OBJECTIVE: To explore the influence of immune-enhanced enteral diet on postoperative metabolism and immunological function in colorectal tumor patient. METHODS: Seventy-two patients with colorectal cancer were enrolled in this study. Immune-enhanced enteral diet was administrated in 36 cases (study group), while regular enteral nutrition (EN) was given in 36 cases (control group). The immunological function, inflammatory function and nutrition index were evaluated before operation and on postoperative day 1 and day 8. RESULTS: Serum levels of glutamine, asparagines, CD4 (+), NK, IgG, IgM were higher in the study group than those in the control group. The levels of IL- 6, TNF-alpha in the study group were lower than those in the control group. The difference was statistically significant (P< 0.05). CONCLUSIONS: Immune-enhanced enteral diet can ameliorate the cell immunological and fluid immunological function in the patients with colorectal cancer after the procedures.
OBJECTIVE: To investigate the frequency of HFE gene variants in patients with hepatocellular carcinoma following chronic hepatitis B and to analyze their relationships. METHODS: 56 patients with hepatocellular carcinoma following chronic hepatitis B (HCC group) and 60 healthy blood donors (control group) were studied for the amino acid dimorphism at codon 63 (His63Asp=H63D) and codon 282 (Cys282Tyr = C282Y) of the HFE gene. The codon 63 and 282 dimorphisms were defined by PCR amplification of genomic DNA samples and restriction enzyme digestion using RsaI for C282Y and BclI for H63D. The association between hepatocellular carcinoma following chronic hepatitis B and HFE mutations were analyzed by Chi-square test. RESULTS: The genotype frequency of C2/C2 in the HCC group was markedly higher than that in the normal control group (10.7% vs 0) and there was a significant correlation between them. At the same time, the allele frequency of C2 in the HCC group was markedly higher than that in the normal control group (16.1% vs 1.7%) and there was a significant correlation between them also. CONCLUSION: The mutation of C282Y may be related with susceptibility to HCC after chronic hepatitis B. This outcome suggests that host HFE mutation may be an important factor related to the pathogenesis of HCC.
OBJECTIVE: To study the predictive factors on mortality and survival of elderly dementia in Shanghai. METHODS: Subjects were from people who had been screened in a prevalence survey study from the selected 5 urban and 4 rural communities from 1997 to 1998. Phase I screening was carried out by a brief memory testing with mini-mental state examination. In phase II, those who scored below the cutoff points and 4% of those whose score was in the normal range were interviewed to identify dementia through a set of diagnostic examinations according to the DSM IV criteria. In phase III which was six months later, a follow-up program was conducted for all persons who were involved in phase II to identify those with Alzheimer's dementia (AD) and vascular dementia (VaD) according to NINCDS-ADRDA and NINDA-AIREN criteria and other types of dementia. RESULTS: The mortality of elderly dementia was 6.06/1000 person-years. Compared to nondementia group, the survival rate of dementia group had a decrease of 57% after 40 months of follow-up (P < 0.05). There was no statistical difference between AD and VaD groups. The relative risk of death caused by dementia, AD and VaD were 1.63 (95% CI: 1.42-1.86), 1.71 (95% CI: 1.44-2.03) and 1.45 (95% CI: 1.16-1.82), respectively. Some factors such as age (RR = 1.0685), severalty (RR = 1.5733), and high ADL (RR = 1.0368) might have increased the risk of death among those patients. CONCLUSION: The mortality of elderly dementia in Shanghai was lower than that seen in other areas in the world. There was no obvious difference between the survival rates of AD and VaD patients. Age, severalty and high ADL seemed to be the risk factors to the death outcome.
OBJECTIVE: To find out whether phospholamban (PLN) is the virulence gene of dilated cardiomyopathy (DCM) in the Chinese of Chengdu. METHODS: DNA was isolated from 89 hospitalized unrelated patients with DCM and 110 healthy Chinese Hans as controls. The PLN mutations was screened by polymerase chain reaction (PCR)-single strand conformation polymorphisms (SSCP) and nucleotide sequence analysis. RESULTS: In polyacrylamide gel lectrophoresis, no abnormal conformer was found in the two groups. In DNA sequence, no C --> T missense mutation at nucleotide 25 was identified in the DCM patients and the controls. Meanwhile, T --> G missense mutation at nucleotide 116 was not found in the affected individuals and in the controls. CONCLUSION: No PLN gene mutation was found in patients with DCM in Chengdu. This result indicated that PLN gene mutation may not be a common cause for DCM in the Chinese population in Chengdu.
