Hedgehog signalling activity of Smoothened requires phosphorylation by protein kinase A and casein kinase I.

The Hedgehog (Hh) family of secreted proteins governs cell growth and patterning in animal development. The Hh signal is transduced by the seven-transmembrane protein Smoothened (Smo); however, the mechanism by which Smo is regulated remains largely unknown. Here we show that protein kinase A (PKA) and casein kinase I (CKI) regulate Smo cell-surface accumulation and activity in response to Hh. Blocking PKA or CKI activity in the Drosophila wing disc prevents Hh-induced Smo accumulation and attenuates pathway activity, whereas increasing PKA activity promotes Smo accumulation and pathway activation. We show that PKA and CKI phosphorylate Smo at several sites, and that phosphorylation-deficient forms of Smo fail to accumulate on the cell surface and are unable to transduce the Hh signal. Conversely, phosphorylation-mimicking Smo variants show constitutive cell-surface expression and signalling activity. Furthermore, we find that the levels of Smo cell-surface expression and activity correlate with its levels of phosphorylation. Our data indicate that Hh induces progressive Smo phosphorylation by PKA and CKI, leading to elevation of Smo cell-surface levels and signalling activity.

Cationic compounds used in lipoplexes and polyplexes for gene delivery.

Gene transfer represents an important advance in the treatment of both genetic and acquired diseases. Many cationic lipids and cationic polymers naturally occurred or synthesized have been used for gene transfer. They have the advantages over viral gene transfer as non-immunogenic, easy to produce and not oncogenic. These cationic compounds, however, have the major limitations of inefficient transfection and toxicity to cells. For overcoming these problems, many new cationic compounds were developed since the first cationic lipid, DOTMA, was found usage in gene therapy. This article reviews cationic lipids for gene therapy from chemistry viewpoint and we classify these compounds as monovalent cationic lipids, polyvalent cationic lipids, cationic polymers, guanidine containing compounds, cationic peptides and cholesterol containing compounds, and hope to provide suggestions on the development of this variety of cationic compounds through the discussion.

A role for 14-3-3 tau in E2F1 stabilization and DNA damage-induced apoptosis.

Genotoxic stress triggers apoptosis through multiple signaling pathways. Recent studies have demonstrated a specific induction of E2F1 accumulation and a role for E2F1 in apoptosis upon DNA damage. Induction of E2F1 is mediated by phosphorylation events that are dependent on DNA damage-responsive protein kinases, such as ATM. How ATM phosphorylation leads to E2F1 stabilization is unknown. We now show that 14-3-3 tau, a phosphoserine-binding protein, mediates E2F1 stabilization. 14-3-3 tau interacts with ATM-phosphorylated E2F1 during DNA damage and inhibits E2F1 ubiquitination. Depletion of 14-3-3 tau or E2F1, but not E2F2 or E2F3, blocks adriamycin-induced apoptosis. 14-3-3 tau is also required for expression and induction of E2F1 apoptotic targets, such as p73, Apaf-1, and caspases, during DNA damage. Together, these data demonstrate a novel function for 14-3-3 tau in the regulation of E2F1 protein stability and apoptosis during DNA damage.

Pose scoring by NMR.

Recently, we have developed a fast approach to calculate NMR chemical shifts using the divide and conquer method at the semiempirical level. To demonstrate the utility of this approach for characterizing protein-ligand interactions, we used the deviation of calculated chemical shift perturbations from experiment to determine the orientation of a ligand (GPI-1046) in the binding pocket of the FK506 binding protein (FKBP12). Moreover, we were able to select the native state of the ligand from a collection of decoy poses. A key hydrogen bond between O1 and HN in Ile56 was also identified. Our results suggest that ligand-induced chemical shift perturbations can be used to refine protein/ligand structures.

Surface plasmon polariton propagation in nanoscale metal gap waveguides.

Based on finite-difference time-domain simulation of the propagation characteristics of surface plasmon polaritons (SPPs) in optical circuits made from metal gap waveguides (MGWs) with nanometric gap widths, we theoretically demonstrate that two structures that consist of splitting and recombining MGWs and of coupling MGWs can be used as nanoscale Mach-Zehnder interferometers. MGW arrays show capabilities for array imaging and for controlling the flow of SPPs. Other potential applications of coupling MGWs, as SPP switches, directional couplers, and even as a nanoscale counterpart for observing linear and nonlinear dynamic behavior of electromagnetic fields, are also predicted and discussed. Our results point to an interesting way to manipulate optical signals and provide efficient sensing in nanophotonic architectures.

