[Electron microscopic findings during hypothermic storage. Preservation of the liver. I. (author's transl)].

Porcine livers were preserved by simple hypothermic storage for 6 h using plasma-protein-fraction 5%, albumin 5%, Collins C2, and Ringer-lactate. Electron-microscopic alterations primarily consist of aggregations of nuclear chromatin, marginal chromatin condensation, swelling and destruction of cristae mitochondriales, swelling and degranulation of the endoplasmic reticulum. These signs of cell damage are of varying intensity depending on the different preservation methods. The best condition of hepatocytes on the whole is guaranteed by using Collins C2-solution.

[Whipple disease of the CNS].

By the example of two cases that could only be cleared up by a postmortem examination the affection of the central nervous system by Whipple's disease is dealt with. The granulomatous inflammatory process which is almost exclusively localised in the grisea and shows a particular concentration in the diencephalon, in the cortex of the frontal, temporal and islet regions as well as in the deeper brain stem, had resulted in a variety of neurological-psychiatric symptoms, in most cases a systemic psychosyndrome, myoclonism and paresis of ocular movement. In both cases the diagnoses was verified by the electromicroscopic identification of the bacteria in the typical SPC cells.

Reactivity in the MEM-test of cancer patients' lymphocytes incubated with brain-derived preparations and extracts from malignant teratomas.

A batch of human encephalitogenic protein (HEP) was compared to preparations of microtubulin and associated proteins (MST) from guinea pig brain by serological techniques and the macrophage electrophoretic mobility (MEM) method. HEP consisted mainly of A1 protein and MST showed the characteristic double band in the 55,000 MW region and some additional weak protein bands. No cross-reactivity could be detected between HEP and MST by rabbit hyperimmune sera to these substances. The MEM test, however, revealed human lymphocyte reactivity to HEP and MST in both cancer and multiple sclerosis patients, but no remarkable responses in other patients. A synthetic nonapeptide (114-122 region of A1 protein) led to lymphokine release in 2 of 5 multiple sclerosis patients, in 1 of 13 cancer patients, and in none out of 13 control individuals. From these findings it is concluded that a minor component (contaminant?) present in both HEP and MST might induce lymphocyte responses in cancer patients. The A1 protein of HEP preparations is, if at all, not the only protein responsible for the so-called HEP-response of cancer patients. Continuing the MEM test experiments with extracts from single organ cancers, it could be shown that patients with malignant teratomas respond to a more or less broad spectrum of extracts bearing organ cancer specificity. On the other side, extracts made from 2 malignant teratoma tissues led to MEM responses in 8 of 13 cancers, in 2 of 11 control persons and in none of 5 multiple sclerosis patients. These findings underline the concept that tumour-associated antigens (possibly oncofetal antigens) do exist, which bear organ cancer specificity and will lead to a specific sensitization of the host. Such antigens may be expressed in teratomas in different qualities and amounts depending on the tissue differentiation capacity of a given teratoma.

Image processing in pathology. X. Electron microscopic morphometric analysis of human lymphocyte subpopulations.

In the present paper, methods of automatic image processing were used for a fine structural differentiation of lymphocyte subpopulations in purified non-labelled lymphocyte suspensions from peripheral blood. It was found that lymphocyte subpopulations differ in several morphological features which have been generated from the whole cell as well as from the nucleus. In general, these differences are greater than those attributed to individual deviations as well as as to influences from the preparation. As found by immunological methods, the composition of morphologically determined subpopulations corresponds to T- and non- T-cells. A preliminary functional interpretation fo the morphological differences is discussed.

Effects of p-chlorophenoxyisobutyric acid (CPIB) on the human liver.

