Study on degradation of methyl orange using pelagite as photocatalyst.

Pelagite of East Pacific Ocean as a low-price, very high reserve photocatalyst, has been successfully used for the first time to realize total degradation and decoloration of methyl orange within 120 min through photocatalysis. The photocatalysis and degradation performance of pelagite for methyl orange under the effect of various factors has been studied. Results show that pelagite has satisfactory photocatalysis effect in degradation of organic compounds.

Simultaneous analysis of seven astragalosides in Radix Astragali and related preparations by liquid chromatography coupled with electrospray ionization time-of-flight mass spectrometry.

A method has been developed for the qualitative and quantitative analysis of pharmacologically active astragalosides isolated from several species of the genus Astragalus by high performance liquid chromatography coupled with electrospray ionization time-of-flight mass spectrometry. Seven astragalosides in Radix Astragali and their commercial pharmaceutical preparations were analyzed using the developed method. The extracted ion current chromatograms were obtained from the total ion current chromatogram using the m/z of [M+Na]+ ions produced by target compounds for peak determination. The limits of detection and limits of quantification were in the range of 0.10-0.22 ng and 0.22-0.52 ng in full scan mode, respectively. All calibration curves showed good linear regression (r2 > or = 0.9965) within the test range. The overall intra- and inter-day precision was less than 2.86% for peak area and the accuracy was higher than 92.9% on using ginsenoside I as internal standard. The assay was successfully utilized to analyze the major biologically active astragalosides in six samples of Astragalus membranaceus (Fisch.) Bge var. mongholicus (Bge.) Hsiao. and eight commercial preparations. The overall results demonstrate that this method is simple, selective, and suitable for the quality control of Chinese medicine and their preparation in the low nanogram range.

Systematic analysis of genes required for synapse structure and function.

Chemical synapses are complex structures that mediate rapid intercellular signalling in the nervous system. Proteomic studies suggest that several hundred proteins will be found at synaptic specializations. Here we describe a systematic screen to identify genes required for the function or development of Caenorhabditis elegans neuromuscular junctions. A total of 185 genes were identified in an RNA interference screen for decreased acetylcholine secretion; 132 of these genes had not previously been implicated in synaptic transmission. Functional profiles for these genes were determined by comparing secretion defects observed after RNA interference under a variety of conditions. Hierarchical clustering identified groups of functionally related genes, including those involved in the synaptic vesicle cycle, neuropeptide signalling and responsiveness to phorbol esters. Twenty-four genes encoded proteins that were localized to presynaptic specializations. Loss-of-function mutations in 12 genes caused defects in presynaptic structure.

Somatic misexpression of germline P granules and enhanced RNA interference in retinoblastoma pathway mutants.

Caenorhabditis elegans homologues of the retinoblastoma (Rb) tumour suppressor complex specify cell lineage during development. Here we show that mutations in Rb pathway components enhance RNA interference (RNAi) and cause somatic cells to express genes and elaborate perinuclear structures normally limited to germline-specific P granules. Furthermore, particular gene inactivations that disrupt RNAi reverse the cell lineage transformations of Rb pathway mutants. These findings suggest that mutations in Rb pathway components cause cells to revert to patterns of gene expression normally restricted to germ cells. Rb may act by a similar mechanism to transform mammalian cells.

[The effect of matrix metalloproteinase-1 on root surface dentin matrix: a scanning electron microscope observation].

