Protein-DNA interactions in the promoter region of the gene encoding diapause hormone and pheromone biosynthesis activating neuropeptide of the cotton bollworm, Helicoverpa armigera.

Diapause hormone (DH) and pheromone biosynthesis activating neuropeptide (PBAN) are two crucial neuropeptides which regulate insect development and sex pheromone biosynthesis respectively. These peptides are encoded by a single gene, termed DH-PBAN gene. In this study, we characterized the promoter of the DH-PBAN gene in Helicoverpa armigera (Har). Transient transfection assays using a series of stepwise deletion fragments linked to the luciferase reporter gene indicate that the promoter contains multiple regulator domains that can activate and repress reporter gene expression. The fragment spanning -467 to -371 bp of the DH-PBAN promoter is an activator domain of transcription, whereas the region from -965 to -534 bp represses the promoter activity in the insect cell line BmN. Electrophoretic mobility shift assays demonstrate that at least two nuclear protein factors from the nuclear protein extracts of H. armigera suboesophageal ganglion, Har-DHMBP-1 and-2 (DH-modulator-binding protein) can specifically bind to the activating region. Furthermore, we characterized in detail that the nuclear protein factor Har-DHMBP-3 can specifically bind to a classical E-box, CAGCTG localized at positions -360 to -355 bp, a potential site for interaction with basic helix-loop-helix transcription factors. Mutation of this E-box results in a significant reduction of the promoter activity, suggesting it can modulate the previously identified activator domain. Taken together, multipartite cis-elements and transcription factors in the DH-PBAN promoter are involved in regulation of the gene expression.

MP2 theory investigation on the halides of D6hC36:C36Xn (X=F,Cl,Br; n=2,4,6,12).

An investigation of C(36)X(n) (X=F,Cl,Br; n=2,4,6,12) formed from the initial C(36) fullerene with D(6h) symmetry has been performed using the MP2 theory. Their equilibrium structures, reaction energies, strain energies, lowest unoccupied molecular orbital-highest occupied molecular orbital (LUMO-HOMO) gap energies, and aromaticities have been studied. The calculation results showed that those addition reaction were highly exothermic and C(36)X(n) were more stable than C(36). Moreover, from the view of thermodynamics it should be possible to detect C(36)X(6). The LUMO-HOMO gap energies of C(36)X(n) were higher than D(6h)C(36) and the redox characteristics of C(36)X(n) were weaker comparing to D(6h)C(36). The analyses of pi-orbital axis vector indicated that the chemical reactivity of C(36) was the result of the high strain, and the nucleus independent chemical shifts research showed that the stabilities of the C(36)X(6) were correlative with the conjugation effect.

Heterologous expression of the AtDREB1A gene in chrysanthemum increases drought and salt stress tolerance.

DNA cassette containing an AtDREB1A cDNA and a nos terminator, driven by a cauliflower mosaic 35S promoter, or a stress-inducible rd29A promoter, was transformed into the ground cover chrysanthemum (Dendranthema grandiflorum) 'Fall Color' genome. Compared with wild type plants, severe growth retardation was observed in 35S:DREB1A plants, but not in rd29A:DREB1A plants. RT-PCR analysis revealed that, under stress conditions, the DREB1A gene was over-expressed constitutively in 35S:DREB1A plants, but was over-expressed inductively in rd29A:DREB1A plants. The transgenic plants exhibited tolerance to drought and salt stress, and the tolerance was significantly stronger in rd29A:DREB1A plants than in 35S:DREB1A plants. Proline content and SOD activity were increased inductively in rd29A:DREB1A plants than in 35S:DREB1A plants under stress conditions. These results indicate that heterologous AtDREB1A can confer drought and salt tolerance in transgenic chrysanthemum, and improvement of the stress tolerance may be related to enhancement of proline content and SOD activity.

A practical VEP-based brain-computer interface.

This paper introduces the development of a practical brain-computer interface at Tsinghua University. The system uses frequency-coded steady-state visual evoked potentials to determine the gaze direction of the user. To ensure more universal applicability of the system, approaches for reducing user variation on system performance have been proposed. The information transfer rate (ITR) has been evaluated both in the laboratory and at the Rehabilitation Center of China, respectively. The system has been proved to be applicable to > 90% of people with a high ITR in living environments.

[Neural engineering and neural prostheses].

The motivation of the brain-computer interface (BCI) research and its potential applications are introduced in this paper. Some of the problems in BCI-based medical device developments are also discussed.

The human Penumbra gene is mapped to a region on chromosome 7 frequently deleted in myeloid malignancies.