[Association between serotonin 2C gene polymorphisms and attention deficit hyperactivity disorder comorbid or not comorbid with learning disorder].

OBJECTIVE: To investigate the relationship between two HTR2C gene polymorphisms, that is C-759T and G-697C polymorphisms, and attention deficit hyperactivity disorder (ADHD) comorbid or not comorbid learning disorder (LD). METHODS: Blood samples were taken from 189 trios with probands of ADHD comorbid LD (ADHD+LD) and 299 trios with probands of ADHD not comorbid LD (ADHD-LD). DNA was extracted and PCR was performed to amplify the fragments containing both C-759T and G-697C polymorphisms. Aci I was used to detect different alleles of the two polymorphisms. Allele- based and haplotype- based TDT analysis were used to test the association of the two polymorphisms of HTR2C gene and ADHD-LD and ADHD+LD. RESULTS: -759C(chi(2)=6.961, P=0.008), -697G(chi(2)=8.346, P=0.004), as well as -759C/-697G haplotype were over- transmitted(chi(2)=9.000, P=0.002 7), while haplotype -759T/-697C was under- transmitted(chi(2)=7.784, P=0.005 3) to probands with ADHD-LD. No biased transmission of any allele and haplotype were found in families with probands of ADHD+LD. CONCLUSION: ADHD-LD and ADHD+LD are different at the level of HTR2C gene polymrohisms of C-759T and G-697C. HTR2C is related to ADHD-LD, while not related to ADHD+LD.

Attention Deficit Disorder with Hyperactivity↗

[A preliminary study on the attentional networks of attention deficit hyperactivity disorder].

OBJECTIVE: To investigate the efficiency of the three attentional networks in attention deficit hyperactivity disorder (ADHD). METHODS: Subjects were 25 children, aged 7 to 12 years, with DSM-IV ADHD and 25 non-ADHD controls matched by age, sex and IQ. Attentional networks tests were performed in all the subjects on an IBM compatible computer. RESULTS: No differences were found between the ADHD children and the normal controls in alerting and orienting effects. However, ADHD children had a significantly larger executive control effect than their normal counterparts. The general accuracy of the ADHD children was significantly lower than that of the normal controls. CONCLUSION: ADHD children may have impairments in their executive control network of attention.

Fast semiempirical calculations for nuclear magnetic resonance chemical shifts: a divide-and-conquer approach.

A new approach to calculate nuclear magnetic resonance chemical shifts has been implemented at the semiempirical modified neglect of diatomic overlap level using gauge-including atomic orbitals. The perturbed density matrix with respect to the magnetic field is obtained by the diagonalization of the complex Fock matrix using the divide and conquer (DC) method, instead of by solving the computationally expensive coupled perturbed Hartree-Fock equations. Adopting the Patchkovskii and Thiel parameters [S. Patchkovskii and W. Thiel J. Comput. Chem. 20, 1220 (1999)], we were able to reproduce their results for small organic molecules. The errors introduced by DC method are negligible, as shown by the calculations on a series of polyalaine structures. Test calculations on proteins have demonstrated that our approach makes it possible to calculate chemical shifts routinely on systems with hundreds of atoms with good accuracy.

[Association of dopamine beta-hydroxylase polymorphism with attention deficit hyperactivity disorder in children].

OBJECTIVE: To study the association of dopamine beta-hydroxylase gene 5' flanking region polymorphism-1021 C-->T with attention deficit hyperactivity disorder (ADHD) in Han children. METHODS: ADHD was diagnosed according to the DSM-IV criteria. DNA was extracted and PCR was performed to examine the DBH-1021C-->T polymorphism. HHRR was used to test the association of dopamine beta-hydroxylase gene with different subtypes of ADHD. RESULTS: HHRR results showed this polymorphism had a tendency to be associated with the inattention subtype (P=0.067) and the combined subtype (P=0.076). The T allele was the protective factor of the inattentive subtype (P=0.07), and the risk factor of the combined subtype (P=0.08). After dividing the 292 nuclear families according to sex, DBH-1021C-->T polymorphism was found to be associated with the combined subtype (P=0.04) with the T allele as the risk factor (P=0.02). There were no positive findings among the girl nuclear families of ADHD. CONCLUSION: DBH gene is related to ADHD combined subtype and inattention subtype. The genetic basis of ADHD combined subtype and is inattention is different, and is influenced by sex factor.

