Characteristics and living patterns of marine myxobacterial isolates.

The growth, morphology, and life cycle of two marine myxobacterial isolates, halotolerant Myxococcus fulvus strain HW-1 and halophilic Haliangium ochraceum strain SMP-2, were studied as models to determine the living patterns of myxobacteria in the ocean. The growth, morphology, and development of halotolerant strain HW-1 shifted in response to salinity. The optimal seawater concentration for growth of HW-1 was 0 to 80% (salinity, 0.1 to 2.9%), and the strain grew poorly in media with a salinity of more than 4%. The cells became shorter as the seawater concentration increased. The fruiting body structure was complete only on agar prepared with low concentrations of seawater or salts (less than 60% seawater; salinity, 2.1%), and rudimentary structures or even simple cell mounds appeared as the seawater concentration increased. In contrast, the halophilic strain SMP-2 was unable to grow without NaCl. The cell length and the morphology of the fruiting body-like structure did not change in response to salts. In seawater liquid medium, the cells of both strains were confirmed to be able to form myxospores directly from vegetative cells, but they could not do so in medium containing a low seawater concentration (10% or less). HW-1 cells from medium containing a high concentration of seawater grew independent of cell density, while cells from medium containing a low concentration of seawater (10% or less) showed density-dependent growth. SMP-2 cells showed density-dependent growth under all salinity conditions. The results suggest that the halotolerant myxobacteria are the result of degenerative adaptation of soil myxobacteria to the marine environment, while the halophilic myxobacteria form a different evolutionary group that is indigenous to the ocean.

Inducible antisense RNA expression in the characterization of gene functions in Streptococcus mutans.

In order to examine gene function in Streptococcus mutans, we have recently initiated an antisense RNA strategy. Toward this end, we have now constructed and evaluated three Escherichia coli-S. mutans shuttle expression vectors with the fruA and scrB promoters from S. mutans, as well as the tetR-controlled tetO promoter from Staphylococcus aureus. Among these, the tetO/tetR system proved to be the most tightly controlled promoter. By using this shuttle plasmid system, modulation of gene function by inducible antisense RNA expression was demonstrated for comC antisense fragments of different sizes as well as for distinct gtfB antisense fragments. It was demonstrated that the size, but not the relative position, of an antisense DNA fragment is important in mediating the antisense phenomenon. Furthermore, by constructing and screening random DNA libraries with the tet expression shuttle system, 78 growth-retarded transformants harboring antisense DNA fragments were also identified. Almost all of them corresponded to homologous essential genes in other bacteria. In addition, a novel essential gene, the coaE gene, encoding dephospho-coenzyme A kinase, which is involved in the final step of coenzyme A catabolism in S. mutans, was identified and characterized. These results suggest that the antisense RNA strategy can be useful for identifying novel essential genes in S. mutans bacteria as well as further characterizing the physiology (including potential virulence factors) of these organisms.

Mechanisms through which bradykinin promotes glomerular injury in diabetes.

