[Activation of endogenous lipid peroxidation in the brain in oxidation stress caused by administration of iron and its prevention by vitamin E].

Looking for an appropriate model of accelerated aging in vivo we investigated the content of endogenous products of lipid peroxidation (LP) in the rat brain after single or 4 day-lasting intramuscular injection of complex-bind iron (ferum Hausman, 50 mg/kg body weight) like promoter of LP. We found that the single administration of this iron complex fails to induce endogenous LP; after 4 day-application of iron we observed significant increase in content of primary (lipid peroxides) and final (fluorescent) products of LP. Iron-promoted activation of endogenous LP could be abolished by animal pretreatment with the natural antioxidant alpha-tocopherol. The calcium antagonist nifedipine didn't affect the content of endogenous LP products neither alone nor in combination with alpha-tocopherol.

Possibilities of extending the indications for application of single antibacterial doses in urological practice.

The authors report very good results of the application of single antibacterial doses in the therapy of acute bacterial cystitis and asymptomatic bacteriuria and point out that in addition to these already established indications, single doses can be applied successfully in some other forms of urinary infection, too. They share the clinical results and on the basis of some experiments maintain the standpoint that postoperative urinary infections are also suitable for this therapy (if no evidence of pyelonephritis is available), as well as the infections of the upper urinary tract (if the isolated microorganisms are not charged with antibodies). A scheme of the so-called "long-term therapy with single dose once a week" is proposed instead of the systematic 3-6 months therapy. The results are compared to the routine 7-10 day course of treatment; as a matter of fact, they are the same. The advantages of the single-dose therapy are discussed.

Pharmacokinetics of vephylline--a new N-substituted theophylline derivative.

Vephylline (7,2-bis-2-hydroxyethylamino-1, 3-dimethylxanthine tartarate) is a xanthine derivative with high bronchodilating activity, low toxicity, and weak effects on the central nervous system. The aim of this study is to determine the pharmacokinetic parameters of vephylline after intravenous and oral (in solution and in tablets) administration to rabbits. Vephylline (dose 50 mg/kg b.w. intravenousely and orally in solution and dose 53.5 mg/kg b.w. in the form of tablets) is administered to the rabbits in an autocontrol crossover design at 7-days intervals. After the intravenous administration the distribution is relatively fast (t1/2 alpha = 3.28h). High values of the apparent volume of distribution--12.15 1/kg suggest tissue accumulation. Elimination is considerably slower (t1/2 beta = 19,00 h) than distribution. After oral administration of the drug in solution the absorption half-life is short and the bioavailability is relatively high. Peak plasma levels are attained at the first hour. The differences in the distribution and elimination patterns for vephylline and theophyline could determine a longer effect for the new bronchodilating drug. The results are discussed in regard to the future clinical application of vephylline.

[Clinical importance of determining the digoxin level in the serum in long-term beta-methyldigoxin treatment].

The digoxin level was determined by radioimmunologic method in 54 patients with cardiac failure on constant maintaining glycoside treatment with beta-methyldigoxin in doses of 0.7-0.1, 4 mg weekly. The results showed that: Between the maintaining dose of Beta-Methyldigoxin and the serum digoxin level there is not always parallelism in cases of achieved cardiac compensation. In spite of that the achievement of cardiac compensation and its maintainance in more than 85% of the cases is possible with a mean dose of Lanitop 1.0 mg weekly and digoxin serum level between 1.0 and 2.0 ng/ml. Between the toxic manifestations observed and the doses applied there is not a strong correlation. But the high digoxin levels are always potentially toxic.

On some presynaptic effects of an aminotetraline derivative.

The article studies the effect of a newly-synthesized aminotetraline derivative with code 1b on some presynaptic mechanisms of nerve transmission. Compound 1b (1 X 10(-9) M) increases the contractions of electrically stimulated isolated rat anococcygeal muscle: on the background of cocaine (1 X 10(-7) M), the contractile response after 1b is potentiated, whereas after phentolamine (1 X 10(-9) M) compound 1b does not increase the contraction of the muscle. Compound 1b in concentration from 1 X 10(-8) M to 1 X 10(-7) M manifests a concentration-dependent potentiating effect (EC50-3.95 X 10(-8) M) on the contractile response of electrically stimulated isolated rat vas deferens. The alpha-adrenergic blocker yohimbin (1 X 10(-7) M) and compound 1b (1 X 10(-6) M) increase the contractions of electrically stimulated mouse vas deferens. The potentiating effect of 1b is eliminated by clonodone (1 X 10(-19) M) and haloperidol (1 X 10(-5) M). On electrically stimulated guinea-pig ileum 1b (2 X 10(-6) M) and morphine (1 X 10(-6) M) inhibit the contractions of the isolated organ, while their combined administration potentiates the inhibitory effect of morphine. Both the combined and the independent effects of 1b and morphine are eliminated by naloxone (1 X 10(-6) M) and 4-aminopyridine (1 X 10(-7) M). Both compound 1b (2 X 10(-6) M) and morphine (1 X 10(-6) M) manifest potentiated inhibitory effect on the contractile response of guinea-pig ileum, administered on the background of dopamine (1 X 10(-5) M) and haloperidol (1 X 10(-6) M).(ABSTRACT TRUNCATED AT 250 WORDS)

[Brulamycin in the treatment of urinary infections--microbiological and clinical research].

The authors report the results frop microbiological and clinical studies of the new aminoglucoside antibiotic brulamycin. The susceptibility of 12 392 bacterial strains was determined versus brulamycin and the other routinely applied antibacterial preparations in our country and established that, according to that index brulamycin, in vitro, gives way only to amikin. Fifty two patients with severe uroinfections were treated (30 adults and 22 children) and healing (sterilization of urine) was attained in 10 per cent of the adults and 40,9 per cent of the children. Improvement (reduction of the bacterial number to insignificant value) was established in 42,3 per cent of all patients. In parallel with the favourable microbiological indices, improvement or normalization of the clinical and paracminical indices of the uroinfection were also established. The antibiotic is very well tolerated, by children one-year of age, including. With those properties brulamycin should widely be administered in the clinical practice of uroinfection treatment.

Pharmacological and toxicological studies of an original xanthine derivative with bronchodilatating activity.

The article studies the pharmacological properties and the toxicity of the original theophylline derivative G112: 7-/2-bis-/2-hydroxy-ethyl/-aminoethyl/-1,3-dimethylxanthine tartarate, synthesized by Y. Gagaouzov and P. Peykov. Considerable bronchorelaxing activity, measurable with novphylline, has been found in experiments in vivo, as well as in isolated tracheal preparations contracted by various spasmogens. In equimolar concentrations the compound has a weaker inhibitory effect on the phosphodiesterase in the lungs and the brain, compared with theophylline, and it almost does not activate the myocardiac phosphorylase "a" and the lipolysis. A weak and brief hypotensive effect has been established, moderate diuretic action and excitatory effect on the central nervous system, similar to that of novphylline. The compound has considerably lower toxicity than novphylline and it does not lead to changes in the body weight and in the basic haematological and biochemical parameters after treatment of rats for three months.
