[Electron microscopic immunohistochemistry in the diagnosis of human lymphotropic viral diseases].

Immunohistochemistry is finding an ever increasing application for electron microscopic diagnosis of human viral diseases. Certain progress has been made in the use of peroxidase-DAB/OsO4 as an electron-dense marker, and colloid gold. The paper discusses immunohistochemistry application for electron microscopic detection of such lymphotropic viruses as VSV (virus of vesicular stomatitis cultured in mink's lung cells), HTLV-1 (virus of human T4-cell C91Pl lymphoma culture) and HTLV-III/HIV-1 (virus growing in human T4-cell H9 culture).

Acquired Immunodeficiency Syndrome↗

[Leiomyosarcoma of the small intestine].

In addition to the literature data the authors describe their own observation of small intestinal leiomyosarcoma. The case was interesting for an uncommon metastatic course and atypical histological appearance presenting difficulties for clinical diagnosis and morphological interpretation. The final diagnosis rested with electron microscopy.

[Catecholamine-producing tumors of the chromaffin tissue. Morphological and ultrastructural characteristics].

Histological and ultrastructural studies were carried out in 41 hypertensive patients showing increased urine excretion of catecholamines and vanilmandelic acid due to chromaffinoma of the adrenal and other sites: malignant chromaffinoma of the adrenal, including multiple--55%, border-line chromaffinoma--34% and malignant--11% (total--53 tumors). Ultrastructurally, malignant chromaffinoma showed pronounced aniso- and poikilocytosis, lack of desmosomes, presence of dark and clear cells, irregular and bizarre-shaped nuclei and enlarged nucleoli. An inverse correlation between the level of secretory granules and that of organelles was established for all sites and patterns of growth. The data obtained suggest certain ultrastructural peculiarities which alongside with macro- and microscopic features serve identification of malignity of tumor, functional status of tumor cells and type of secretion.

[Tolerance of different cell types for hypotonic action].

The behavior of cells with different structure of cytoskeleton was studied in the hypotonic media: protoplasts, embryonal mouse fibroblasts and transformed mouse fibroblasts (L-line). Protoplasts were most sensitive to the hypotonic media. They began to disrupt in the hypotonic medium 4:1, and in the hypotonic medium 1:7 a complete lysis of cells was observed. Transformed mouse fibroblasts were disrupted in the medium diluted 1:15, while embryonal mouse fibroblasts were not disrupted in the medium diluted 1:31. Moreover, in the hypotonic conditions an essential difference was observed between the cells studied. Embryonal mouse fibroblasts are more tolerant to the hypotonic conditions than L-cells and protoplasts. It is suggested that the cytoskeleton may define the cell tolerance in hypotonic media.

[Prognostic value of dark cells and desmosomes in squamous cell carcinoma of the lung].

An electron microscopic study of 30 cases of squamous cell carcinoma of the lung of varying degree of cell differentiation was aimed at evaluating prognostic significance of ultrastructural features of tumor. A high level of dark cells was found to be associated with poor prognosis whereas the presence of well-formed and multiple cellular contacts (desmosomes) was in direct correlation with survival.

[Electron microscopic characteristics of oncocytoma of the lung, small intestine and adrenal gland].

Electron microscopic characteristic of 3 oncocytomas (of lung, small intestine, adrenal) is presented. All the tumours had a similar ultrastructure with a large number of mitochondria in their cytoplasm. The electron microscopic study confirmed the diagnosis of oncocytoma in 2 cases. In one case the diagnosis of oncocytoma was made on the basis of the electron microscopic examination.

[Adaptive properties of the cultured cells from a pig embryo kidney exposed to hypotonic media].

A high tolerance of the cultured SPEV-cells was found under vital examination of their behaviour in hypotonic conditions. The dilution of cultured medium 1:1 hardly affects the cell morphology. With a 1:3 dilution, some minute alterations in the cells were noticed associated with the cell shape and the structure of the nucleus and of the cytoplasm, which later gradually disappear. A marked tendency of morphological alterations in the cells increases with increasing the medium dilution (1:7 and 1:15), however no cell destruction was observed. The complete lysis of the cells occurs only in the medium diluted by 32 times (1:31).

[Functional evaluation of cultured cells under hypotonic conditions by intravital staining with neutral red].

A high stability of pig embryo kidney-cells to hypotony was shown with neutral red vital staining. In the medium, diluted 1 : 1, the cells kept their ability to make granules during 6 days. In the medium diluted by water 1 : 3, both granular and diffuse staining of cells was shown within 6-12 hours. Later (18-48 hours) the cell ability to make granules was kept, but the neutral red was deposed as large vacuoles. In the normal medium the cell ability to make granules was kept too. In the medium, diluted by water 1 : 7, the cells lost their ability to make granules. The ability was recovered when cells were returned to the normal medium. Thus, after 6-18 hours of incubation in such a hypotonic medium, the cells produced small granules of neutral red, and 24-48 hours later they made large granules.

[Electron microscopic and biochemical characteristics of the hormonal activity of small-cell lung cancer].