The site-directed mutagenesis of HA gene was made by using PCR, and mismatches were introduced into primers. Mutagenesis was performed in a three-step PCR. The amplified fragments from the second PCR which contained the mutation site were cloned into the pcDNA3 vector, named pHAm. The sequencing analysis showed that the mutation site was correct. The amino acid sequence at the cleavage site of the HA protein was from RKKR decrease GLF to RSSR decrease GLF. The recombinant plasmid pHAm was transiently transfected into 293T cells by the calcium phosphate precipitation method. Indirect immunofluorescent assay (IFA), confirmed expression of the HA protein on the cell membrane, the mutant HA gene was a promising candidate for further studies.
OBJECTIVE: To identify the species of biological samples by amplifying intron 8 of the TP53 gene. METHODS: Collected the bloodstains or muscle tissues of 14 kinds of common animals and human. DNA were quantified after extraction. The PCR amplification products were analyzed by PAGE and silver staining. RESULTS: The human and monkey have one amplification band of 460 bp while the eel, fish, frog, duck, rabbit, cat, mouse, cavy, pig, ox and sheep could amplify different unspecial bands. No amplification products were found in chicken and dog. CONCLUSION: The method of species identification by amplifying intron 8 of the TP53 gene is simple and sensitive.
The relationship between Human papillomavirus (HPV) 16 infection and the natural course of colorectal adenocarcinoma has not been fully defined. In this study, the HPV 16 E7 DNA was detected in 82 patients with primary colorectal adenocarcinoma to study the relationship between HPV 16 infection and colorectal carcinoma. Samples were taken from both the tumors and the adjacent normal mucosa in the same patient. Polymerase chain reaction (PCR) was used to amplify the HPV16 E7 DNA fragment. The PCR products were gel-purified and sequenced for HPV genotyping. DNA sequence analysis indicated that PCR product was 297 bp. It was the equivalent of 562-858th pairs in the HPV 16 primitive sequences. Our results showed HPV16 E7 DNA expression was significantly higher in colorectal carcinoma (42/82) than in adjacent normal mucosa (4/82). The correlation was found between HPV16 E7 expression and tumor's location; the positive rate was 18.18% in the ascending colon carcinoma group and 64.10% in the rectal carcinoma group. High HPV16 E7 expression was also associated with lower Dukes stage (P < 0.01). These results indicated that there was correlation between colorectal adenocarcinoma and HPV 16 infection. HPV16 infection was relatively higher in the colorectal carcinoma and rare in the adjacent normal mucosa. Specimens expressing higher levels of HPV 16 E7 DNA were associated with lower Dukes stage and more distal location.
This paper reviewed application of chitin and its derivatives in pharmaceutical industry of traditional Chinese medicine in recent years. Chitin and its derivatives could be used as a clarificant, retaining active substances, stabiliting preparation, reducing the heavy metals of traditional Chinese medicine extraction. Because of their churacteristics in biodegradation, biocompatibility and its tendency to film easily, chitin and its derivatives was ideal assistant materials in traditional Chinese medicine. Its application was largely enhanced after chitosan was modified. The problems and prospects of the application of chitin and its derivatives were also discussed.
OBJECTIVE: To study the effects of doxofylline on calcium-activated potassium (K(Ca)) channels of human peripheral blood eosinophils in asthma. METHODS: Peripheral blood eosinophils from patients with asthma were isolated and divided equally into two groups, a control group and a doxofylline incubated group. The data were recorded using cell-attached configuration of patch-clamp technique and the kinetic changes of K(Ca) channels activated by 0.2 micromol/L platelet activating factor (PAF) were compared. RESULTS: As compared with the control group, the open probability of the K(Ca) channels decreased from 0.135 +/- 0.021 to 0.044 +/- 0.018, the open time from (5.75 +/- 0.40) ms to (2.39 +/- 0.13) ms, while the close time increased from (2.17 +/- 0.50) ms to (23.73 +/- 2.50) ms in the doxofylline incubated group. The differences were significant between the two groups (all P < 0.05). CONCLUSION: Doxofylline could decrease the open probability of the channels as a result of both the shortening of open period and the prolongation of close time.
The Simon effect refers to the finding that reaction times are faster when stimulus and response locations are congruent than when they are not, even stimulus locations are task irrelevant. Zhang and Johnson reported a Simon-like spatial congruence effect in working memory. This study examined the neural mechanisms of this memory-based spatial congruence effect by recording event-related potentials to probe stimuli. Behavioral results showed a clear congruence effect. The P300 amplitudes were larger in the congruent condition than in the incongruent condition. The results suggest that the memory-based congruence effect and the classical Simon effect were mediated by similar neural mechanisms and support theories assigning response selection an essential role in spatial congruence effects.