We previously cloned the murine Penumbra gene based on its differential expression in proerythroblasts/erythroblasts. Subsequently, we identified human Penumbra cDNA from a human bone marrow cDNA library and the human Penumbra gene from a BAC library. Penumbra is a new member of the tetraspanin protein family and exhibits growth-suppressive activity in vitro. In this study, we designed a human Penumbra probe contig and used fluorescent in situ hybridization (FISH) to analyze seven cases of myeloid malignancies with 7q deletions. Five patients with cytogenetic deletions involving 7q31.2 approximately q32 also showed deletions of Penumbra by FISH; these were not present in two patients with cytogenetic deletions not involving 7q31.2 approximately q32. Our findings provide the first FISH evidence supporting the mapping of human Penumbra to 7q31.2 approximately q32 and demonstrate the potential of the Penumbra probe in the detection of 7q31 approximately q32-related deletions in myeloid malignancies.

Characterization of papillary squamotransitional cell carcinoma of the cervix.

OBJECTIVE: Does papillary squamotransitional cell carcinoma (PSTCC) behave differently from conventional squamous cell carcinoma of the cervix and does PSTCC have true transitional cell differentiation? MATERIALS AND METHODS: Twenty cases of PSTCC were identified from archival files. Clinical data were compiled. Immunoperoxidase stains for uroplakin III, p63 and p16 were performed on available tissue blocks. RESULTS: Patients ranged in age from 27 to 85 years. Twelve patients were FIGO Stage I, 4 were Stage II, and 2 were Stage III. Thirteen patients had clinical follow-up ranging from 5 to 132 months. Three patients subsequently had more extensive disease than initial clinical staging indicated. Nine patients had no tumor progression, three had local recurrence and one had metastatic disease. Eight cases were strongly immunoreactive for p63 and p16 and 14 were negative for uroplakin III. CONCLUSIONS: PSTCC lacks true transitional cell differentiation and probably shares similar clinicopathologic features with conventional cervical squamous cell carcinoma.

Suppression of invasion in human U87 glioma cells by adenovirus-mediated co-transfer of TIMP-2 and PTEN gene.

TIMPs and PTEN are known to be inhibitors of the invasive activities of malignant glioma. But there has been no literature reported concerning the effect of combined gene transfer of these two genes on invasiveness of glioma. This study was designed to evaluate the effect of adenovirus-mediated in vitro gene transfer of tissue inhibitor of metalloproteinases-2 (TIMP-2) and phosphatase and tensin homology deleted on chromosome ten (PTEN) on invasion of human U87 glioma cells. The mRNA and protein expressions of TIMP-2 and PTEN in U87 cells infected with AdTIMP-2 and AdPTEN were determined by RT-PCR and Western blot, respectively. The relative activity of MMP-2 and MMP-9 were measured by Gelatin zymogram and invasion of U87 in vitro were detected using Boyden chamber. The number of invasion cell of U87, U87 infected with Ad-gal, AdPTEN, AdTIMP-2 and AdPTEN/TIMP-2 was 55.63+/-13.27, 48.27+/-14.75, 35.27+/-10.94, 27.37+/-12.81, and 19.17+/-5.45, respectively. In vitro invasiveness of glioma cells was significantly inhibited by infection with AdTIMP-2 and/or AdPTEN, which was not consistent with the change of MMPs activity. And in the combinated group, the inhibition effect was more remarkable than in single group. Our studies suggest that adenovirus-mediated combined TIMP-2 and PTEN gene therapy is possibly useful for anti-invasion therapy of malignant glioma.

[Safety and short-term results of stent-assisted angioplasty for the treatment of intracranial arterial stenosis].

OBJECTIVE: To evaluate the safety and short-term outcome of endovascular stent-assisted angioplasty for the treatment of intracranial arterial stenosis. METHODS: Angioplasty and stent placement were administered to treat 46 patients with lesions of atherosclerotic intracranial stenosis, including 16 lesions of basilar artery, 12 of vertebral artery, 13 of internal carotid artery and 9 of middle cerebral artery. RESULTS: Technical success was achieved in 49 of 50 vessels (98%), with no procedure-related death or cerebral ischemic attack. Extracranial internal carotid artery dissection occurred in 1 patient and was successfully treated with a self-expand stent. One patient had subarachnoid hemorrhage because of perforation by microwire with no permanent neurologic deficit, and another 2 patients had groin hematoma. Angiographic examination immediately after stenting revealed that the stenosis rate was significantly reduced (72.4% +/- 12.3% vs 10.6% +/- 7.8%). There was no cerebral ischemic attack in the 37 patients who were clinically followed up (6 - 18 months, mean of 8.5 months). CONCLUSION: Angioplasty and stenting for the treatment of intracranial stenosis is safe and feasible, and it may be favorable for decreasing the incidence of ischemic attack.