In diabetes, mesangial cell proliferation and extracellular matrix expansion are critical components in the development of glomerulosclerosis. We reported that diabetes alters the activity of the kallikrein-kinin system and that these alterations contribute to the development of diabetic nephropathy. The present study examined the influence of streptozotocin-induced diabetes on the renal expression of bradykinin (BK) B2 receptors (B2KR), connective tissue growth factor (CTGF), transforming growth factor-beta (TGF-beta), and TGF-beta type II receptor (TGF-betaRII) and assessed the signaling mechanisms through which B2KR activation may promote glomerular injury. Eight weeks after the induction of diabetes, renal mRNA levels of B2KR, CTGF, and TGF-beta as well as protein levels of CTGF and TGF-betaRII were measured in control (C), diabetic (D), and insulin-treated diabetic (D+I) rats. Renal B2KR and TGF-beta mRNA levels expressed relative to beta-actin mRNA levels and CTGF and TGF-betaRII protein levels were significantly increased in D and D+I rats compared with C rats (P < 0.03, n = 5). To assess the contribution of B2KR activation on modulating the expression of CTGF, TGF-betaRII, and collagen I, mesangial cells (MC) were treated with BK (10(-8) M) for 24 h and CTGF and TGF-betaRII protein levels were measured by Western blots and collagen I mRNA levels were measured by RT-PCR. A two- to threefold increase in CTGF and TGF-betaRII protein levels was observed in response to BK stimulation (P < 0.001, n = 6). In addition, a marked increase in collagen I mRNA levels was observed in response to BK stimulation. Treatment of MC with BK (10(-8) M) for 5 min significantly increased the tyrosine phosphorylation of p60src kinase and of p42/p44 MAPK (P < 0.05, n = 4). Inhibition of src kinase by PP1 (10 microM) inhibited the increase in p42/p44 MAPK activation in response to BK. Finally, to determine whether BK stimulates CTGF, TGF-betaRII, and collagen I expression via activation of MAPK pathways, MC were pretreated with an inhibitor of p42/p44 MAPK (PD-98059) for 45 min, followed by BK (10(-8) M) stimulation for 24 h. Selective inhibition of p42/p44 MAPK significantly inhibited the BK-induced increase in CTGF, TGF-betaRII, and collagen I levels. These findings are the first to demonstrate that BK regulates the expression of CTGF, TGF-betaRII, and collagen I in MC and provide a mechanistic pathway through which B2KR activation may contribute to the development of diabetic nephropathy.

Effects of prenatal irradiation with an accelerated heavy-ion beam on postnatal development in rats: I. Neurophysiological alterations.

Effects on postnatal neurophysiological development in offspring were studied after exposure of pregnant Wistar rats to accelerated carbon-ion beams with an LET of about 13 keV/ mum at doses ranging from 0.1 Gy to 2.5 Gy on the 15th day of gestation. The age at which four physiological markers appeared and five reflexes were acquired was examined prior to weaning. Gain in body weight was monitored until the offspring were 3 months old. Male offspring were evaluated as young adults using two behavioral tests. The effects of X rays estimated for the same biological end points were studied for comparison. For most of the end points at early age, no significant alterations were observed in offspring that received prenatal irradiation with 0.1 Gy of either accelerated carbon ions or X rays compared to the offspring of sham-irradiated dams. However, all offspring whose dams received 2.5 Gy died prior to weaning. Offspring from dams irradiated with accelerated carbon ions generally showed higher incidences of prenatal death and preweaning mortality, markedly delayed accomplishment in their physiological markers and reflexes, and gain in body weight compared to those exposed to X rays at doses of 0.5 to 2 Gy. Significantly reduced ratios of main organ weight to body weight at the postnatal ages of 30, 60 and 90 days were also observed within this dose range. The results indicate that irradiation with 0.5 to 2 Gy on day 15 of gestation caused permanent alterations in offspring that were dependent on dose. The alterations include permanent growth retardation, morphological malformations in main organs, including microcephaly, diminished reflex attainment, delayed appearance of physiological markers, and changes in adult behavior. Exposure to 1 to 2 Gy of radiation resulted in growth retardation and behavioral alterations that persisted throughout life. Accelerated carbon ions generally induced more detrimental effects than X rays.

Fine-mapping gene-by-diet interactions on chromosome 13 in a LG/J x SM/J murine model of obesity.

