Cytotoxic labdane diterpenoids from Croton oblongifolius.

Three labdane diterpenoids, 2-acetoxy-3-hydroxy-labda-8(17),12(E)-14-triene, 3-acetoxy-2-hydroxy-labda-8(17),12(E)-14-triene, and 2,3-dihydroxy-labda-8(17),12(E),14-triene were isolated from stem bark of Croton oblongifolius. Their structures were established by spectroscopic data, and each was also tested for cytotoxicity against various human tumor cell lines. The latter compound showed non-specific, moderate, cytotoxicities against all the cell lines; whereas the first two compounds were less active.

Oxidized heptenes from flowers of Melodorum fruticosum.

Three oxidized heptenes, 7-benzoyloxy-6-oxo-2,4Z-heptadiene-1,4-olide, [7-benzoyloxy-4-hydroxy-1-methoxy-2E,4Z-heptadiene-1,6-dione] and 7-benzoyloxy-6-oxo-2,4E-heptadiene-1,4-olide, were isolated from flowers of Melodorum fruticosum. Their structures were determined by spectral data analysis. The first two compounds exhibited moderate cytotoxicity against a panel of human tumor cell lines, whereas the third was inactive.

Cassane diterpenoid from Caesalpinia major.

A new cassane diterpenoid, 14-deoxy-epsilon-caesalpin was isolated from the seed kernels of Caesalpinia major and its structure was determined by spectroscopic data.

Identification of deoxymiroestrol as the actual rejuvenating principle of "Kwao Keur", Pueraria mirifica. The known miroestrol may be an artifact.

Miroestrol (1) has been isolated previously as an active principle from "Kwao Keur" (Pueraria mirifica), a rejuvenating folk medicine from Thailand. Reinvestigation using bioassay-guided purification has resulted in the isolation of a new potent phytoestrogen, deoxymiroestrol (2). The facile aerial oxidation of 2 into 1 suggests the possibility that 1 may be an artifact.

Isoflavonoids from Pueraria mirifica and their estrogenic activity.

Nine isoflavonoids including a new pterocarpene, puemiricarpene, were isolated from the tuberous root of Pueraria mirifica (Leguminosae). The structure of puemiricarpene was determined by spectroscopic means. Estrogenic activity of the isolated isoflavonoids was tested using MCF-7 human breast cancer cells. Moderate activity was observed for kwakhurin, a prenylated isoflavonoid.

Norpregnane glycosides from a Thai soft coral, Scleronephthya pallida.

Two new norpregnane glycosides, 19-norpregna-1,3,5(10), 20-tetraen-3-O-alpha-fucopyranoside (3) and 19-norpregna-1,3,5(10), 20-tetraen-3-O-beta-arabinopyranoside (4), were isolated from the soft coral Scleronephthya pallida, along with two known steroids, pregna-1,20-dien-3-one (1) and 19-norpregna-1,3,5(10), 20-tetraen-3-ol (2). 19-Norpregna-1,3,5(10), 20-tetraen-3-O-alpha-fucopyranoside (3) exhibits moderate antimalarial and cytotoxic activities. The chemical structures of 1-4 were elucidated from spectroscopic data.

Neocrotocembranal from Croton oblongifolius.

A new cembranoid diterpene, neocrotocembranal (3), was isolated from the stem bark of Croton oblongifolius. Its structure was established on the basis of spectroscopic analysis. This compound inhibited platelet aggregation induced by thrombin, with an IC50 value of 47.21 microg/mL, and exhibited cytotoxicity against P-388 cells in vitro, with an IC(50) value of 6.48 microg/mL.

Additional bioactive heptenes from Melodorum fruticosum.

Four additional new bioactive heptenes, melodorinol [2], homomelodienone [4], 7-hydroxy-6-hydromelodienone [5], and homoisomelodienone [7] have been isolated from Melodorum fruticosum. These compounds were slightly to significantly cytotoxic to human tumor cell lines. Their structures have been determined by comparison of their 1H-nmr, 13C-nmr, and mass spectral data with those of prototype compounds (acetylmelodorinol [1], melodienone 3, and isomelodienone [6]).