An electron microscopic study of small-cell carcinoma of the human lung showed that tumors identified by light microscopy as small-cell lung cancer were, in actual fact, histologically heterogenous, ranging from those free from any signs of specific cell differentiation, neoplasms with endocrine differentiation, squamous cell cancer, adenocarcinomas to tumors with mixed cell differentiation. Elevated blood-ACTH concentrations occur in patients with tumors which do not incorporate cells with endocrine differentiation. In such cases, activation of ACTH synthesis may be due to hyperfunction and hyperplasia of apudocytes (elements of a diffuse endocrine system--the APUD system) of the lung or other organs in response to tumor growth.

[Correlation between the electron microscopic picture of small cell cancer of the lung and treatment results].

Electronmicroscopic study of 35 small-cell lung carcinomas showed these to be a group of lung carcinomas having different histogenesis (cytogenesis). Apart from carcinoma without signs of differentiation, this group also includes squamous-cell carcinoma, adenocarcinoma, apudoma and mixed carcinoma with various differentiation of tumour cells. A certain correlation between the ultrastructural features of the tumour and its susceptibility to the treatment is established. Small-cell carcinomas consisting of undifferentiated cells are most susceptible. A relatively good prognosis can be expected when a variant with the endocrine differentiation of tumour cells is treated. Tumours interpreted light microscopically as small-cell carcinoma but ultrastructurally identified as having squamous-cell differentiation, or as adenocarcinoma and mixed carcinoma have the least favourable prognosis after treatment.

[Electron microscopy in the diagnosis of small cell lung cancer].

The paper deals with an ultrastructural classification of small cell lung cancer distinguishing between wholly-undifferentiated tumors which do not contain cells with organ-, tissue- or cytospecific features (group 1) and tumors which incorporate both undifferentiated and differentiated cells (group 2). Depending on certain cytospecific characteristics, group 2 tumors histologically identifiable as small cell lung cancer may prove to be endocrine cancer, squamous cell cancer, adenocarcinoma or a mixed type tumor incorporating differentiated cells of two or more patterns. There is a correlation between the ultrastructural features of small cell lung cancer and its response to radiation and chemotherapy.

[Mechanisms of cellular multinucleation as affected by cytochalasin B in a culture of transformed fibroblasts].

The multinucleation of cytochalasin B was studied by means of phase contract microscopy and time-lapse cinematography. Several types of alteration of normal cytokineses resulted in the formation of multinucleated cells. The main way of multinucleation consists in the complete development of a cleavage furrow with the formation of daughter cells and an intercellular bridge that is followed by the fusion of these cells due to extending the entercellular bridge. These results suggest that the terminal stage of cytokinesis is the most sensitive to cytochalasin B.

[Ultrastructural variants of the cellular composition of human pulmonary adenocarcinomas].

Fifty-four lung adenocarcinomas of various degrees of differentiation were studied electron microscopically. Two groups of cells-differentiated, with the ultrastructural organ- and tissue-specific features, and undifferentiated, without such ultrastructural features were distinguished as a result of this study. Differentiated tumour cells may have ultrastructural features of pneumocytes II (osmiophilic membranous bodies), goblet cells and serous cells (mucous or serous granules), cylindrical cells (microvilli, cilia), squamous epithelium (tonofibrils, desmosomes) and possibly endocrine type cells (endocrine secretory granules) and pneumocytes I (pinocytotic vesicles). The quantitative correlation between differentiated and undifferentiated cells significantly varies with adenocarcinomas, in some tumours only undifferentiated cells being present. One or several cell types may be found in one tumour. There is a correlation between the degree of tumour differentiation at the histologic and ultrastructural level.

[Multiple primary carcinoid tumor of the stomach of complex structure].

The description of a polymorphic primary multiple stomach carcinoid tumour in a male of 36 is given. The tumour consisted of the nodes showing leiomyolike (spindle cell), pericytic, acinar-trabecular and solid structure; some cells resembled goblet cells. The diagnosis was precised by electron microscopy of the material fixed in formaldehyde in the course of which the neurosecretory granules in the tumour cell cytoplasm were revealed.

[Ultrastructural classification of tumor cells (based on the example of human squamous cell lung cancer)].

Ultrastructural classification of tumour cells of human neoplasms is suggested on the basis of ultrastructural examination of 110 lung squamous cell carcinomas. All tumour cells may be subdivided into two groups: the tumour cells of the first group show the ultrastructural organ- and (or) tissue specific features; the tumour cells of the second group do not exhibit such features. The tumour cells of the 1st group may be represented by one, two or several different types. The tumour cells of both groups, depending on the intracellular organelles development, may belong to one of the following three types: 1) rich in organelles, 2) with moderate number of organelles and 3) poor in organelles. All the tumours on the whole should be divided into three varieties: the first variety--the number of the 1st group cells dominate; the second variety--the number of cells of the 1st and the 2nd group is the same; the third variety--the 2nd group cells dominate. The structural-functional alterations of tumour cells (change of nuclei and other organelles) should be fixed in every case. The proposed classification reflects the diagnostic needs of electron microscopy.