Obesity is one of the most serious threats to human health today. Although there is general agreement that environmental factors such as diet have largely caused the current obesity pandemic, the environmental changes have not affected all individuals equally. To model gene-by-environment interactions in a mouse model system, our group has generated an F(16) advanced intercross line (AIL) from the SM/J and LG/J inbred strains. Half of our sample was fed a low-fat (15% energy from fat) diet while the other half was fed a high-fat (43% energy from fat) diet. The sample was assayed for a variety of obesity- and diabetes-related phenotypes such as growth rate, response to glucose challenge, organ and fat pad weights, and serum lipids and insulin. An examination in the F(16) sample of eight adiposity quantitative trait loci previously identified in an F(2) intercross of SM/J and LG/J mouse strains reveals locus-by-diet interactions for all previously mapped loci. Adip7, located on proximal chromosome 13, demonstrated the most interactions and therefore was selected for fine mapping with microsatellite markers. Three phenotypic traits, liver weight in male animals, serum insulin in male animals, and reproductive fat pad weight, show locus-by-diet interactions in the 127-kb region between markers D13Mit1 and D13Mit302. The phosphofructokinase (PFK) C (Pfkp) and the pitrilysin metalloprotease 1 (Pitrm1) genes are compelling positional candidate genes in this region that show coding sequence differences between the parental strains in functional domains.

[Diagnosis and surgical treatment of inflammatory pseudotumor of the lung: a report of 51 cases].

BACKGROUND & OBJECTIVE: Clinical symptoms and imaging presentations of inflammatory pseudotumor of the lung (IPL) lack of specialties, and always result in misdiagnoses. This article was to summarize features of IPL, and to improve diagnosis accuracy and appropriate operation selection. METHODS: Records of 51 patients with pathologically confirmed IPL, treated surgically in Department of Thoracic Surgery, Cancer Institute/Hospital, Chinese Academy of Medical Sciences from Jan. 1990 to Mar. 2000, were retrospectively reviewed. All patients were followed-up for at least 3 years. RESULTS: Of the 51 patients, before operation, only 3 (5.9%) had been correctly diagnosed, 24 (47.1%) were suspected having tuberculoma or other benign lung tumors, 10 (19.6%) with undefined occupied lesion in the lung, 14 (27.4%) were misdiagnosed as lung cancer. All patients received surgery, including lobectomy in 23 patients (45.1%), tumor excision in 22 patients (43.1%), wedge resection in 4 patients (7.8%), and segmental resection in 2 patients (3.9%). Only 1 (2.0%) had postoperative complication. No death occurred with a follow-up rate of 94.1% (48/51). CONCLUSION: Preoperative diagnosing IPL is quite difficult. Surgery should be performed as early as possible. The selection of surgery patterns, mainly include lobectomy and limited resection, depends on intraoperative pathology.

[Association between dopamine beta hydroxylase gene and attention deficit hyperactivity disorder complicated with disruptive behavior disorder].

OBJECTIVE: Attention deficit hyperactivity disorder (ADHD), a common behavior disorder of childhood, is a highly heterogeneous disease frequently accompanied by various mental disorders, including disruptive behavior disorder (DBD). Studies show that children suffering from ADHD with DBD are at higher risk of antisocial personality, substance abuse, and social adaptations disorder at their adulthood. The dopamine beta hydroxylase (DbetaH) is the key enzyme to ADHD since it catalyzes the conversion of dopamine to norepinephrine, and dysfunction there of is believed to be one of the causes of the disorder. To explore the association between DBH gene and ADHD complicated with or without DBD, the authors analyzed the transmission of a novel polymorphism DBH -1021C-->T, which is found associated with plasma DbetaH activity, in ADHD nuclear families using transmission disequilibrium test (TDT). METHODS: Consensus diagnoses were based on the DSM-IV. The samples included those from 292 Chinese Han nuclear families with ADHD probands. Genotypes of DBH -1021C-->T polymorphism were determined by PCR amplification, endonuclease digesting and electrophoresis. The transmission of DBH -1021C-->T polymorphism in ADHD nuclear families with or without DBD was analyzed by TDT. RESULTS: The results showed that there was transmission disequilibrium between DBH-1021C-->T polymorphism and ADHD with or without DBD. In ADHD comorbid with DBD, T allele was preferentially transmitted (P < 0.05); and in ADHD without DBD, so was the C allele (P < 0.05). Among the three subtypes of ADHD, only ADHD-C subtype with DBD had an increased transmission of T allele (P < 0.05). CONCLUSION: There is an association between DBH gene and ADHD comorbid with or without DBD, but the preferential transmission alleles are different. The low activity T allele is increased to transmit in ADHD with DBD, while the high activity C allele is preferentially transmitted in ADHD without DBD. The results support the proposition that the genetic mechanism is different between ADHD comorbid with or without DBD. We also found that only ADHD-C subtype with DBD is associated with DBH -1021C-->T polymorphism in three subtypes of ADHD, which may suggest that there is a more intense relationship between DBD and ADHD-C subtype.

