Studies of erythrocyte sequestration. II. Liver homing.

Xenogeneic and enzyme-treated erythrocytes rapidly home to the rat liver as measured by scintigraphic procedures. Histology and scanning electron microscopy revealed that pronase digested erythrocytes are taken up as whole cells by liver histiocytes.

Therapy of androgenetic sympatomatology with cyproterone acetate and ethinyl estradiol.

The effectiveness of therapy with cyproterone acetate and ethinyl estradiol was studied in 103 women. Acne and seborrhea responded best with 91.7 and 93.3% respectively, including complete and partial therapeutic success. For hirsutism complete remission and partial improvement were found in 75.3% of the treated women. Under therapy, body weight did not change in 51.9%, while 24.7% of the patients gained weight and 23.4% lost weight. The cycle length remained normal after therapy in 35.8%. Normalization or improvement was found in 54.7%. In 6.3% no improvement was noted after therapy and in 3.2% cycle irregularity developed in women with previous undisturbed pattern. According to BBT, improvement of the functional capacity of the reproductive system was found in 32.9% of the patients. Only 3% of the women studied demonstrated a deterioration. The 17-ketosteroid excretion was diminished in 35.3% after therapy and remained unchanged in 64.7%. The therapeutic regimen used for the study was well tolerated and good cycle control was obtained.

Effect of PTH on osteocyte ultrastructure.

Osteocyte ultrastructure was studied in the cortical bone of the tibia of rats after acute or chronic administration of supraphysiological doses of PTH. Confirming previous reports, an increase in the width of the cytoplasm with the appearance of numerous thin cytoplasmic processes, an increase in rough ergastoplasmic reticulum and Golgi apparatus, an increase in lacunar width and lysis of the lacunar wall ("brush border" after Bonucci) were observed. Particularly striking was the appearance of numerous microfilaments and microtubules in the cytoplasm of activated osteocytes. The appearance of microfilaments, often densely packed in cytoplasmic processes or running parallel to the plasma membrane, points to a role of the cytoskeleton in mediating the effects of PTH on conformational changes of the plasma membrane (and possible on cell motility); microtubules were particularly prominent in the Golgi field and are presumably involved in the exocytosis of lysosomes. Another striking feature was the non-random distribution of periosteocytic osteolysis along the lacunar perimeter. Osteolysis was particularly pronounced at the cell pole opposite to the cell nucleus. After chronic administration of PTH, autolysis of osteocytes, associated with signs of excessive periosteocytic osteolysis, was frequently encountered.

Dose-response study with a new LH-RH analogue, D-Ser (TBU)6 LH-RH 1-9 (EA)10 during the follicular phase of the menstrual cycle.

D-Ser (TBU)6 LH-RH 1-9 (EA)10 (HOE 766) a highly active LH-RH analogue, was studied with regard to its effects on the release of follicle stimulating hormone (FSH), luteinizing hormone (LH) and oestradiol-17beta (Oe2) during the follicular phase of the menstrual cycle. Forty-two regularly menstruating women were allowed to five different treatment groups with different doses (1.25 microgram; 2.5 microgram; 5.0 microgram; 10.0 microgram; 20.0 microgram) of HOE 766 given as intravenous bolus injections and the plasma concentrations of FSH, LH and Oe2 were measured up to 24 h after injection using specific radioimmunoassays. In the majority of cases, peak values of both FSH and LH occurred 4 h after injection being significantly different from pre-injection levels (P less than 0.02 in the 1.25 microgram treatment group, P less than 0.005 for the other treatment groups). Statistical analysis of maximum values as well as the absolute and relative increase in the different treatment groups revealed a dose-dependent effect of HOE 766. Maximum values of Oe2 occurred 8 h after injection and were found to be significantly different from pre-injection levels (P less than 0.005). However, no dose dependent effect was observed. It was concluded that HOE 766 is a potent and long-acting stimulator of FSH, LH and OE2 release in women. The effect of HOE 766 is dose dependent for FSH and LH but not for Oe2.

[The effect of low dose gestagens on the hypothalamic-pituitary-ovarian axis].

The contraceptive effect of continuous treatment with low dose progestogens (minipill) has been attributed mainly to alterations of the cervical mucus and the endometrium. This study was undertaken to investigate the effect of low dose progestogens on hypothalamic-pituitary-ovarian function. Plasma concentrations of follicle stimulating hormone (FSH), luteininzing hormone (LH), estradiol-17 beta (E-2) and Progesterone were measured daily during apparently ovulatory menstrual cycles and during treatment cycles with different low dose progestogens. From the results obtained it was concluded that the minipill has a clear-cut effect on the LH/FSH peak at midcycle and on corpus luteum function. A cyclic secretion of E-2 is maintained in the majority of cases. In a few treatment cycles however, follicular maturation was suppressed as indicated by low E-2 concentrations. It was concluded that the minipill exerts a profound effect at the central and ovarian level which contributes to its satisfactory contraceptive efficacy.