OBJECTIVE: To observe the effect of matrix metalloproteinase-1 (MMP-1) from human host on degradation of dentin organic matrix of root dentin. METHODS: The freshly extracted caries-free impacted teeth were selected. Teeth were cut transversely under the enamel-cementum junction into dentin sections with a thickness of about 5 mm. Then all sections with removal of cementum, pulp and predentin were randomly divided into four groups. In the first group, dentin sections were demineralized with acid solution for 21 days, and then incubated with MMP-1 solution for 7 days; the second group were only treated with acid solution for 21 days; the third group were only attacked by MMP-1 solution for 7 days; and the fourth group were untreated as a control. Then all sections were dehydrated in ascending strength of alcohol, critically dried, coated with platinum, and then observed under scanning electron microscope(SEM). RESULTS: The dentin sections of root surface attacked by acid and MMP-1 showed that demineralization of dentin mineral and degradation of dentin matrix fibrae synchronously happened. The dentin matrix fibrae wasn't degradated in the groups treated with acid or MMP-1. CONCLUSION: The proteinases from human host may play an important role in the development of root surface caries. MMP-1 may distinctly degradate the organic matrix of demineralized dentin.

[Animal experimental study of the treatment of periapical disease with recombinant human bone morphogenetic protein-2 composite].

OBJECTIVE: To investigate the effect of the treatment of periapical diseases with recombinant human netic protein (rhBMP-2) composite in dog models so as to provide basis for its clinical application. METHODS: The endotoxin-bacteria mixed fluid was injected into each root canal of the experimental teeth of dogs and the animal periapical were built. The rhBMP-2 composite was mixed by rhBMP-2, TCP, collagen and metronidazole and used in the treatment cal diseases in dog models as root apex screen. The imageology and pathology research were went on. RESULTS: The effective rate of the experimental group was remarkably higher than that of the carrier group and the control group with remarkable ference (P < 0.05). The quality of the repair of tissue was obviously better than the two other groups. CONCLUSION: The rhBMP-2 composite is a promising biological root-canal filling material.

[The expression of c-fos and transmitter calcitonin gene-related peptide in the chronic compressive injury of the nerve root].

OBJECTIVES: To investigate the significance of c-fos oncogene morphogenetic protein's locational expression, and the correlativity between nerve transmitters calcitonin gene-related peptide (CGRP) expression and nerve root's functional change using the animal model of the chronic compressive injury in the nerve root. METHODS: The animal model of chronic compressive injury of the nerve root was established by transplanting autogenous cancellous bone into the intervertebral foramen. During different injury phase (1, 2, 4, 8, 12, 24 weeks after operation), the functional status of the nerve root was determined under the monitoring of evoked potential, and the expression changes of c-fos oncogene morphogenetic protein and nerve transmitter CGRP were detected using in situ hybridization technique and their expression intensity was determined using automatic image analytic instrument respectively. RESULTS: One week after operation, the c-fos expression strengthened in both anterior and posterior root fiber obviously. Two to four weeks after operation, the expression of the posterior root fiber weakened than the anterior root fiber. After 12 weeks, the anterior root fiber expression turned down obviously, however the posterior root fiber expression backed up slightly compared with that of the 8 weeks. By the time of 24 weeks after operation, the expression enhancement in all roots disappeared. CGRP expression increased obviously at the site of compressive axon of both anterior and posterior root. The expression of the posterior root axon and ganglion cell was higher than that of the anterior root axon. CGRP expression was diminished in the second week than the first week, and that was especially obvious in the posterior root and ganglion cell. But 4 weeks after operation, the expression enhanced once more, and that was more obvious inside the anterior root axon. Eight weeks after operation, the expression intensity attained the high peak. Twelve weeks after operation, the expression started the slow-moving descent. CONCLUSIONS: The expression of c-fos gene protein is beneficial to localize the damaged part of certain nerve. During chronic injury, the degeneration of posterior root sensory fiber is earlier than the anterior root motor fiber. The expression of CGRP strengthened when the nerve fiber degenerated by the harmful stimulation, and the expression intensity is positively related with pain. That suggests when the nervous tissue is hurt, the information of warning and regulation should be sent out to our body.

A conserved siRNA-degrading RNase negatively regulates RNA interference in C. elegans.