Molecular cytogenetic characterization of a case of Müllerian adenosarcoma.

Müllerian adenosarcoma is a distinctive type of mixed Müllerian tumor of the female genital tract. To our knowledge, no cytogenetic data have been documented on Müllerian adenosarcoma in the literature so far. We report here the chromosomal findings of a Müllerian adenosarcoma in a 15-year-old female. Cytogenetic and molecular cytogenetic analysis revealed a complex karyotype involving chromosomes 2, 8, 10, 13, 19, and 21. These numerical and structural abnormalities may be of etiologic significance. This report may highlight the potential value of molecular cytogenetic analysis in differential diagnosis of Müllerian tumors. More cases are warranted to further genetically characterize this type of neoplasm.

BCI Competition 2003--Data set IIb: enhancing P300 wave detection using ICA-based subspace projections for BCI applications.

An algorithm based on independent component analysis (ICA) is introduced for P300 detection. After ICA decomposition, P300-related independent components are selected according to the a priori knowledge of P300 spatio-temporal pattern, and clear P300 peak is reconstructed by back projection of ICA. Applied to the dataset IIb of BCI Competition 2003, the algorithm achieved an accuracy of 100% in P300 detection within five repetitions.

Combined stent and coil in endovascular treatment of intracranial wide-necked and fusiform aneurysms.

BACKGROUND: Wide-necked and fusiform aneurysms are challenges for interventional neuroradiologists and neurosurgeons since they may involve entire vessels or be irregular in shape, making is possible for Guglielmi detachable coil to protrude into the parent arteries. This study was to evaluate the endovascular treatment of intracranial wide-necked and fusiform aneurysms by using a combination of stents and coils. METHODS: From October 2000 to July 2001, eighteen patients with intracranial wide-necked or fusiform aneurysms were treated with intraluminal stenting in combination with endosaccular coiling. RESULTS: Stent placement and coil packing were technically successful in all patients. There were no complications related to the procedure except for the migration of the stent in four cases. The aneurysms were embolized with the coils after precise stenting, and total occlusion was obtained in 13 cases, neck remnants in 3, and loose occlusion in 2. All the patients recovered well. Short-term follow-up angiograms suggested patency of the parent arteries without recanalization. CONCLUSIONS: A combination of endovascular stenting and coil packing may be a favorable alternative for the treatment of intracranial aneurysms that are unsuitable for surgical clipping or coil embolization. Further study is necessary to assess the long-term outcome.

[Bone segmentation in human CT images].

In 3D visualization of human skeleton, distinguishing bones from soft tissue in 2D CT slides is the first and most critical procedure. This article presents the methods for image pre-processing, segmentation and smoothing. 1733 CT images of human body from Visible Human Project provided by the American National Library of Medicine are treated in this paper. We use the technique of Chebyshev uniform approximation filtering for denoising and present a new simple adaptive threshold method in segmentation, which combines the similarity of consecutive slices with the region-growing method. In post-processing, we use the algorithms of mathematical morphology and multi-resolution filtering. The accuracy of segmentation is examined and certified by comparing the segmented images with the original one. The results also demonstrate a wide applicability of the method.

Molecular cytogenetic analysis of a pleuropulmonary blastoma.

We report a case of pleuropulmonary blastoma with complex cytogenetic abnormalities, including trisomy 2, trisomy 8, dup(7), der(10) t(8; 10)(q13; q22), add(17), and double minutes (dmin). Fluorescence in situ hybridization FISH analysis demonstrated TP53 deletion and amplification of MYCN; the latter has not been reported in PPB.

[Endovascular stent-assisted angioplasty for atherosclerotic intracranial stenosis, a clinical analysis of 39 cases].

OBJECTIVE: To assess the preliminary outcome of endovascular stent-assisted angioplasty for intracranial stenosis. METHODS: Thirty-nine patients with atherosclerotic intracranial stenosis (19 cases in posterior circulation and 20 in anterior circulation) were treated with endovascular angioplasty using balloon-expandable coronary stents. Follow-up was made for 4 - 24 months. The clinical manifestations were observed and CT, MR, or DSA were conducted before and after the operation. RESULTS: All of the stents were successfully implanted on the first procedure without procedural and periprocedural complications. The mean carotid artery stenosis rate was 73.5 +/- 6.7% before the procedure and 11.2 +/- 4.3% after the procedure. The patients were clinical asymptomatic and neurologically intact. Follow-up angiographies made in 29 patients revealed no restenosis. CONCLUSION: With effective short-term results and helpful in decreasing incidence of stroke, endovascular stenting is safe and feasible for treatment of intracranial stenosis.

