Metal induced changes in the erythrocyte membrane of rats.

Lead, zinc and copper were administered i.p singly or in combination as acetate salts to rats for 14 consecutive days. It was observed that lead induced drastic changes, copper induced moderate changes but zinc did not cause any significant change in the cholesterol and phospholipid content, hexose, hexosamine and sialic acid levels and activities of the erythrocyte membrane enzymes--acetylcholinesterase (AChE), NADH dehydrogenase and Na(+)-K+ ATPase. In the combined metal treatment the presence of zinc considerably reduced the changes induced by lead and copper.

Effect of perchlorate on mitochondrial function.

Administration of sodium perchlorate to rats for 45 days leads to decreased activities of citric acid cycle enzymes. The oxidation of succinate both in state 3 and state 4 conditions and endogenous ATP content of mitochondria decreased during perchlorate toxicity. The significant decrease in cytochrome aa3 in perchlorate-treated rats may be one of the prime factors involved in the decreased rate of respiration. The permeability of mitochondria of perchlorate-treated rats is altered as indicated by increased oxidation of NADH and low respiratory control ratio (RCR).

Effect of a Siddha drug (Vatharasavangam) on insulin release from isolated pancreatic islets in rabbits.

The Siddha drug--Vatharasavangam (VRV) was studied in rabbits for its hypoglycemic activity. The insulin level in alloxan-induced hyperglycemic animals increased significantly after treatment with VRV. The insulin release from the isolated islets to glucose stimulus and acid phosphatase activity in islets as an index to islet function were measured. The insulin released from the islets in hyperglycemic rabbits decreased from 70 microU/ml (control) to 32 microU/ml to low glucose stimulus. VRV-treated rabbits showed a significant increase in the insulin released from the islet cells (65 microU/ml) to the low glucose stimulus. Similarly, a significant increase of insulin secretion was observed for high glucose stimulus after 30 min in VRV treated rabbits (100 microU/ml), when compared to hyperglycemic rabbits (70 microU/ml). The islet cell acid phosphatase activity in the hyperglycemic rabbits increased significantly after VRV treatment. These findings suggest that VRV acts by stimulating beta-cells to secrete insulin to the glucose signal and improves blood glucose homeostasis.

Lipoprotein profile during perchlorate toxicity.

Perchlorate administration to rats for 45 days alters the lipoprotein profile in plasma. The levels of cholesterol, phospholipids and triglycerides in HDL, LDL and VLDL fractions are significantly increased in perchlorate-treated rats. Post-heparin lipolytic activity of plasma of sodium perchlorate-treated rats is decreased. The risk factor, i.e. the total cholesterol/HDL cholesterol, increases in the experimental animals, indicating that the treatment of rats with perchlorate may develop the susceptibility of the animals to cardiac heart disease.

Influence of black gram (Vigna mungo) trypsin inhibitory fraction on the hepatic protein catabolism in male albino mice.

The effect of black gram and black gram trypsin inhibitor on the protein catabolism of male albino mice has been investigated. Group 1 was given autoclaved black gram (control), Group II raw black gram and Group III the autoclaved black gram incorporated with 1% black gram trypsin inhibitor. Blood as well as urinary urea and creatine were found to be elevated in Groups II and III. Increased levels of arginase, ornithine transcarbamylase and transaminases were noted in Groups II and III. The results suggested an enhanced catabolism of proteins evoked by the native black gram trypsin inhibitor.

Effect of the dietary black gram trypsin inhibitory fraction on the pancreatic activity of male albino mice.

The effect of raw black gram and black gram trypsin inhibitory fraction on the pancreatic function of male albino mice has been evaluated. Animals were fed autoclaved black gram (Group I), raw black gram (Group II) and autoclaved black gram incorporated with black gram trypsin inhibitory fraction (Group III). Pancreatic total protease, trypsin and amylase activities were elevated significantly in Group II and III animals. Analyses of intestinal contents showed a constant level of total protease activity in all the groups, along with a significant reduction in trypsin level in Groups II and III. Increased U-14C-lysine incorporation in pancreas and an enhanced level of amylase in intestinal contents show the hyperactivity of the pancreas in Groups II and III. Pancreas weights increased in Groups II and III. Pancreatic protein/mg pancreas and RNA/DNA ratio remained constant in all three groups. It is concluded with histological studies that the black gram trypsin inhibitor increased the pancreatic activity of mice by developing the cellular hyperplasia of the pancreas.

Effect of dietary green gram trypsin inhibitor and raw green gram meal (Phaseolus aureus Roxb) on the pancreatic activity in rats.

To evaluate the effect of green gram trypsin inhibitor (GGTI) and raw green gram meal (RGG) the experimental animals were fed with the following diets: (i) AGG meal-control (autoclaved green gram meal), (ii) RGG meal and (iii) AGG meal + 1% GGTI. Pancreatic enlargement was observed in rats fed RGG diet and AGG + GGTI diet. Elevated levels of protease and amylase were observed in pancreas. Intestinal contents showed no significant variation in the protease activity, but a significant variation in the amylase activity was evident. Total RNA, DNA and protein content were increased in the pancreas of group II and group III set of animals, suggesting an active proliferation of acinar cells. RNA and DNA content per unit weight of pancreas and the RNA/DNA ratio remained almost constant indicating the hyperplasia. Histological studies revealed hyperplasia of the acinar cells. The results obtained have been discussed.

Influence of dietary raw green gram (Phaseolus aureus Roxb) and green gram trypsin inhibitors on the activity of certain protein metabolism enzymes in rats.

The influence of a raw green gram (RGG) diet, an autoclaved green gram (AGG) diet and green gram trypsin inhibitors (GGTI) incorporated in AGG diet on urinary and blood urea and creatinine levels in rats was studied. The activities of certain liver enzymes of pathways associated with protein or amino acid metabolism were also studied. The levels of urea and creatinine in urine and blood were found to be significantly increased in rats fed the RGG and GGTI-incorporated AGG diets when compared to the animals fed with the AGG diet. The levels of enzyme activities of arginase, ornithine transcarbamoylase, aspartate aminotransferase and alanine aminotransferase were also found to be significantly increased along with that of urea and creatinine, The possible role of GGTI on the altered levels of the above-mentioned parameters is discussed.

Effect of vatharasavangam on U14C-glucose absorption and utilization in hyperglycemic rabbits.

Vatharasavangam (VRV) is a hypoglycemic drug mentioned in the Siddha Literature. Previous studies showed that VRV therapy brings down the blood glucose levels in the hyperglycemic rabbits. The present intestinal perfusion studies with labelled glucose indicate that the blood glucose homeostatis in VRV treated animals is brought about by a significant reduction in the rate of glucose absorption in the intestine. Further an enhanced incorporation of 14C glucose into tissue glycogen is observed in the VRV treated rabbits when compared to the hyperglycemic rabbits.
