Evidence for the importance of a positive charge and an alpha-helical structure of the C-terminus for biological activity of human IL-6.

Thirteen point mutations of human interleukin-6 at the C-terminus were constructed at the cDNA-level. Two degenerate oligonucleotide primers were used for PCR synthesis of two groups of point mutations at positions 182 and 184. The mutated cDNAs were in vitro transcribed and translated and subsequently assayed for biological activity in the B9 cell proliferation test. Our results confirm our former findings obtained with deletion mutants on the importance of the C-terminus of IL-6 for biological activity. In addition, we now present evidence for the importance of an alpha-helix at the C-terminus of IL-6 and the presence of the positive charge at position 182 for biological activity.

Saliva composition and caries development in asthmatic patients treated with beta 2-adrenoceptor agonists: a 4-year follow-up study.

In an earlier study, we found that chronic treatment with beta 2-adrenoceptor agonists in asthmatic subjects gave an impaired saliva secretion and a higher caries prevalence than in healthy controls. Twenty-one of the asthmatics and their matched controls were examined 4 yr later in a follow-up study. Samples of whole saliva stimulated by chewing and parotid saliva stimulated by citric acid were collected and dental caries was scored. In the asthmatic group the secretory rates of stimulated whole and parotid saliva decreased by 20% and 35%, respectively, compared to the control group. The number of lactobacilli increased. The asthmatic subjects had a decreased output per minute of total protein, amylase, hexosamine, salivary peroxidase, lysozyme, secretory IgA, a bacteria-aggregating glycoprotein, potassium, and calcium in stimulated parotid saliva. Initial and manifest caries lesions as well as the number of DFS were significantly increased in the asthma group. We conclude that asthmatic patients treated with beta 2-adrenoceptor agonists have an increased caries susceptibility due to an impaired saliva secretion caused by the use of beta-adrenergic agonists.

Correlation between dynamic posturography, clinical investigation, and neurography in patients with polyneuropathy.

Dynamic posturography (PG) is a new objective method to study functional performance in diseases affecting balance. It measures muscle response latencies and sway angles to standardized alterations of a moveable platform and moveable surroundings. Twenty-eight patients with polyneuropathy (PN) were studied by clinical investigation, vibrametry, neurography, and dynamic PG. The results of dynamic PG and vibrametry were compared with those of 29 healthy controls. In the patients with PN, clinical scores correlated to the latencies of the muscle response when the platform was suddenly moved forward, and to equilibrium performance (sway angles) in dynamic PG test conditions with eyes closed and the platform either stable or 'sway-referenced'. That is, the platform moves in response to the patient's anterior-posterior sway, creating a disturbed proprioceptive input to the brain. Clinical scores also correlated to the equilibrium performance when both platform and surroundings were sway-referenced. In conclusion dynamic PG, in addition to clinical investigations and neurophysiology, is a valuable and objective method for estimating the equilibrium performance in patients with PN.

Quantitative comparison of insulin-like growth factor mRNA levels in human and rat tissues analysed by a solution hybridization assay.

In this study a solution hybridization assay was evaluated for its application to the measurement of levels of specific mRNAs. The evaluation included parameters such as incubation time, hybridization stringency and probe concentration/structure. Both short (50 bases derived from synthetic oligonucleotides) and long (125-147 bases) RNA probes, derived from cloned sequences, could be used to obtain quantitative information on specific mRNA species. The solution hybridization assay was used to compare the levels of insulin-like growth factor-I (IGF-I) and IGF-II mRNAs in various rat and human tissues. In the rat the liver was the main source of IGF-I mRNA (approximately 400 molecules/cell), but significant levels were also found in extrahepatic tissues such as fat and muscle (3-50 molecules/cell). Human liver contained approximately 100-fold less IGF-I mRNA than rat liver. Human fat, muscle and placenta contained levels of IGF-I mRNA (2-8 molecules/cell) similar to those in the liver. Levels of IGF-II mRNA in rat and human tissues were similar, in that the expression was greatest in the placenta (approximately 200 molecules/cell). Species differences were evident, however, since human liver and fat contained significant amounts of IGF-II mRNA (15-20 molecules/cell), while the rat counterparts had almost undetectable levels. Young and old rats were used to examine the influence of age on the expression of IGF-I and GH receptor mRNAs in the liver. Levels of both IGF-I mRNA and GH receptor mRNA were found to decrease with age (2.8-fold and 1.7-fold respectively). It is concluded that low levels of IGF mRNAs can be detected using the solution hybridization assay and that there are considerable species differences within and between tissues with regard to steady-state levels of IGF-I and IGF-II mRNAs.

