[Cardiovascular action of 1,2,4-benzothiadiazine-1,1-dioxide derivatives. VI].

A series of 3,4-dihydro derivatives of 6-chloro-, 7-chloro-, 5,7-dichloro-, 6,7-dichloro-, 5,7-dibromo-, 5-nitro-7-chloro-, 7-nitro-, 7-amino-1,2,4-benzothiadiazine-1,1-dioxide, various substituted on the heterocyclic carbon, was prepared and tested for cardiovascular activity. It was found that in this series of compounds cardiac activity predominates and is exclusively of the depressant type. Bradycardial activity seems to be affected both by the nature of the substituent on the heterocyclic carbon atom and by the substituents on the benzene ring. An alkyl chain of three carbon atoms, normal or alpha-ethylsubstituted, on C3 and the presence of a chlorine atom in positions 6 or 7 seem to be the most significant structural features for this activity. Some of the substances tested showed a pressor effect (hypotension and/or increase in differential pressure).

[Cardiovascular action of 1,2,4-benzothiadiazine-1,1-dioxide derivatives. VII].

A series of dimethylamino-, diethylamino-, pyrrolidyl- and morpholylalkyl derivatives of 3-amino-1,2,4-benzothiadiazine-1,1-dioxide (compounds I leads to XX) was prepared and tested for cardiovascular activity. It was found that substitution of the alkyl group with the above mentioned groups in 3-amino-1,2,4-benzothiadiazine-1,1-dioxide derivatives in most cases causes marked dissociation of hypotensive activity from bradycardial activity and also disappearance or great reduction in the effect on differential pressure. It was also found that hypotensive activity is particularly dependent on the nature of the basic chain in the two components: length of the carbon chain between the two nitrogens and nature of the basic grouping. For the first component a chain of three carbon atoms seems the most effective and for the second the diethylamino group seems the most favourable. In addition, substituents in the benzene ring influence hypotensive activity, often positively, and 6,7-dichlorosubstitution in some compounds also affects the increase of differential pressure.

Anti-inflammatory activity of newly synthesized 2,6-bis-(1,1-dimethylethyl)phenol derivatives.

Four new 2,6-bis-(1,1-dimethylethyl)phenol derivatives were synthesized and evaluated for their anti-inflammatory activity, in comparison with indomethacin and vitamin E, using the carrageenan paw oedema assay in rats. Among the tested compounds, the 4-(6,7-dichloro-4H-1,2,4-benzothiadiazine-3-yl)-2,6-bis-(1,1- dimethylethyl)-phenol-S,S-dioxide (4) was the most active compound markedly reducing oedema formation. Moreover compound 4 potentiated, particularly in the early phase of carrageenan paw swelling (2-4 h), the antiphlogistic effect of indomethacin. This compound, on the other hand, did not show ulcerogenic activity at therapeutic doses and did not potentiate the gastric damage induced by high doses of indomethacin. In vitro, the studied compounds (1-4) showed to be moderately active as scavengers of 2,2-diphenyl-1-(2,4,6-trinitrophenyl)hydrazil, 4 being the most active among them. Compound 4, on the contrary, was practically inactive on liposome peroxidation where compounds 1, 2, 3, which are modest anti-inflammatory agents, exerted a good or moderate effect.

Evidence that exposure to methyl mercury during gestation induces behavioral and neurochemical changes in offspring of rats.

On day 15 of gestation, pregnant Sprague-Dawley rats were orally treated by gavage with 8 mg/kg of methyl mercury (MMC). At day 1 of postnatal life the levels of MMC in whole brain of exposed pups were found to be about 100 times higher than those of saline-exposed rats, while they were near to the control values at 21 days and practically normal at 60 days of age. Behavioral experiments showed that exposure to MMC in late gestation did not affect at any tested time (14, 21 and 60 days) locomotor activity or development of ultrasonic vocalization. An increased response to a challenge dose of amphetamine was, however, detected in MMC-exposed pups at day 14. This phenomenon was no longer evident at day 21 and 60 of age. In parallel, an increased density of dopamine receptors was found in the striatum at 14, but not at 21 and 60, days of age. From these data, we tentatively suggest that a high level of MMC induces a transient phenomenon of disuse-supersensitivity of the dopaminergic system. Moreover, further evidence that acute MMC exposure during prenatal life might induce permanent disturbances in learning and memory which could be partially related to a reduced functional activity of the glutamatergic system is provided.

Follow-up of methylmercury concentration in brain areas of developing rats exposed during prenatal life using cold-vapor absorption spectrometry.

The concentration and the distribution of mercury in six different brain areas of developing rats (21 and 60 days of age) exposed to methylmercury (MMC) during prenatal life were determined by using pressure decomposition of the tissues and cold-vapor atomic absorption spectrometry. The distribution of mercury in brain samples showed that the metal distributes to all brain areas, but with different levels. The amount of mercury in the brain areas was about 10-100 times higher, depending on the tested area, in MMC exposed rats than in control, at day 21 of age, while it was practically equal to controls at 60 days of age. These data seem to be of importance in order to correlate the presence of mercury in the brain and its distribution to brain areas after a single exposure to MMC with transient or permanent changes in specific neurotransmitter system and altered behaviors.

[Common bile duct calculosis and stenosis of Vater's ampulla. Results of surgical treatment of 307 cases].

A systematic account is offered of a personal series of patients operated for calculosis of the common bile duct, with or without stenosis of Vater's ampulla, over a period of 15 yr. Reference to the clinical and anatomopathological features of these cases is followed by a description of the criteria followed in the choice of the surgical approach. Recent and earlier results are illustrated and some comments are made with regard to the methods employed: ileal choledochotomy, transduodenal papillosphincterotomy, and choledochoduodenostomy). A preference is expressed for papillosphincterotomy in all cases where the correct indications are present.
