Three different monoclonal rat antibodies, Acr1, Acr2, and Acr3, have been established against boar proacrosin. They are shown by enzyme-linked immunosorbent and immunoblot assays to react with boar proacrosin and several different acrosin molecules derived therefrom during activation. The epitopes detected by the three antibodies are different from each other, one being highly sensitive to reduction and periodate treatment. The antibodies crossreact with various proacrosin and acrosin molecules derived from human sperm extract; they also show indirect immunofluorescent staining of the acrosomal region of ejaculated sperm from normal men but fail to react with round-headed spermatozoa.
Mitoxantrone, a recent anthracenedione derivative is a potentially useful drug for direct intraperitoneal (i.p.) application because of its high tissue-binding and therapeutic index. We have carried out studies to establish maximum tolerated doses as well as pharmacokinetic studies with i.p. mitoxantrone in 21 patients (5 male, 16 female) with gastrointestinal (9), ovarian (6), unknown (2) and other (4) primary cancers and peritoneal carcinomatosis. Increasing doses (10-40 mg/m2) were given i.p. every 4 weeks. Five partial remissions (2-8+ months) and 7 stable disease courses (2-6+ months) were achieved. A reduction or disappearance of ascites was seen in an additional 3 patients. Severe toxicity (leukopenia) was observed in 4 patients only after 35 mg/m2 and 40 mg/m2 i.p. Pharmacokinetic analysis using high-performance liquid chromatography yielded the following data: The mean ratio of area under curve peritoneal fluid to serum was 1,108. The peritoneal clearance ranged from 2 ml/min to 9,617 ml/min and the disappearance half-life from 0.6-28.9 h. Mean urinary excretion within 24 h was 0.42% of the i.p. dose. These data indicate that mitoxantrone is sequestered in the intraperitoneal tissue compartment and only slowly released. Based on the outcome of this phase-I study we recommend phase-II studies at a dose of 30 mg/m2 i.p., repeated every 3-4 weeks.

The incidence of newborn infections caused by group B streptococci (GBS) has risen significantly in recent years. In 1983 and 1984 ten GBS diseases of the newborns were registered in the Department of Neonatology of the University Hospital (Charité). This is a morbidity of 1.2 per 1,000 live births. Eight infants showed an early onset and 2 a late onset type. Three of the neonates died. These were term infants with a birth weight of more than 2,500 g. The isolated types in these cases were Ia, Ia/c and III. In 1983 and 1984 430 nonselected women between the 34th and 40th weeks of gestation were examined for GBS colonization. One rectal, vaginal and cervical swab was collected from each women and cultured both in a selective broth medium and a selective blood agar plate. The colonization rate was 11.4% in pregnant women. The most frequent colonized region was the rectum, followed by vagina and cervix. Cultures from infants were obtained from throat, left and right ear. 2.7% of the newborns were positive for GBS. The most frequent isolated types are III/R and Ia. In all cases, positive for maternal and neonatal colonization, the isolated types were identical.
From January 1975 to March 1985 37 newborns and infants with B streptococcal infections were treated in the Pediatric Hospital of the Karl Marx University in Leipzig. The percentage of B streptococcal infections of all neonatal bacterial infections rose in this time from 6.9% to 15.7%. There were 23 early onset type infections and 14 late infections; 12 of the 37 infants died. Septicemia and meningitis were the most important clinical features. The time of beginning of the clinical symptoms in the postnatal period and the results of serotyping suggest a connatal infection in most cases.

As so far a singularity among opioid peptides, bovine beta-casomorphin-5 has been shown to display pronounced cardiatic activities. The gene sequence of a human beta-casomorphin-5, including plasmid insertion and fusion protein expression and cleavage instrumentary, has been synthesized by a manual phosphoramidite procedure and cloned in a bacterial system.
The close linkage between hisI and mutations of the cysII group in Pseudomonas aeruginosa was confirmed by mapping experiments with new mutants. All mutants of the cysII group were blocked in the cysteine biosynthesis before sulfite (this include the mutant cys-5605 which had previously been mapped by Mee and Lee, 1969). Most of the sulfite requiring cys mutants belonged to this group. The farthest distance between two mutations of the cysII group was calculated by the Wu formula to be approximately 2.5 kbp. We, therefore, suggest that more than one gene of the cysteine pathway may be located within this region.
A temperature-sensitive mutant (dna-11) with the phenotype of a mutant defective in the initiation of DNA replication, was isolated from an Hfr-like FP2 donor of Pseudomonas aeruginosa. Reversion of its temperature-sensitive character was achieved by integrative suppression rather than by backmutation or an additional suppressor mutation. The dna-11 mutant proved to be helpful in stabilizing the Hfr status of the original host.
A genetic locus controlling the electrophoretic mobility of an acid phosphatase in the rat (Rattus norvegicus) is described. The locus, designed Acp-2, is not expressed in erythrocytes but is expressed in all other tissues studied. The product of Acp-2 hydrolyzes a wide variety of phosphate monoesters and is inhibited by L(+)-tartaric acid. Inbred rat strains have fixed either allele Acp-2a or allele Acp-2b. Codominant expression is observed in the respective F1 hybrids. Backcross progenies revealed the expected 1:1 segregation ratio. Possible loose linkage was found between the Acp-2 and the Pep-3 gene loci at a recombination frequency of 0.36 +/- 0.06.
An immunohistological method for detection of rat MHC antigens by unconjugated primary alloantibodies is described. The procedure is based on secondary reagents specific for an allotypic marker of rat kappa light chains and makes possible differentiation between specifically bound alloantibody and the ubiquitously distributed interstitial immunoglobulin in the section. This technique can be used to pre-select monoclonal antibodies to be coupled to biotin or enzymes for histological application.