Changes of osteocalcin and IGF-I during bone lengthening.

OBJECTIVE: To examine the changes of osteocalcin and insulin-like growth factor I (IGF-I) during bone lengthening, and to clarify the mechanism of bone healing. METHODS: Thirty-two shepherd dogs were divided into five groups randomly. Their tibiae were lengthened by Ilvzarov's external fixator at the rate of 1 mm/day. The lengthening area was the experimental side and the opposite side was the control. Samples were obtained on the 2nd, 4th, 6th, 8th, 12th weekend respectively. The samples were defatted, dried, powdered, centrifuged and measured by radioimmunoassay. RESULTS: The osteocalcin concentration increased at the subsequent periods, but it was significantly lower in the experimental side than that of the control side, P<0.05 and the IGF-I concentration was not significantly lowered. CONCLUSIONS: Different noncollagenous bone growth factors may be different at different stage.

[Nitric oxide and nitric oxide synthase in patients with chronic hepatitis B].

OBJECTIVE: To investigate nitric oxide (NO) and nitric oxide synthase (NOS) in patients with chronic hepatitis B. METHODS: Nitric oxide and nitric oxide synthase, including inducible NOS (iNOS) and constitutive NOS (cNOS), were measured in patients and control groups, then were statistically analyzed. RESULTS: NO and iNOS were significantly higher in patients with hepatitis B than in the controls (P < 0.05). NO and iNOS were significantly higher in patients with increased ALT than in the controls and in patients with normal ALT (P < 0.05). NO was significantly higher in patients with normal ALT than in the controls (P < 0.05). cNOS were not significant different among these groups. NO and iNOS significantly correlated with ALT in patients with hepatitis B (r=0.367 and r=0.474). No significant relationship was found among NO, NOS and HBV DNA. Among different genotype groups, NO and NOS had no significant difference. CONCLUSION: NO and NOS were higher in patents with chronic hepatitis B. In patients with increased ALT, NO's damage was severe. In patients with normal ALT, there was no significant damage caused by NO. NO should be detected in patients with hepatitis B in addition to HBV markers.

[Craniofacial characteristics of the Class II division 1 malocclusion patients with mouth-breating: a posteroanterior].

OBJECTIVE: To evaluate the craniofacial characteristics of the Class II malocclusion patients with mouth-breating by posteroanterior cephalometry. METHODS: To measure craniofacial width of the 12 Class II malocclusion patients with mouth-breathing, and to compared these measures with corresponding measures in a group of normal children. RESULTS: The width of the maxillary base bone (J-J) was less than that in normal children significantly (P < 0.01). The mouth-breathing children's upper and lower arch width (at first molar and cuspid) were comparatively narrower, and lateronasal width (Lap-Lap) was narrower too. CONCLUSION: Mouth breathing may lead to craniofacial morphological abnormal development in craniofacial transverse structures.

[The effect long-term administration of a selective beta1 blocker bisoprolol on glucose metabolism in patients with essential hypertensive and type 2 diabetes mellitus].