Dynamic posturography in cervical vertigo.

Cervical vertigo, the entity of neck disorder and associated vestibular symptoms, was investigated in 15 suspected subjects and results were compared with 15 age-matched controls. A modified dynamic posturography investigation with different head positions was used. Head position was recorded 3-dimensionally with electrogoniometry. Differences on a sway-referenced forceplate were found.

Structure-function analysis of human interleukin-6. Evidence for the involvement of the carboxy-terminus in function.

C-terminally deleted analogs of human interleukin-6 (IL-6) have been constructed at the cDNA level, and after cell-free transcription and translation their biological activity was analyzed. Removal of only 4 amino acids resulted in complete loss of biological activity as determined by the B9 cell proliferation assay. Secondary structure prediction of human IL-6 resulted in 58% helix, 14% beta-structure, and 28% turn and coil (average of 3 independent methods). The circular dichroism of recombinant human IL-6 was measured in the near and far UV. Evaluation of the latter in terms of secondary structures gave 67% helix, 15% beta-structure, and 18% turn and coil.

Saliva composition in asthmatic patients after treatment with two dose levels of a beta 2-adrenoceptor agonist.

Changes are known to occur in the salivary composition of asthmatic patients treated with beta 2-adrenoceptor agonists. To evaluate the precise contribution of the agonist to the impaired saliva secretion, 15 asthmatic patients, 15-23 yr old, were given two dose levels of agonist, either terbutaline or salbutamol. The lower dose, 0.15-3.0 mg/day, represented the therapeutic level used by the patients. During a wash-out period of one month, the asthma was treated with budesonide, a corticosteroid spray. Then a daily dose of 32 mg of terbutaline or salbutamol was given for one month. Samples of whole saliva, stimulated by chewing, and parotid saliva, stimulated by citric acid, were collected on three occasions: (1) at the end of the low-dose agonist treatment; (2) at the end of the wash-out period; and (3) at the end of the high-dose agonist treatment. During the high dosing the secretion rate of parotid saliva decreased and the concentrations of its total protein, amylase, hexosamine and the ratio of hexosamine/total protein were lowered. The output per minute of total protein, amylase, hexosamine, peroxidase, lysozyme, secretory IgA and potassium decreased. There were only small differences in secretion rates or saliva composition between samples collected at the end of the low-dose and at the end of the wash-out period. Thus, treatment with beta 2-adrenoceptor agonists impairs saliva secretion in asthmatics.

Localization of Usher syndrome type II to chromosome 1q.

Usher syndrome is characterized by congenital hearing loss, progressive visual impairment due to retinitis pigmentosa, and variable vestibular problems. The two subtypes of Usher syndrome, types I and II, can be distinguished by the degree of hearing loss and by the presence or absence of vestibular dysfunction. Type I is characterized by a profound hearing loss and totally absent vestibular responses, while type II has a milder hearing loss and normal vestibular function. Fifty-five members of eight type II Usher syndrome families were typed for three DNA markers in the distal region of chromosome 1q: D1S65 (pEKH7.4), REN (pHRnES1.9), and D1S81 (pTHH33). Statistically significant linkage was observed for Usher syndrome type II with a maximum multipoint lod score of 6.37 at the position of the marker THH33, thus localizing the Usher type II (USH2) gene to 1q. Nine families with type I Usher syndrome failed to show linkage to the same three markers. The statistical test for heterogeneity of linkage between Usher syndrome types I and II was highly significant, thus demonstrating that they are due to mutations at different genetic loci.

