The demonstration of indole formation is of great value for the identification of pathogenic anaerobes. A reduction of the pH to values below the critical range of 5.1-5.3 in complex media 5.5 in simple testing substrates, respectively, which is caused by the presence of fermentable carbohydrates and will result in false-negative reactions. For this reason, the indol test should be performed with an adequate buffer capacity and an addition of fermentable carbohydrates should be avoided. Strains showing a false-negative reaction in the spot indole test can be reliably accounted for by means of the rapid test in buffered tryptophan solution described here without having to use additional anaerobic culture methods.

The concentration kinetics of combinations of antibiotics similar to those taking place after i.v. administration was simulated in an in vitro model taking into account the different half-life periods of the antibiotics. A hose pump was used for continuous supply of sterile nutrient to a liquid bacterial culture containing both antibiotics in a preselected concentration and for removal of fluid from the system at an identical flow rate. To account for the different half-life periods of the antibiotics, a solution containing the antibiotic having a longer half-life in a corresponding concentration was continuously added by pumping. Samples were taken at fixed intervals to determine bacterial counts and check the expected concentrations of active substance did not contain antibiotics. This model has been developed to enable a determination of the dynamic relationship between the course of concentrations of antibiotics and their antibacterial activity. To permit a statement on the synergistic, indifferent or antagonistic activity of the combination, in analogy to the FIC indices in the checkerboard technique, the relative reduction of the bacterial count by the combination as compared to the individual components at time t was calculated. The model has been demonstrated by the example of the combined action of azlocillin and gentamicin against a strain of Klebsiella pneumoniae. In a simple way, the model permits any increase in the number of combination partners having different half-life periods. The mathematical development of the model is explained in detail.
A disperse dye immunoassay (DIA) using Fantagen-dye sol particles as label was developed for the measurement of rubella antibodies in human serum specimen. The DIA has a sensitivity comparable to the hemagglutination-inhibition (HI) test, whereas the detection limit for a similar ELISA-system developed in parallel is lower by a factor of about two orders of magnitude. The results of testing 34 random serum specimens with the 3 different methods showed a significant correlation (v = 0.73 for DIA vs. ELISA). Titer rises in 10 paired sera (acute/convalescent) estimated with the DIA- and ELISA-methods were in good agreement with H1-results. Only two of the 10 pairs differed in titer rise by no more than one dilution step.

The virucidal effect, the inhibition of virus adsorption and penetration and the influence on later phases of replication of human pathogenic viruses were studied. The compounds showed a significant virucidal effect to enveloped viruses (measles virus, herpes virus type 1 and 2), whereas the compounds were ineffective against nonenveloped viruses (Coxsackie virus A9 and B1, attenuated poliovirus type 1, adenovirus type 1). Interactions with the viral envelope are supposed.

A clinical relevant diagnostic must be performed early and quickly with sufficient sensitivity and specificity. Under these aspects the virological diagnostics with its methods for the demonstration of viruses as well as for the establishment of an immune response is estimated. As to the proof of pathogenic organisms or antigens is referred to electron-microscopic and immunologic methods (e.g. the fluorescence-antibody-technique, enzyme immunoassay) as well as to the hybridization of nucleic acid. The diagnostics of the immune response is above all based on the demonstration of antibodies. Emphasis is laid on the methods for the establishment of IgM-antibodies, which mostly conclude to an acute virus infection.

Among the numerous labels employed in immunoassays dye sol particles have been introduced but recently. This report deals with a modification of the disperse dye immunoassay (DIA). It is shown that Fantagen dyes to which antibodies were adsorbed physically and tablet blisters as solid phase give satisfactory results with the model antigens alpha 2-macroglobulin and the group specific component of human plasma.