Macrophage migration inhibitory factor deficiency impairs atherosclerosis in low-density lipoprotein receptor-deficient mice.

BACKGROUND: Macrophage migration inhibitory factor (MIF) is a proinflammatory cytokine expressed widely by vascular cells. However, scant in vivo evidence supports direct participation of MIF in atherogenesis. Therefore, we investigated whether deficiency of MIF modulates atherosclerotic lesion formation and composition in low-density lipoprotein receptor-deficient (LDLr-/-) mice. METHODS AND RESULTS: MIF-/-LDLr-/- and LDLr-/- mice were generated and consumed an atherogenic diet for 12 or 26 weeks. MIF-/-LDLr-/- mice had significantly reduced abdominal aorta lipid deposition and intimal thickening from aortic arch throughout the abdominal aorta compared with LDLr-/- mice. Marked retardation of atherosclerosis over time in MIF-deficient mice accompanied decreased lesion cell proliferation. At 26 weeks, 20% of MIF-deficient mice developed only early, fatty streak-like lesions, whereas >80% of LDLr-/- mice developed advanced lesions containing calcification and lipid cores. Analysis of smooth muscle cells from mouse aortae demonstrated that MIF deficiency reduced smooth muscle cell proliferation, cysteine protease expression, and elastinolytic and collagenolytic activities. CONCLUSIONS: Deficiency of MIF reduces atherogenesis in LDLr-/- mice. These results provide novel insight into inflammatory pathways operating in atheromata and identify a new potential target for modulating atherogenesis.

Dielectric properties and frequency response of self-assembled monolayers of alkanethiols.

This paper presents dielectric properties of alkanethiol self-assembled monolayers (SAMs) under an ac electric field. Using a Hg-SAM/SAM-Hg junction, we measured the ac impedance of alkanethiol SAMs using a sinusoidal perturbation of 30 mV (peak-to-peak) with frequency ranging from 1 Hz to 1 MHz at zero bias. Semicircles at higher frequencies and at middle frequencies along with Warburg lines at lower frequencies were observed in complex plane impedance plots, that is, Nyquist plots. The frequency response of SAMs was analyzed by modeling the junction using an equivalent circuit and fitting the Nyquist plots. The semicircles at higher frequencies are attributed to the effect of the SAM/SAM interfaces, and the ones at middle frequencies are attributed to the effect of alkanethiol SAMs. The comparison in the plots of the imaginary part of the impedance Z against frequency for the bare Hg electrodes (in pure ethanal) and the SAM-covered Hg electrodes (in alkanethiol solution) supports the analysis. The Warburg lines are attributed to a certain ionic impurity. The dielectric loss spectra are further analyzed. Chain-length-dependent peaks, which correspond to different relaxation mechanisms, at higher frequencies and middle frequencies were observed in the spectra of the dissipation factor (tan delta vs frequency). The peaks move to small frequency with the increase of chain length of alkanethiols. Using a correlation of peak position with the chain length, we then derived active energies of 39-99 meV for alkanethiol SAMs of C7-C18 under an ac electric field.

Reaction of a terminal phosphinidene complex with azulenes: eta1-complexes, C--H bond insertions, and 1,4-adducts.

Reaction of an in situ generated phosphinidene complex [PhPW(CO)(5)] with the aromatic azulene and guaiazulene leads to unexpected 1,4-adducts of the seven-membered ring and to C--H bond insertion of the five-membered ring. A DFT analysis suggests that the reaction is initiated by formation of a eta(1)-complex between the phosphinidene and the five-membered ring of the aromatic substrate. Four conformations of this complex were identified. Two convert without barrier to the slightly more stable syn- and anti-1,2-adducts. These undergo pericyclic 1,7-sigmatropic rearrangements with remarkably low barriers to give 1,4-adducts, with an inverted configuration at the phosphorus center. An X-ray crystal structure is presented for one of the 1,4-adducts of guaiazulene. The other two eta(1)-complexes insert with modest barriers into a C--H bond of the five-membered ring.

Separation of quaternary ammonium diastereomeric oligomers by capillary electrophoresis.

The separation of novel diastereomeric trimers (3M) and pentamers (5M), derived from quaternary ammonium salts, was studied in conventional, uncoated and coated capillaries using capillary zone electrophoresis (CZE) with a variety of buffers and additives. Resolution of 5M diastereomers was best achieved using gamma-cyclodextrin (gamma-CD) as a chiral selector, while no diastereomeric resolution was realized for the 3M material.
