A simple two-step method for purification of secretory IgA from human colostrum.

This paper describes an extremely simple method for the purification of secretory IgA in two steps from a colostrum pool. The first step of this technique utilizes the agar transparency effect produced by high concentrations of secretory IgA contained in colostrum pool when it is subjected to electrophoresis in this support. The secretory IgA is obtained by extraction and elution of the transparent zone adjacent to the well. In the second step, in order to obtain the maximum degree of purity, the eluate is subjected to precipitation with ammonium sulfate (1.33 M) or chromatographed through Sephadex G-200 gel. The simplicity of the method and the purity of the protein obtained make this procedure suitable for purification of macromolecular substances in concentrated state.

DNA antibodies detected by microhemagglutination test in the diagnosis of systemic lupus erythematosus.

A method of tanned erythrocyte agglutination of wide applicability is described for the study of DNA antibodies. The practical aspects of the method have been explored in estimation of antibodies to DNA in human cases of systemic lupus erythematosus, discoid lupus erythematosus, rheumatoid arthritis, other collagenoses and not related diseases. This method was found to be of high sensitivity and specificity, detecting DNA antibodies only in sera of systemic lupus erythematosus. Results were compared with those obtained by immunofluorescence test using rat liver hepatocytes as source of nuclei.

[Effect of L-tetramisol associated with rifampicin in patients with lepromatous leprosy. L-tetramisol in patients with lepromatous leprosy].

In this work are presented results obtained in the treament of thirty patients suffering of lepromatous leprosy. In a group of fifteen patients L-tetramisol was administrated in association to antimocrobian drugs. The control group received only the last medication. Immunological modifications were not observed in any case. However, in six patients treated with L-tetramisol associated to rifampicin during three months it was possible to observe a notable improvement of the clinical state. In the patients that received L-tetramisol associated to other drugs or in those that received only antimicrobian drugs the clinical improvement was very low or null.

A comparative study on the development of rabbit male accessory gland organ-specific autoantigens.

Experimental cryosurgery of rabbit male accessory glands (RMAG) induced highly specific autoantibodies. They revealed in rabbit male accessory glands the presence of autoantigens when tested by passive cutaneous anaphylaxis reaction and passive haemagglutination test. Using the same techniques, the antisera were confronted with extracts of male accessory glands from rabbits of different ages. It was demonstrated that the autoantigens involved in both tests appear at about the age of three months. The significance of these findings in correlation with the initiation of the organ growth is discussed.

A simple method for detection of IgG rheumatoid factor.

A simple method for detection of IgG rheumatoid factor (RF) in sera and joint fluid is described. The technique is based on the action of 0.1 M 2-mercaptoethanol mixed directly with the material. After 2 h of incubation determinations of human anti-IgG were performed by latex agglutination test. Comparison with data obtained by using the conventional method, sequential 24 h treatment with 0.1 M 2-mercaptoethanol and 0.01 M iodoacetamide, shows similar results for both methods. A correlation was observed between the presence of IgGRF in synovial fluid and a severe clinical course with invalidating forms in patients suffering rheumatoid arthritis.

Potentiation of autoimmune response in rats infected with Toxoplasma gondii. Effect of the infection route.

The aim of this report is to describe the influence of the way of infection with 3 X 10(3) trophozoites of Toxoplasma gondii on the auto- and heteroimmune responses of rats immunized with chemically modified rat male accessory glands (MRAG) and human serum albumin (HSA) 6 and 36 days after infection. The delayed hypersensitivity (DTH) response to MRAG was potentiated in rats infected in thymus proximity (p less than 0.02) when compared with rats only immunized. The parasites introduced intraperitoneally did not modify the cellular autoimmune response. The titers of hemagglutinating antibodies to MRAG were increased in animals infected by both ways (p less than 0.05). The response to HSA did not show significant difference between the infected and uninfected rats. Thymic involution and proliferation of lymphocytes and plasma cells were observed at 6 and 52 days post-infection only in rats infected in thymus proximity (p less than 0.05). A correlation between the potentiation of cellular autoimmune response and the injection of parasites in thymus proximity was observed.