[A serial study of in cytogenetic change and morphology on the tetra-arsenic tetra-sulfide treatment in untreated or recurrent acute promyelocytic leukemia].

OBJECTIVE: To study the morphologic and cytogenetic change in acute promyelocytic leukemia (APL) patients during the tetra-arsenic tetra-sulfide (TATS) treatment and the mechanism of TATS. METHODS: The bone marrow cells of 13 newly diagnosed and 7 recurrent APL patients were studied through FISH and morphology during the TATS treatment by special and repeated marrow culture. RESULTS: Cytomorphological study of 8 cases (6 untreated and 2 recurrent) showed that TATS could differentiate APL cells forward to mature granulocytes. There was obvious correlation between the reduced t (15; 17) positive cells and the reduced APL cells (r range 0.7298 - 0.9989). Except one patient with t (11; 17), 19 (13 untreated and 7 recurrent) with translocation t (15; 17) achieved clinical CR including 16 who achieved cytogenetic CR. CONCLUSION: TATS has a differentiation-inducing effect on untreated and recurrent APL, with haematological CR and cytogenetic CR achieved. Measurement of t (15; 17) positive cells by FISH technique can reflect the change of APL cells objectively.

[Factors influencing phosphorus loss by runoff process from farmlands in the dianchi watershed].

By laboratory simulated experiment, the effects of soil factors on the farmland phosphorus losses and its concentration were studied under artificial rainfall and nature rainfall. A number of soil and landscape factors were taken into account such as slopeness, soil and plant covers, fertilizers and application performances as well as rainfall intensity. The study showed that the concentration of total phosphorus (TP) and the loss rate varied with slopeness, being highest at slopness of 18 degrees with the values of 0.138 mg.L-1 and 6.14 kg.hm-2. There was no significant difference in TP concentration and the loss rate between under artificial rainfall and under nature rainfall. Moreover, TP concentration in runoff was smaller with the courser soil surface texture, and obviously lower with crop covering than without covering, among which the loss decreased 0.9-2.8 kg.hm-2 at different level of slope. Fertilizer application remarkably enchanced TP concentration and, thus, total phosphorus loss by runoff.

[Ecological characteristics of zooplankton in frequent HAB areas of the East China Sea in spring].

A comprehensive oceanographic survey was made in the East China Sea (122 degrees-123 degrees 30'E, 29 degrees-32 degrees N) during April to May, 2002, and the zooplankton samples were collected by zooplankton net (mesh size 0.567 mm). Analyses on the ecological characteristics of zooplankton such as its quantity, community characteristics, species composition and dominant species showed that 128 species of diet zooplankton were identified (16 pelagic larvae and fish juveniles were not involved). Among them, copepods occupied dominant position, including 40 species, composing 31.25% of diet zooplankton. The community was divided into 3 ecological types, in which, eurythermy and euryhalinity occupied dominant position. Key species, Calanus sinicus, was the first dominant species (142.10 ind.m-3, composing 68.09% of diet zooplankton abundance). The average biomass of zooplankton was 243.80 mg.m-3 (55.53-773.92 mg.m-3), and the highest density (> 500 mg.m-3) was found in the estuary area of the Yangtze River (30 degrees 45'-31 degrees 15'N, 122 degrees 45'-123 degrees 15'E). The average biomass of diet zooplankton was 195.96 mg.m-3 (55.53-496.09 mg.m-3). It was also showed that the biomass value of the estuary area of the Yangtze River (the north of 30 degrees 30'N) was higher than that of the southwest of Zhoushan waters, and that of the offshore of the estuary area of the Yangtze River (122 degrees 45'E) was higher than that of coastal waters. The mean value of diversity index (H') was 2.12 (1.09-3.73), and the stations having low H' (< 2) occupied 37.03%. The index of diversity, evenness and richness in the estuary area of the Yangtze River were low, but dominance was high. These results showed that the community structure of zooplankton in studied areas was not stable. Regression analysis results showed that main factors in relation to the characteristic distribution of zooplankton were temperature, diatom abundance, and dinoflagellate abundance.