In many organisms, introducing double-stranded RNA (dsRNA) causes the degradation of messenger RNA that is homologous to the trigger dsRNA--a process known as RNA interference. The dsRNA is cleaved into short interfering RNAs (siRNAs), which hybridize to homologous mRNAs and induce their degradation. dsRNAs vary in their ability to trigger RNA interference: many mRNA-targeting dsRNAs show weak phenotypes, and nearly all mRNAs of the Caenorhabditis elegans nervous system are refractory to RNA interference. C. elegans eri-1 was identified in a genetic screen for mutants with enhanced sensitivity to dsRNAs. Here we show that eri-1 encodes an evolutionarily conserved protein with domains homologous to nucleic-acid-binding and exonuclease proteins. After exposure to dsRNA or siRNAs, animals with eri-1 mutations accumulate more siRNAs than do wild-type animals. C. elegans ERI-1 and its human orthologue degrade siRNAs in vitro. In the nematode worm, ERI-1 is predominantly cytoplasmic and is expressed most highly in the gonad and a subset of neurons, suggesting that ERI-1 siRNase activity suppresses RNA interference more intensely in these tissues. Thus, ERI-1 is a negative regulator that may normally function to limit the duration, cell-type specificity or endogenous functions of RNA interference.

[The test of metal-ceramic bonding strength among three ceramic alloies].

PURPOSE: To compare the metal-ceramic bonding strength of different ceramic alloies. METHODS: 30 wax sheets were divided into 6 groups at random. Each group included 5 sheets. After being invested conventionally with Bellaves SH,the samples were casted and fused with porcelain.The metal-ceramic bonding strength was evaluated. RESULTS: The highest bonding strength was found in Bio Herador N alloy, and then in TILITE alloy and Heraenium S alloy respectively. Bio Herador N alloy and TILITE alloy all had significant differences of the bonding strength before and after treatment (P<0.05). There was no significant differences in the bonding strength of the Heraenium S alloy after treatment (P>0.05). CONCLUSION: The bonding strength of precious metal-ceramic was higher than that of non-precious metal-ceramic.Pre-oxygen treatment can improve the metal-ceramic bonding strength of Bio Herador N alloy and TILITE alloy,but it had no effect on the metal-ceramic bonding strength of the Heraenium S alloy.

[Recovery and evoked potential of nerve root under variable chronic injury].

OBJECTIVE: To study the recovery and mechanism of nerve root under variable chronic injury and to determine the alerting index of the evoked potential of the irreversible injury to the nerve root, so as to offer the evidence for selecting treatment methods, judging prognosis and grasping treatment juncture. METHODS: Autogenous cancellous bones were planted into the right C7-8 and C8-T1 intervertebral foramens in 30 cats with weight 3-5 kg to make chronic injury models. The left side was for auto-contrast. By 24 weeks' observation and dynamic supervisory of evoked potential, the injury degrees were ascertained and classified into I to V degree groups. Then the operation of decompression was performed on every group. During the following 32 weeks, the evoked potential survey and pathological tissue examination were made every week to observe the function recovery of the injured roots. RESULTS: The tissue form and function of the nerve roots with I and II degree injuries recovered well within 2 to 6 weeks. Those with II degree injury began to recover in the 4th week, and gradually recovered to normal 12 weeks later. In the group of IV degree injury, the recovery was slow. They could recover completely in 3 of 6 cats, partly in 2 and hardly recovered in 1. The function and tissue form of nerve roots with V degree injury could hardly recovered. CONCLUSION: In the case that the compression has been removed before the nerve roots suffer II degree injury. The tissue form and function of the nerve roots will recovered satisfactorily, whereas they may suffer irreversibly injury by IV degree injury, and even can hardly recover by V degree injury. During the supervisory process of the recovery of the injured nerve roots, sensitive evoked potential is a sensitive sign of early recovery and motion evoked potential is a reliable sign of recovery extent.

[The preparation and in vitro release test of sustained release delivery gutta-percha point containing metronidazole].