The cancer metastasis suppressor protein KAI1/CD82 is a member of the tetraspanin superfamily. Recent studies have demonstrated that tetraspanins are palmitoylated and that palmitoylation contributes to the organization of tetraspanin webs or tetraspanin-enriched microdomains. However, the effect of palmitoylation on tetraspanin-mediated cellular functions remains obscure. In this study, we found that tetraspanin KAI1/CD82 was palmitoylated when expressed in PC3 metastatic prostate cancer cells and that palmitoylation involved all of the cytoplasmic cysteine residues proximal to the plasma membrane. Notably, the palmitoylation-deficient KAI1/CD82 mutant largely reversed the wild-type KAI1/CD82's inhibitory effects on migration and invasion of PC3 cells. Also, palmitoylation regulates the subcellular distribution of KAI1/CD82 and its association with other tetraspanins, suggesting that the localized interaction of KAI1/CD82 with tetraspanin webs or tetraspanin-enriched microdomains is important for KAI1/CD82's motility-inhibitory activity. Moreover, we found that KAI1/CD82 palmitoylation affected motility-related subcellular events such as lamellipodia formation and actin cytoskeleton organization and that the alteration of these processes likely contributes to KAI1/CD82's inhibition of motility. Finally, the reversal of cell motility seen in the palmitoylation-deficient KAI1/CD82 mutant correlates with regaining of p130(CAS)-CrkII coupling, a signaling step important for KAI1/CD82's activity. Taken together, our results indicate that palmitoylation is crucial for the functional integrity of tetraspanin KAI1/CD82 during the suppression of cancer cell migration and invasion.
Gene targeting in the mouse has become a standard approach, yielding important new insights into the genetic factors underlying cardiovascular development and disease. However, we still have very limited understanding of how mutations affect developing cardiovascular function, and few studies have been performed to measure altered physiological parameters in mouse mutant embryos. Indeed, although in utero lethality due to embryonic heart failure is one of the most common results of gene targeting experiments in the mouse, the underlying physiological mechanisms responsible for embryonic demise remain elusive. Using in utero ultrasound biomicroscopy (UBM), we studied embryonic day (E) 10.5 to 14.5 NFATc1-/- embryos and control littermates. NFATc1-/- mice, which lack outflow valves, die at mid-late gestation from presumed defects in forward blood flow with resultant heart failure. UBM showed increasing abnormal regurgitant flow in the aorta and extending into the embryonal-placental circulation, which was evident after E12.5 when outflow valves normally first develop. Reduced NFATc1-/- net volume flow and diastolic dysfunction contributed to heart failure, but contractile function remained unexpectedly normal. Among 107 NFATc1-/- embryos imaged, only 2 were observed to be in acute decline with progressive bradyarrhythmia, indicating that heart failure occurs rapidly in individual NFATc1-/- embryos. This study is among the first linking a specific physiological phenotype with a defined genotype, and demonstrates that NFATc1-/- embryonic heart failure is a complex phenomenon not simply attributable to contractile dysfunction.
Cantharidin is a natural toxin that has antitumor properties and causes leukocytosis as well as increasing sensitivity of tumor cells resistant to other chemotherapeutic agents. There is limited information, however, on the molecular pharmacological mechanisms of cantharidin on human cancer cells. We have used cDNA microarrays to identify gene expression changes in HL-60 promyeloid leukemia cells exposed to cantharidin. Cantharidin-treated cells not only decreased expression of genes coding for proteins involved in DNA replication (e.g., DNA polymerase delta), DNA repair (e.g., FANCG, ERCC), energy metabolism (e.g., isocitrate dehydrogenase alpha, ADP/ATP translocase), but also decreased expression of genes coding for proteins that have oncogenic activity (e.g., c-myc, GTPase) or show tumor-specific expression (e.g., phosphatidylinositol 3-kinase). In contrast, these treated cells overexpressed several genes that encode intracellular and secreted growth-inhibitory proteins (e.g., BTG2, MCP-3) as well as proapoptotic genes (e.g., ATL-derived PMA-responsive peptide). Our findings suggest that alterations in specific genes functionally related to cell proliferation or apoptosis may be responsible for cantharidin-mediated cytotoxicity. We also found that exposure of HL-60 cells to cantharidin resulted in the decreased expression of multidrug resistance-associated protein genes (e.g., ABCA3, MOAT-B), suggesting that cantharidin may be used as an oncotherapy sensitizer, and the increased expression of genes in modulating cytokine production and inflammatory response (e.g., NFIL-3, N-formylpeptide receptor), which may partly explain the stimulating effects on leukocytosis. Our data provide new insight into the molecular mechanisms of cantharidin.
