[Improvement of effectiveness of antibiotic therapy by pyrimidine derivatives in reparative regeneration of skin in rats].

It was shown on noninbred albino rats with various affections of the skin that pyrimidine derivatives stimulated the skin reparative regeneration and increased the efficacy of antibiotic therapy of Staphylococcus and Proteus infected wounds. The therapeutic effect of 2-methyl-4-amino-6-oxypyrimidine was much higher than that of oxymethyluracil or methyluracil. The pyrimidine derivatives proved to be universal accelerators for reparative regeneration, were compatible with antibiotics and increased their efficacy.

[Pyrimidine derivatives increase antibiotic therapy efficacy after ionizing irradiation].

Pyrimidine derivatives increased the antibiotic therapy efficacy in albino rats irradiated with RUM-7 apparatus for close-focus roentgenotherapy. 2-Methyl-4-amino-6-oxypyrimidine was twice as efficient as oxymethyluracil and 6 times as efficient as methyluracil in the stimulation of the skin reparative regeneration. When the total irradiation was performed with LUCH-1 apparatus in a dose of 6 Gy the pyrimidine derivatives also increased the antibiotic therapy efficacy. After the prophylactic use of the pyrimidine derivatives for 7 days prior to the total irradiation their therapeutic effect increased, the level of the exudative component lowered, the tissue epithelization increased, the terms of the wound healing decreased and the animal lifespan increased.

[Immunomodulating therapy of adjuvant disease in rats using cyclophosphamide in combination with prodigiozan--its effect on anti-infectious resistance].

The effect of immunomodulating therapy of adjuvant disease in rats with cyclophosphamide, prodigiozan and their combinations on infection resistance, weight of the lymphoid organs and leukocyte counts in peripheral blood, as well as the effect of prodigiozan on acute toxicity of cyclophosphamide in intact mice and mice exposed to the Freund's complete adjuvant (FCA) was studied. Prodigiozan did not increase acute toxicity of cyclophosphamide in the intact mice. It lowered the cyclophosphamide toxicity at the background of the FCA and decreased the levels of leukopenia induced by the immunosuppressor in the rats with adjuvant arthritis. It was shown on the models of local infectious inflammation caused by Proteus and lethal sepsis due to P. aeruginosa that the combined use of prodigiozan and cyclophosphamide resulted in correction of the infection resistance impairment induced by both the arthritis development and the immunosuppressor administration.

[Effectiveness of prodigiozan, methyluracil and levamisole in experimental infection after administration of non-steroidal anti-inflammatory drugs].

The effect of nonsteroid antiinflammatory drugs (NAID) such as voltaren, acetylsalicylic acid, indometacin and naproxen on the efficacy of immunostimulators in treatment of infections caused by Proteus and Staphylococcus was studied. It was shown that except naproxen the NAID were capable of decreasing resistance to Proteus infections. Acetylsalicylic acid increased resistance to staphylococcal infections. The rest of the drugs did not change it. Prodigiozan increased resistance to the infections at the background of the NAID. However, volteran and indometacin lowered its effect in infections caused by Proteus. Levamisol stimulated resistance of mice to the infections at the background of volteran and increased resistance to Proteus infections lowered by acetylsalicylic acid. Methyluracyl was not able to increase resistance to the infections at the background of the NAID. It only increased resistance to Proteus infections lowered by acetylsalicylic acid. Levamisol and methyluracyl did not as a rule increase efficacy of antibiotic therapy at the background of the NAID.

[Effect of prodigiozan, levamisole and methyluracil on the course of an experimental infection and primary immune response].

The effect of prodigiosan, levamisol and methyluracil on the process of experimental sepsis caused by Proteus was studied comparatively on mice, and the primary immune response was studied on intact animals treated with prednisolone, cyclophosphan or azathioprin. It was shown that prodigiosan was a more active stimulator of antiinfectious resistance of the intact animals than levamisol or methyluracil. Its effect was evident with administration before the infection, while the effect of levamisol or methyluracil was evident in administration after the infection. When used prophylactically, prodigiosan increased the average life-span of the mice given prednisolone, had no effect on the infection process in the presence of cyclophosphan and increased the antibiotic therapy efficiency at the background of either prednisolone or cyclophosphan. Administration of prodigiosan after the infection without the use of antibiotics was not efficient in immunosuppression, whereas in combination with levamisol it increased the tolerance to the infection at the background of cyclophosphan. When used for the treatment purposes, levamisol had no effect on survival of the animals. However, it increased the average life-span of the intact mice also at the background of prednisolone. Prodigiosan stimulate the primary immune response in the intact animals at the background of azathioprin or cyclophosphan but not prednisolone. Levamisol increased the number of the antibody-producing cells (APC) in the intact animals in some experiments. At the background of azathioprin it had no effect on the number of the APC and at background of prednisolone or cyclophosphan it even lowered their number.(ABSTRACT TRUNCATED AT 250 WORDS)

[Combined use of prodigiozan and methyluracil with immunosuppressive drugs in adjuvant arthritis in rats].