Gingival inflammatory reactions in children with rhinoconjunctivitis due to birch pollinosis.

The present investigation was designed to study the degree of gingival inflammation in children with rhinoconjunctivitis due to birch pollinosis. Thirty-four children, 8-17 yr of age, participated. As controls, healthy classmates of the same age and sex were chosen. Dental examination was performed on three occasions: pollen season I (Spring 1987), off-season (Autumn 1987) and pollen season II (Spring 1988). The controls were examined on one occasion, mixed with the allergic children at the off-season examination. The degree of gingival inflammation was studied by determining the gingival bleeding tendency by standardized probing. Absence or presence of dental plaque was recorded after sulcular probing. In order to compensate for differences in oral hygiene level between the participants, the bleeding/plaque ratio according to Abbas et al. was calculated. The comparisons of the bleeding/plaque ratios revealed statistically significantly higher mean ratios in the allergic children during pollen seasons compared with off-season and with the controls. The results indicate that during the pollen season, children with allergic rhinoconjunctivitis exhibit an enhanced degree of gingival inflammatory reaction.

Seasonal variation of the conjunctival provocation test, total and specific IgE in children with birch pollen allergy.

In 20 children with birch pollinosis, conjunctival provocation tests (CPT) with a birch pollen preparation were performed and blood was drawn on five different occasions during 1 year. From November to February there was a decrease in the activity of the atopic disease as measured by CPT (p = 0.03), total serum IGE (p = 0.02) and birch pollen-specific serum IgE (not significant). Then all parameters increased until August (p less than 0.05). From August to November there was a decrease, significant for total IgE and birch-specific IgE (p less than 0.01). The correlation between CPT and the in vitro tests was poor. The conclusion is that the seasonal variations of CPT, total and specific IgE are similar but without close correlation between CPT and the antibody levels. The priming during the season is retained at least for 1 year.

Otoneurological findings in workers exposed to styrene.

An otoneurological test battery was administered to 18 workers with long-term exposure (6-15 years) to styrene at levels well below the current Swedish limit (110 mg/m3). The results were compared with those of a reference group. Disturbances were found in the central auditory pathways of seven workers. Tests reflecting central processing of impulses from different sensory equilibrium organs were abnormal for 16 workers. The most relevant tests seemed to be static posturography and the rotatory visual suppression test. In the posturography the styrene group had a significantly larger sway area than the reference group. In the visual suppression test, the styrene workers displayed a significantly poorer ability to suppress vestibular nystagmus than the reference group. It was concluded that styrene exposure in industrial environments at moderate or low levels causes central nervous system disturbances which are not always diagnosable with psychometric tests but can be apparent in special otoneurological tests.

Visual suppression tests in acoustic neuroma patients.

Fifty-five patients with a unilateral acoustic neuroma were investigated preoperatively with visual suppression tests during rotatory oscillation and caloric irrigation. During a sinusoidal oscillation, 29% of the patients showed a reduced suppression compared to 9% during pseudo-random oscillation and 11% in the caloric test. In the sinusoidal and caloric tests the majority of the patients with deficient suppression had large or medium-sized tumors. In a few patients with small tumors, pathology was observed in both sinusoidal and randomized tests, presumably as a sign of unconcentration. The study shows that the sinusoidal visual suppression test and to a lesser degree suppression during caloric irrigation are of value for identification of brainstem-cerebellum compression of acoustic neuromas.

Effect of pollen immunotherapy on food hypersensitivity in children with birch pollinosis.