OBJECTIVE: To investigate the effects of bisoprolol on glucose metabolism and blood pressure in essential hypertensive patients with type 2 diabetes mellitus. METHODS: A total of 92 hypertensive patients with type 2 diabetes on stable antidiabetic therapy with HbAlc less than 7% were recruited. In a randomized, open trial the patients were treated for 12 weeks with bisoprolol or captopril following a 1-week placebo run-in period, the main parameters measured were HbA1c, fasting blood glucose and 2-hour postprandial glucose following a standard dinner as well as systolic and diastolic blood pressure. RESULTS: There were no differences in HbA1 [(6.0 +/- 0.8)% vs (6.2 +/- 0.8)%, P > 0.05], fasting glucose [(7.0 +/- 1.8) mmol/L vs (7.0 +/- 1.9) mmol/L, P > 0.05], 2-hour postprandial blood glucose [(10.7 +/- 2.5) mmol/L vs (11.2 +/- 3.4) mmol/L, P > 0.05], systolic blood pressure [(147.3 +/- 9.7) mm Hg (1 mm Hg = 0.133 kPa) vs (146.2 +/- 8.3) mm Hg, P > 0.05] and diastolic blood pressure (88.3 +/- 8.9 mm Hg vs 87.8 +/- 7.9 mm Hg, P > 0.05) between the bisoprolol and captopril group before treatment. After administration of bisoprolol or captopril, there were still no differences between these two groups in HbA1c [(5.7 +/- 0.9)% vs (5.7 +/- 1.1)%, P > 0.05], fasting blood glucose [(6.8 +/- 1.6) mmol/L vs (6.4 +/- 2.1) mmol/L, P > 0.05], 2-hour postprandial glucose [(10.0 +/- 2.9) mmol/L vs (10.2 +/- +/- 2.9)mmol/L, P > 0.05], systolic blood pressure [(124.8 +/- 10.6) mm Hg vs (126.6 +/- 7.8) mm Hg, P > 0.05] and diastolic blood pressure [(74.5 +/- 7.7) mm Hg vs (77.6 +/- 7.6) mm Hg, P = 0.05]. CONCLUSIONS: Based on these results, bisoprolol appears to be a beta1-selective blocker possessing a satisfactory antihypertensive effect without any adverse effects on glucose metabolism and is therefore a choice for treating hypertensive patients with type 2 diabetes mellitus.

[Inhibitory effect of sodium butyrate on 1,2-dimethylhydrazine-induced tumorigenesis of colorectal cancer in mice].

BACKGROUND & OBJECTIVE: In vitro studies proved that sodium butyrate (NaB) could stimulate cell differentiation, inhibit cell proliferation, and induce cell apoptosis in various tumors. This study was to evaluate the preventive effects of coloclysis of NaB on 1,2-dimethylhydrazine (DMH)-induced tumorigenesis of colorectal cancer in mice. METHODS: The mice of Kunming species were divided into 5 groups and received relevant treatments: DMH alone group (with subcutaneous injection of 30 mg/kg of DMH weekly for 11 weeks), control (normal saline, NS) group, DMH plus low dose of NaB (1.25 mol/kg, 24-week coloclysis) group, DMH plus high dose of NaB (2.5 mol/kg, 24-week coloclysis) group, and high dose of NaB group. The mice were killed in batches at the 12th, 18th, and 24th weeks of carcinoma induction separately. The incidence of colorectal tumor in each group was observed. Meanwhile, the general condition, body weight increasing, liver and renal functions, and pathologic changes of liver, kidney, lungs, and pancreas of the mice in DMH plus high dose of NaB group were also observed. RESULTS: No tumor was observed at the 12th week in each group. At the 18th week, the tumor incidence was significantly higher in DMH alone group and DMH plus low dose of NaB group than in DMH plus high dose of NaB group (58.3%, and 25.0% vs. 0, P < 0.01). At the 24th week, the tumor incidence was 95.0% in DMH alone group, 45.0% in DMH plus low dose of NaB group, and 15.0% in DMH plus high dose of NaB group; the differences among the 3 groups were significant (P=0.01, P < 0.01). No tumor was observed in control group and high dose of NaB group. There was no difference in the general condition, body weight increasing, and liver and renal functions of the mice between control group and high dose of NaB group (P > 0.05); no pathologic changes in liver, kidney, lungs, and pancreas were observed in the mice in high dose of NaB group. CONCLUSION: NaB could inhibit DMH-induced tumorigenesis of colorectal cancer in mice with no obvious toxicity.