The effect of the combined use of prodigiozan (0.5 mg/kg bw) prodigiozan with immunosuppressants on the development of adjuvant arthritis was studied on rats. The drugs were given during the prearthritis period from the 1st to the 16th day after injection of complete Freund's adjuvant. It was shown that the combined use of prodigiozan and cyclophosphamide (5 mg/kg) induced significant inhibition of the arthritis development and extraarticular signs of the disease. The efficacy of the combination was higher than that of cyclophosphamide alone. Azathioprine (4 mg/kg/bw) produced no immunosuppressant effect and did not influence the inhibitory effect of prodigiozan on the development of the adjuvant disease. Prednisolone (1.6 mg/kg) either did not inhibit the arthritis development. However, it eliminated the prodigiozan effect. Methyluracil did not change the effect of the immunosuppressants on the articular syndrome. Still, it increased the number of nodular affections in the animals treated with cyclophosphamide and prednisolone. The data obtained show the possibility of prodigiozan combination with certain immunosuppressants in autoimmune affections and confirm the suggestion that the inhibitory effect of this drug is mediated through macrophages.

[Effect of prodigiozan, levamisole and methyluracil on delayed hypersensitivity against a background of immunosuppressor use].

Immunostimulants, such as prodigiosan, levamisol and methyluracil, as well as their combinations with imuran, prednisolone or cyclophosphamide were studied for their effect on the delayed type hypersensitivity (DTH) induced by dinitrofluorobenzene alcohol solutions applied in challenge doses to the floor of the auricle of mice. It was shown that the immunostimulants did not affect the DTH in intact mice. In mice treated with imuran the DTH was significantly increased only by prodigiosan. Prednisolone used for a prolonged period before and after the sensitization and cyclophosphamide administered 24 hours before the sensitization increased the DTH. According to the literature data it was connected with T-suppressor inhibition. With the use of cyclophosphamide it was also connected with B-suppressor inhibition. Under such conditions the DTH was decreased to the control level by prodigiosan after the use of prednisolone or by levamisol after the use of cyclophosphamide. This was probably associated with increasing of the suppressor effect of the macrophages and activation of the T-suppressor effect by levamisol. Methyluracil had no effect on the DTH.

[Aspects of the immunodepressive action of gentamycin and rifampicin].

The effects of rifampicin and gentamicin on the count of the antibody-forming cells (AFC), the titers of hemagglutinins, endogenous colony-formation and activity of the reticuloendothelial system (RES) were studied on uninbred albino mice. An attempt was made to correct the immunity defects induced by rifampicin. In addition to the inhibitory effect of the antibiotics on the primary immune response, i.e. the count of the AFC described earlier, suppression of hemagglutinin accumulation on the 4th day after immunization and inhibition of the absorption function of the RES after administration of rifampicin in a dose of 250 mg/kg were shown. Both the antibiotics induced stimulation of endogenic colony formation when administered for 10 days before the irradiation. It was only gentamicin that produced a stimulating effect after irradiation. The combined use of rifampicin with the immunostimulants prodigiozan, methyluracil or levamisole according to different schemes showed that the only effective combination was that of rifampicin with methyluracil (400 mg/kg) or levamisole (5 mg/kg).

[Stimulation of nonspecific body resistance in transplantation].

The experiments on mice showed in principle a possibility of using prodigiozan, a stimulant of the host immunobiological reactivity together with immunodepressants in transplantation. Imuran in doses of 25 or 60 mg/kg and prednisolone in doses of 30 or 50 mg/kg were used on alternate days at a rate of 5 injections of every drug per course. Prodigiozan was administered intramuscularly in a dose of 0.5 mg/kg on the 4th and 8th-9th days of the immunodepressant administration. Addition of prodigiozan to the scheme of the combined use of the immunodepressants provided an increase in the survival rate of mice with staphylococcal and coli infections and prolongation of the skin allotransplant rejection time. No effect of prodigiozan on immune depression due to imuran was observed. The results suggest that screening of drugs for the use in transplanthology for prevention of infectious complications in the presence of immunostimulants is advisable among stimulants of the host nonspecific resistance.

[The effect of erythromycin on immunity in immunodeficiency].

The effect of 12.5 and 25 mg/kg doses of erythromycin on the immunity of intact mice and in cyclophosphan- and azathioprine-induced immunodeficiency was studied. Erythromycin has no effect on the number of antibody-forming cells in the spleen of intact and immunocompromised with cyclophosphan animals. It increases their number against the background of azathioprine. Immunosuppression may change the character of the effect of erythromycin on delayed hypersensitivity. Erythromycin stimulates phagocyte activity in intact animals and in immunosuppression.

[The metabolic effects of isradipine, ramipril, Ednit and beta-adrenoblockers in patients with non-insulin-dependent diabetes mellitus and arterial hypertension].

The effect of isradepin, ramipril, ednyt, and beta-adrenoblockers on carbohydrate and lipid metabolism and free-radical oxidation in patients with insulin-independent diabetes mellitus was studied. Isradepin caused an increase in high-density lipoproteins (HDLP) in the blood serum and the content of esterified cholesterol in HDLP, and reduced the arterogenicity coefficient. Rampiril reduced the basal level of glycemia and insulin. Treatment with ednyt lead to increase in the content of HDLP and the content of free cholesterol in them. In the course of treatment isradepin normalized chemoluminescence of the serum and urine, rampiril that of serum, and beta-adrenoblockers the chemoluminescence of the blood. The favorable changes induced by the drugs under study allow them to be recommended for the treatment of arterial hypertension in patients with insulin-independent diabetes mellitus.