Of 72 children with severe rhinoconjunctivitis due to birch pollinosis, 67 (93%) reported sensitivity against nuts, apples, etc. The hayfever symptoms improved by immunotherapy but neither subcutaneous (n = 42) nor oral (n = 14) immunotherapy with birch pollen allergen preparations made the food sensitivity decrease significantly more than the placebo oral immunotherapy (n = 16).

cDNA cloning, sequence, and regulation of a major female-specific and growth hormone-inducible rat liver cytochrome P-450 active in 15 beta-hydroxylation of steroid sulfates.

cDNA clones encompassing the complete coding sequence for the female-specific rat liver microsomal 15 beta-hydroxylase have been isolated and sequenced. This cytochrome P-450 species (P-450 15 beta) has a total of 490 amino acid residues, and its deduced amino acid sequence as well as direct sequencing of the amino-terminal portion of the purified protein revealed its identity with P-450 i. P-450 15 beta was found to be 66% similar to the male-specific rat liver microsomal 16 alpha-hydroxylase, P-450 16 alpha. Furthermore, P-450 15 beta is 47% similar to the major phenobarbital-inducible cytochrome P-450 in rat liver, P-450 b, while its structural similarity to P-450 c, P-450 pregnenolone-16a-carbonitrile, and P-450 lauric acid omega-hydroxylase is less than 30%. However, the developmentally regulated cytochromes P-450 f and P-450 PB-1 both have 68% similarity to P-450 15 beta. It is therefore concluded that P-450 15 beta belongs to the same gene subfamily (P450IIC) that also includes cytochromes P-450 f, PB-1, and 16 alpha. The plasma pattern of growth hormone (GH) is known to be a major determinant for the sex-specific expression of P-450 15 beta and P-450 16 alpha. Continuous infusion of GH to hypophysectomized male rats causes an increase in P-450 15 beta mRNA after 6 days of treatment, while intermittent injections have no effect. On the other hand, no effect on P-450 16 alpha mRNA is observed after continuous administration of GH, while intermittent injections cause an increase after 2 days of treatment. This relatively long period before P-450 15 beta and P-450 16 alpha mRNA induction is seen might indicate that protein factors mediating GH action are involved.

Expression of insulin-like growth factor I (IGF-I) and IGF-II mRNA during hepatic development, proliferation and carcinogenesis in the rat.

Insulin-like growth factor I (IGF-I) and IGF-II are peptides that presumably are required for normal fetal and postpubertal growth. The production of IGFs is developmentally regulated and the liver appears to be a major site of production. By analysing mRNA levels for IGF-I and IGF-II in the rat liver we have attempted to further study the expression of these growth factors during development and regeneration as well as during the course of hepatic carcinogenesis. Fetal livers are characterized by a high level of IGF-II mRNA and a low level of IGF-I mRNA, while in adult livers the opposite situations occur, i.e. a high level of IGF-I mRNA and a non-measurable level of IGF-II mRNA. During the course of experimentally induced hepatic cancer, IGF-I mRNA was consistently reduced and in a majority of cancers analysed (6/9) IGF-II mRNA was increased, i.e. a fetal type of IGF expression can be switched on in some experimentally induced hepatocellular carcinomas. The onset of IGF-II production during hepatic carcinogenesis appears to be a late phenomenon since liver nodules, preceding the development of hepatocellular cancer, were found not to contain IGF-II mRNA. Furthermore, during hepatic regeneration following partial hepatectomy no marked change in IGF-I or IGF-II mRNA levels was noted. The above results suggest that the fetal growth factor IGF-II could have a role in hepatic cancer.

Regulation of insulin-like growth factor messenger ribonucleic acid in rat growth plate by growth hormone.

The mechanism of action for the stimulatory effect of GH on longitudinal bone growth is not yet clarified. Several recent reports indicate that GH has a direct effect at the site of the epiphyseal growth plate, as opposed to the somatomedin hypothesis which holds that the effect of GH is mediated by circulating insulin-like growth factors (IGFs). Using a RNA probe in a solution hybridization assay we investigated the presence of IGF-I mRNA in rat rib growth plate. Hypophysectomy resulted in a decrease in the number of IGF-I mRNA copies compared to that in normal rats. Replacement treatment with GH restored the number of transcripts in a specific and dose-dependent manner. The results show that GH regulates the level of IGF-I mRNA in rat rib growth plate and give further support to the hypothesis that locally produced IGF-I might contribute to the stimulatory effect of GH on longitudinal bone growth.
