CSF producing gall bladder cancer: case report and characteristics of the CSF produced by tumor cells.

Due to an increase in mature neutrophils, a 72-year-old female with gall bladder cancer showed leukocytosis of up to 1.32 X 10(11)/l; hypercalcemia was up to 7.7 mEq/l in her terminal stage. Leukocyte counts and calcium values increased as the tumor progressed. There was no sign of infection or bone marrow metastasis. Cultured cells from the tumor tissue produced high colony-stimulating factor (CSF) activity into the supernatant. The tumor cell-conditioned medium stimulated exclusively granulocytic colonies. The study of this patient shows that leukocytosis was caused by the CSF produced by tumor cells. Approximately 90% of CSF activity was lost by heat treatment at 60 degrees C for 30 min. The CSF was stable over a pH range of 3-11 and was inactivated by treatment with proteolytic enzymes, but was not affected by treatment with DNase or neuraminidase. Molecular weight of the CSF, demonstrated by fractionation using Sephacryl S-200, was approximately 27,000 to 30,000.

Uptake of estrone and estrone-3-sulfate by lung of macaca fuscata.

An equimolar mixture of 3H-E1-S2 and 14C-E1 was injected in one shot into the inferior vena cava near the heart of female Japanese monkey. Following the injection, blood was collected from the aortic arch at intervals of 15 s over a period of 10 min. The concentration of radioactivities in the whole blood and serum was measured. The metabolites were analyzed by DEAE-Sephadex A-25 column chromatography, enzyme hydrolysis, thin layer chromatography and paper chromatography. Both radioactivities of 3H-E1-S and 14C-E1 rapidly decreased in the first 90-s serum sample. The 3H/14C ratio in the 0-15-s serum sample was 5 times higher than the initial ratio of injected compounds. The radioactivities in the serum gradually decreased after 90 s of injection. The 3H/14C ratio in the pulmonary tissue was very low after collecting the final blood sample. This result shows that the most of 3H-E1-S passed through the lung and the larger part of 14C-E1 remained in the lung following injection of these materials. So, it is probable that E1-S is in a form to be carried in the general circulation.

Estrogen biosynthesis in human liver--a comparison of aromatase activity for C-19 steroids in fetal liver, adult liver and hepatoma tissues of human subjects.

After incubation of various tritiated C-19 steroids (androstenedione, testosterone, dehydroepiandrosterone, dehydroepiandrosterone sulfate) with human fetal liver, adult liver and hepatoma tissue homogenates, estrone, estradiol and estriol were analysed after a series of purification steps involving column chromatography, thin layer chromatography and co-crystallization. The findings indicated that the human fetal liver extensively aromatized various C-19 steroids to estrogens, whereas human adult liver and hepatoma tissues exhibited little or no aromatase activities. The formation of estradiol from androstenedione in human fetal liver indicated the presence of 17 beta-hydroxysteroid dehydrogenase in this tissue. It was therefore concluded that although the liver participated in the aromatization process during the fetal stage, extensive aromatization did not take place in the adult liver.

[Aromatization of C-19 steroid in human fetal liver tissue].

In this study we examined whether or not human fetal liver has the ability to aromatize various C-19 steroids to estrone, estradiol and estriol. The human fetal liver homogenate (1g in 10ml of 0.1 M-phosphate buffer) was incubated with the labelled C-19 steroids ([7-3H]-DHEA,-DHEAS, -A:10 microCi, [4-14C]-A:3 microCi) and NADPH at 37 degrees C for 2h in air. [14C]-E1,-E2,-E3 or [3H]-E1,-E2,-E3 (1 x 10(4)dpm, 250 micrograms) were added as tracers after the addition of 3 volumes of ethanol, respectively. E1,E2 and E3 produced were decided by submission to extraction with ethyl acetate, Bio-Rad AG1-X2 resin column chromatography, thin layer chromatography and then co-crystallization to constant specific activity and the 3H/14C ratio. The yield of isolated E1,E2 and E3 were calculated from the 3H/14C ratio of the final crystal. Consequently, DHEA was transformed to 0.78 pmol E1/h/g, 0.14 pmol E2/h/g and 0.18 pmol E3/h/g in human fetal liver, respectively. E1,E2 and E3 were converted from DHEAS to 0.04 pmol/h/g, under 1 fmol/h/g and 0.04 pmol/h/g, respectively. A was transformed to 17.5 pmol E1/h/g, 26.6 pmol E2/h/g and under 1 fmol E3/h/g, respectively. These results indicate that human fetal liver is able to aromatize C-19 steroid to estrogens.

[Therapeutic results of DCMP therapy (2M-80) in acute non-lymphocytic leukemia--the relationship between the residual leukemic cells and duration of remission].

In this study the duration of complete remission by daunomycin, cytosine arabinoside (Ara-C), 6MP and prednisolone (DCMP) therapy using a large dose of Ara-C (200 mg) (protocol 2M-80) was compared with that of DCMP therapy with a low dose of Ara-C (80-160 mg) in acute non-lymphocytic leukemia (ANLL). In protocol 2M-80, the chemotherapy was continued until leukemic blasts in marrow were attained below 3%. Complete remission was induced in about 80% of ANLL patients in both chemotherapies. However, the duration of remission in protocol 2M-80 appeared to be much longer than that of DCMP therapy with a low, dose of Ara-C. This difference was dependent not on the consolidation and maintenance therapy, but on the total dose of Ara-C used and leukemic blasts left in marrow at the end of chemotherapy. This suggests that it is important to reduce leukemic blasts in marrow as low as possible by induction chemotherapy to obtain a long-term remission in ANLL.

A new method for colony formation in vitro by leukemic cells in acute nonlymphocytic leukemia with PHA-stimulated lymphocyte conditioned medium as stimulating factor.

A new agar culture system to clone specifically acute nonlymphocytic leukemia (ANLL) is described, which consists of two phases: an initial liquid phase in the presence of PHA-stimulated lymphocyte conditioned medium (PHA-LCM) and a semi-solid phase. By preincubation of leukemic cells with PHA-LCM in a liquid phase, leukemic cells were found to be more sensitive to forming colonies and clusters in agar medium. In addition, this PHA-two step assay could eliminate colony formation by T cell progenitors. The leukemic origin of the colonies was proven by electron-microscopic analysis, which demonstrated the presence of nuclear blebs and abnormal perinuclear fibril formation. Morphological studies also indicated myeloid differentiation of the cells in the leukemic colonies, although considerable variation was observed among patients. The wide range of linearity between the number of cells plated and the number of colonies grown permits quantitative assay of colony-forming leukemic cells (L-CFU). This assay should be valuable for studies of chemotherapy, growth regulation, and differentiation of L-CFU.
