did not affect the expression of Fas, FasL and Bcl-2, which suggested that different drugs induce apoptosis of the same kind of cells by different signal transduction system and apoptosis induced by Fas system needed the coexistence of Fas and FasL. Although IL-2, IL-6 and GM-CSF upregulated the expression of Fas protein when adding to RMA cells separately, none of them induced apoptosis. Apoptosis could be induced by combination of IL-2 and IL-6 along with the upregulation of Fas and FasL. The cytokines facilitated the apoptotic action of chemotherapeutic drugs, the drug concentration for inducing apoptosis decreased and the time period of starting apoptosis shortened. Apoptosis could be observed without the expression of FasL when anti-Fas-antibody was added to RMA cells. The results demonstrated that there was synergistic effect of chemotherapeutic drugs and some cytokines for induction of apoptosis. Fas-FasL system participated in the apoptosis induced by DEX and VP-16; different drugs induce apoptosis by different pathway of signal transduction.

[Effect of shengji huayu recipe and its disassembled formulae on type I and III collagen synthesis in wound healing fibroblasts].

OBJECTIVE: To explore the mechanism of Shengji Huayu Recipe (SJHYR) and its 3 disassembled formulae in accelerating wound healing toward skin restoration. METHODS: Using in vitro cultured fibroblasts from granulation tissue of wound and compared with the normal skin fibroblast of suckling rat, who were treated separately by drug serum containing high and low dose SJHYR and its disassembled prescriptions (Shengji formula and Huayu formula). The type I and III collagen contents in the fibroblasts were determined with immunocytochemical ABC method. RESULTS: Shengji formula could increase the levels of type I and III collagens in fibroblasts, which was significantly higher than those in model cells and control (P < 0.01). Huayu formula lowered them to the levels below those in model cells (P < 0.01), while in comparing the levels in the cells treated by high or low dose SJHYR with those in control, no significant difference was shown(P > 0.05). CONCLUSION: SJHYR might accelerate wound healing toward skin restoration through regulating the ratio of collagen type I and III, to adjust their metabolism.

[Comparative study of transurethral electrovaporization of prostate versus transurethral resection of prostate on benign prostatic hyperplasia].

OBJECTIVES: To compare the efficacy of transurethral electrovaporization of prostate (TUVP) with transurethral resection of prostate (TURP). METHODS: 206 patients with symptomatic benign prostatic hyperplasia (BPH) whose prostatic sizes were all less than 60 grams were randomly divided into two groups. 97 cases were treated by TUVP while the other 109 cases were treated by TURP. The patients who underwent either TUVP or TURP were followed up for 12-34 months with an average of 20 months postoperatively. RESULTS: Both groups showed the significant decline in the mean IPSS (international prostatic symptom score) (P < 0.01), the mean PVR (Postovoiding Residual Volume) (P < 0.01), while increase in mean Qmax (Peak uroflow rate) (P < 0.01) in 12 months, 24 months after the operation. There were significant differences in the mean duration of operation or catheterization postoperatively (P < 0.05). The main complications of post-operation in the two groups were stress incontinence, TUR syndrome, urethral stricture, secondary bleeding. CONCLUSIONS: Both TUVP and TURP are effective treatment for the patient with BPH whose prostatic size is less than 60 grams. TUVP spends shorter time of the operation and postoperative catheterization than that of TURP.

[Function analysis of a wheat phosphate transporter in yeast mutant].

Phosphorus is a major nutrient acquired by plants and phosphorus availability is considered one of the major growth-limiting factors for plants in many natural ecosystems. For a better understanding of the function of the wheat phosphorus transporter gene named TaPT2, we did Southern blot analysis and studied its function with complementation test in yeast mutant strain MB192. Southern blot indicated that TaPT2 gene is a low-copy member and has several different members in wheat genome. In the function complementation study, TaPT2 has a similar function as PHO84 which is the phosphate transporter in yeast Saccharomyces cerevisiae. Both TaPT2 and PHO84 are able to complement the PHO84 mutant phenotype of yeast strain MB192 which lacks the phosphate transporter activity. Therefor, this assessment presents evidence that TaPT2 gene plays an important role in Pi acquisition.

Ultrasound-Induced Formation of CdS Nanostructures in Oil-in-Water Microemulsions.

In this study the formation of approximately spherical assemblies of nanocrystalline primary particles of CdS in a CS(2)-water-ethylenediamine (CWE) microemulsion induced by ultrasonic irradiation is described. CS(2) was employed as the sulfur source for CdS and also as the oil phase in the microemulsion. The particles were studied with X-ray powder diffraction, transmission electron microscopy, UV/Vis absorption spectroscopy, and photoluminescence spectroscopy. A blue shift compared to bulk CdS was observed in absorption and photoluminescence spectra. Copyright 2001 Academic Press.

Associations between XRCC1 and ERCC2 polymorphisms and DNA damage in peripheral blood lymphocyte among coke oven workers.

A wide variety of base damages and single-strand breaks formed by reactive oxygen species during metabolic activation of polycyclic aromatic hydrocarbons (PAHs) have been recognized to be involved in PAH carcinogenesis. In this study, alkaline comet assay was used to detect the DNA damage in peripheral blood lymphocytes among 143 coke-oven workers and 50 non-coke-oven workers, and the effects of genetic polymorphisms of XRCC1 and ERCC2 genes on DNA damage were evaluated. The olive tail moment was significantly higher in coke-oven workers than in non-coke-oven workers (2.6, 95% CI=2.1-3.3 versus 1.0, 95% CI=0.8-1.2, p<0.01), and significant correlation between ln-transformed urinary 1-OHP and ln-transformed olive tail moment was found in total population (n=193, Pearson's r=0.393, p<0.001) and in coke-oven workers (n=143, Pearson's r=0.224, p=0.007). The olive tail moment was significantly higher in coke-oven workers with GA genotype of G27466A polymorphism of XRCC1 than those with GG genotype (4.6, 95% CI=2.5-8.7 versus 2.4, 95% CI=1.9-2.9, p<0.01 with adjustment for covariates). No significant associations between C26304T, G28152A and G36189A polymorphisms of XRCC1 and G23591A and A35931C polymorphisms of ERCC2 and olive tail moment were found in both groups. The study showed that the alkaline comet assay is a suitable biomarker in the detection of DNA damage among coke-oven workers and it suggested that the A allele of G27466A polymorphism of XRCC1 may be associated with decreased DNA repair capacity toward PAH-induced base damage and strand breaks.