Serial liver biopsies were carried out in 67 patients with HLP and/or fatty liver before, during short- and long-term therapy with CPIB and after termination of therapy. Results (1) Decrease of liver glycogen from 4.17% to 2.69% (wet weight, P less than 0.02). (2) Insignificant changes of liver triglyceride content. (3) Significant decrease of manganese, while the concentrations of zinc and copper in the liver biopsy specimens remained unchanged. (4) No signs of liver intoxication or cancerogeneous effects of light-microscopic pictures. (5) Significant increases in numbers of mitochondria and cristae as well as a hypertrophy of endoplasmic reticulum with longer lasting therapy. (6) Striking focal proliferation of cristae mitochondriales in 3 cases on longterm treatment. (7) Regression of the mitochondrial alterations after termination of the CPIB therapy. Our findings suggest that an increased number of mitochondria and of their inner membranes in the liver cells induced by CPIB could play an important role in the hypolipidemic action of the drug.

Detection of type D retrovirus in a human amnion cell line (FL).

This paper describes the detection of viral particles in a human amnion cell line (FL). These particles belong to the group of type D retroviruses, because of their characteristic morphology, their major antigenic determinants, and the presence of particle-associated Mg++-preferring RNA-dependent DNA polymerase. In addition to budding and mature type D virus particles, intracytoplasmic type A structures have also been found. Immunological analysis provided evidence of cross-reactivity between the particles described and the Mason-Pfizer monkey virus as well as type D retroviruses from other human cell lines (PMF, HEp-2). Particles isolated from FL cell medium were shown to infect type D virus-susceptible human TU 197 cells.

[Demonstration of hepatitis A virus in the feces].

On the basis of own examinations methodology and results of the demonstration of the virus A in the stools and of the proof of antibodies in the serum are described. The parameters of the virus-A-hepatitis which are important for practice are further the basis for diagnosis, therapy and surrounding prophylaxis in routine work. They were compiled in a table.

[A case of malacoplakia of the prostate (author's transl)].

A case of malacoplakia of the prostate gland in a 75-year-old man is reported. This special type of granulomatous prostatitis was diagnosed by histological examination of biopsyn material. There were typical Michaelis-Gutmann corpusculates within the inflammation area. The structure of these inclusion bodies was investigated by electron microscopy. Characteristic concentric ring-shaped structures were seen as well as corpusculates which are composed of irregular cristalloid material. Some Michaelis-Gutmann bodies were found to contain short cell membrane-like structures. By histochemical investigation, both the cells possessing inclusion bodies as well as the Michaelis-Gutmann corpusculates themselves gave positive acid phosphatase reactions. The case is described because of the rarity of the malacoplakia of the prostate gland and with respect to the differential diagnosis of malignant prostate gland disease.

Demonstration of mouse mammary tumour virus (MuMTV) antigenicity in human milk by means of immunodiffusion technique.

Using the micro-Ouchterlony immunodiffusion technique, we were able to demonstrate mouse mammary tumour virus (MuMTV) antigenicity in the 1.26-1.28 g/ml density fraction prepared from pooled human milk samples after treatment with detergents and ether. The cross-reacting antigen(s) was precipitated by rabbit antisera to MuMTV-B particles prepared from murine milk and intracytoplasmic A particles (iAp) isolated from mouse mammary tumour tissue. By confluence of precipitin lines, the human milk "core" antigen(s) was shown to be identical with the main antigen(s) of iAp which are known to share antigenicity with the MuMTV-B particle cores. Electron microscopy of the human "core" fraction revealed abundant particulate structures of 40-70 nm in diameter. However, the structural entities bearing the cross-reactive antigen(s) remain to be identified.

Image processing in pathology. IV. Internal structure of mitochondria.

For extensive recording of mitochondrial membrane lengths hitherto statistical methods were applied which only provide mean values. Conventional measurements of the lengths by means of curvimetry or by intersection counts are required for estimating size distributions and secondary parameters. The former method requires expenditure in time, the latter yields only very rough values for single organelles. Automatic image processing is a possible way to obtain more informations. A method for an automatic definition of the internal mitochondria structure is given in this paper and preliminary results are discussed.
