Use of rigid spherical inclusions in Young's moduli determination: application to DNA-crosslinked gels.

Current techniques for measuring the bulk shear or elastic (E) modulus of small samples of soft materials are usually limited by materials handling issues. This paper describes a nondestructive testing method based on embedded spherical inclusions. The technique simplifies materials preparation and handling requirements and is capable of continuously monitoring changes in stiffness. Exact closed form derivations of E as functions of the inclusion force-displacement relationship are presented. Analytical and numerical analyses showed that size effects are significant for medium dimensions up to several times those of the inclusion. Application of the method to DNA-crosslinked gels showed good agreement with direct compression tests.

Mechanical properties of a reversible, DNA-crosslinked polyacrylamide hydrogel.

Mechanical properties of a polyacrylamide gel with reversible DNA crosslinks are presented. In this system, three DNA strands replace traditional chemical crosslinkers. In contrast to thermoset chemically crosslinked polyacrylamide, the new hydrogel is thermoreversible; crosslink dissociation without the addition of heat is also feasible by introducing a specific removal DNA strand. This hydrogel is characterized by a critical crosslink concentration at which gelation occurs. Below the critical point, a characteristic temperature exists at which a transition in viscosity is observed. Both temperature-dependent viscosity and elastic modulus of the material are functions of crosslink density.

Parameter estimation in a crossbridge muscle model.

Models of muscle crossbridge dynamics have great potential for understanding muscle contraction and having a wide range of application. However, the estimation of many model parameters, most of which are difficult to measure, limits their applicability. This study developed a method of estimating parameters in the Distribution Moment crossbridge model from measurements of force-length and force-velocity relationships in cat soleus single muscle fibers. Analysis of the parameter estimates showed that the detachment rate parameters had more uncertainty than the attachment rate parameter, which could reflect physiological variations in the contractile protein content and in the response of muscle to lengthenings.

Probiotics as functional foods.

Probiotic products have been widely used in Japan and Europe for years. Probiotics are now emerging as an important category of food supplement in the United States. Questions about the biologic nature, available products, claimed health benefits, and safety and regulation of probiotics are important for both consumers and nutrition professionals. Probiotics can be considered functional foods because they provide health benefits beyond the traditional nutrition function. With few exceptions, most probiotic products currently available contain lactic-acid-producing bacteria, which mainly belong to the genera Lactobacillus and Bifidobacterium. We reviewed the scientific papers published in major nutrition journals and abstracts available on the PubMed website regarding probiotics. Evidence suggests the following beneficial effects of probiotics: normalization of the intestinal microflora, ability to block the invasion of potential pathogens in the gut, prophylactic or therapeutic treatment for several types of diarrhea, relief of symptoms of irritable bowel syndrome and inflammatory bowel disease, amelioration of lactose intolerance, prevention of colon cancer, modulation of immune function, inhibition of Helicobacter pylori, and possible enhancement of calcium absorption and reduction of blood cholesterol levels. Mechanisms for the above benefits have been proposed, but none has been proven. An adequate level of viable bacteria in a probiotic product and an appropriate daily dose are critical to achieve a health benefit. Because probiotics are not known to be pathogenic and their upper tolerable level is high, they could be promoted as a beneficial food supplement. Currently, no disease-prevention or therapeutic claim for probiotics is legally allowed.