PURPOSE: To develop sustained release delivery gutta-percha point containing metronidazole (SRDGM) for root canal disinfection, and determine the drug concentration in vitro and the time that the device maintains the effective drug concentration. METHODS: The gutta-percha was selected as the carrier and adsorbed metronidazole etc. The point was coated with mixation of ployethylene glycol and acetate fibre to prepare the groups of SRDGM.The group that released the longest period of the effective drug was selected for the following study. Eight teeth were selected, the SRDGM was used to disinfect the root canal. The adsorption was measured for 24 hours and 10 days. RESULTS: The SRDGM contained metronidazole 2013 microg, it could release 68.24% of the total drug in 24 hours in vitro. The effective metronidazole concentration released lasted more than 10 days. On the 10th day, there was also 33.13 microg/ml metronidazole released which was more than minitory inhibitory concentration of metronidazole. CONCLUSIONS: SRDGM could carry the metronidazole and control the release rate. It could maintain the effective drug concentration for a long time (>10 days) in root canal.

[Research advances of adult stem cells and dental stem cells].

Human stem cells have a major effect in the development of human bodies. Acting as an in vitro model, human stem cells have a great value in the study of the mechanism of individual development and in the cell-replacement treatments and gene therapy of tissue deficiency diseases. The study of stem cells has been given rise to a keen interests in the bio-medical area at present. This paper reviews the present status and progress of adult stem cells and dental stem cells.

[Empirical analysis and forecasting for SARS epidemic situation].

Based on the SIR epidemic model, the empirical analysis for the SARS epidemic situation is presented. The results show that the SIR model s applicable approximately for describing the SARS epidemic situation. The forecasting for SARS epidemic situation is also given.

[The development and in vitro release rate determination of controlled-release delivery gutta-percha point containing metronidazole compound].

OBJECTIVE: To prepare the controlled-release delivery gutta-percha points containing metronidazole compound (CDGMC) and to determine its release rate in vitro. METHODS: The drug points were made by using compound drugs and gutta-percha as a carrier, and CDGMC were prepared followed by enveloping a release membrane outward. The best formula was selected according to the release parameters of the drugs extracted in the release experiments in vitro. The CDGMC were placed into the extracted teeth after root canals were routinely prepared. The non-drug CDGMC was used as the control. The absorbency of the drugs in normal saline (37 degrees C, pH 7.4) was determined timely. The percentage of release and cumulated release of the drugs were calculated according to the concentrations of drugs in medium. RESULTS: The in vitro experiments showed that this system contained 1,880 micrograms of metronidazole and 267 micrograms of ciprofloxacin. The experiments in healthy single root canal showed that the drug release amounts around the perioapical area were metronidazole 88.54 micrograms/ml and ciprofloxacin 9.05 micrograms/ml in 10 days. CONCLUSION: CDGMC can continuously release effective drug concentrations more than 10 days and could be considered as an ideal method for clinical application.

[The observation of the effect of metronidazole-chlorhexidine solution on treatment of periapical periodontitis].

OBJECTIVE: To observe the effect of metronidazole-chlorhexidine solution on treatment of periapical periodontitis. METHODS: 202 teeth suffering from chronic periapical periodontitis were selected and divided into two groups. One group including 102 teeth were treated with metronidazole-chlorhexidine solution. Another group including 100 teeth were treated with formocresol. The acute symptoms during root canal therapy were observed. 164 teeth suffering from acute periapical periodontitis were also selected and divided into two groups. One group including 80 teeth were treated with metronidazole-chlorhexidine solution. Another group including 84 teeth were treated as the above except the pulp chamber keeping open. The other 44 teeth, in which purulence also presented in the root canal after treated with formocresol and iodoform for three times or more, were treated with metronidazole-chlorhexidine solution. The presence of purulence was evaluated. RESULTS: There are significant differences between the two groups of chronic and acute periapical periodontitis(P<0.05). The effective rate was 97.6% after changed to metronidazole-chlorhexidine solution treatment. CONCLUSION: Good treatment effect could be achieved by using metronidazole-chlorhexidine solution as the root canal cleaning and disinfection agent.