In order to study the character of periodontal ligament cells (PDLCs) attaching on commercially pure titanium (cpTi) by morphology and metrology on the early stage (24 h), 1 x 10(5)/ml PDLCs in 2 ml culture medium were seeded on cpTi discs fixed in 24-well culture plates. Morphology of cell attachment was observed by contrast phase microscope, scanning electron microscope (SEM) and fluroscence microscopy. Cell adhesion was analyzed by MTT at 0.5, 1, 2, 4 h respectively. PDLCs could attach and spread on cpTi discs. SEM showed that PDLCs had pseudopod-like protuberance. PDLCs showed different attaching phases and reached saturation in cell number at 2 h. It was concluded that PDLCs had good biocompatibility with cpTi, and showed a regular and dynamic pattern in the process of attaching to cpTi.
The Household Responsibility System initiated in the late 1970s in China has brought a profound change to its rural economy. The shift from the collective farming system to individual family farms has changed land management. The change, including fertilization and crop systems, may have significant effects on soil quality and agro-environmental sustainability. However, very little research is being carried out on the impact of reformed land tenure systems on the spatial variability of soil nutrients. In this study, geostatistics was applied to analyze changes in the spatial variability of soil organic matter and nutrients in paddy fields in Pinghu County, China after 20 years of land management change. In 1984 and 2002, 617 and 131 locations were selected, respectively, for collecting surface soil samples to analyze soil organic matter (SOM), total nitrogen (TN), available phosphorus (AP), and available potassium (AK). From 1984 to 2002, variability of the SOM and TN changed from strongly to moderately spatial-dependent, and the variability of AP remained weakly dependent on space, but that of the AK changed from moderately to weakly spatial-dependent. That the trend of the variability of four soil properties in 2002 became weaker than that in 1984 showed that the extrinsic factors (soil management practices, such as fertilization) weakened the effect by intrinsic factors (soil formation factors, such as soil parent materials) owing to a long period of land management change. The temporal geographic maps of the SOM and nutrients spatial distributions suggested that the concentrations of the SOM and nutrients had been changed to different extents during the period. Significant increase in AP and decrease in AK were noted. The changes were likely due to the imbalance between N, P, and K fertilizers and increased grain yield.
A flavone (5,7,4'-trihydroxy-3',5'-dimethoxyflavone), a cyclohexenone (3-isopropyl-5-acetoxycyclohexene-2-one-1) and a liquid mixture of low polarity, containing long-chain and cyclic hydrocarbons, were isolated from leaves of allelopathic rice accession PI 312777 using column chromatography. Their structures and constituents were identified by means of HR-MS, NMR and GC/MS analyses, respectively. Bioassays showed that both the flavone and cyclohexenone significantly inhibited the growth of weeds Echinochloa crus-galli, Cyperus difformis and Cyperus iris, and the spore germination of fungal pathogens Pyricularia oryzae and Rhizoctonia solani at all tested concentrations. Moreover, the combination of the inactive mixture of low polarity and the active flavone or cyclohexenone significantly enhanced the inhibitory activities on weed growth. In addition, the two compounds and the mixture of low polarity from the leaves of PI312777 did not inhibit the rice growth at the same concentrations. It was also established that both compounds could be released into the soil, and was especially induced by E. crus-galli. The results suggest that 5,7,4'-trihydroxy-3',5'-dimethoxyflavone and 3-isopropyl- 5-acetoxycyclohexene-2-one-1 may act as allelochemicals participating in the defense of rice against weeds and pathogens.
This article presents two approaches for automated building of knowledge bases of soil resources mapping. These methods used decision tree and Bayesian predictive modeling, respectively to generate knowledge from training data. With these methods, building a knowledge base for automated soil mapping is easier than using the conventional knowledge acquisition approach. The knowledge bases built by these two methods were used by the knowledge classifier for soil type classification of the Longyou area, Zhejiang Province, China using TM bi-temporal imageries and GIS data. To evaluate the performance of the resultant knowledge bases, the classification results were compared to existing soil map based on field survey. The accuracy assessment and analysis of the resultant soil maps suggested that the knowledge bases built by these two methods were of good quality for mapping distribution model of soil classes over the study area.
