Cyproterone acetate (CA) was toxic for Didelphis virginiana at a dose range of 22-28 mg/kg IM. A loss of spermatozoal pairing int the cauda epididymidis did not occur until 2 wk after testicular degeneration caused by CA. The isomers of 1-amino-3-chloro-2-propanol hydrochloride (CL 88,236 and CL 88,237) were well tolerated by the opossum at doses up to 30 mg/kg IM. After 8 days of CL 88,236 and CL 88,237 treatment there was no effect on spermatozoal pairing in the cauda epididymidis or on the seminiferous epithelium.
Sperm without acrosome of two males--spouses of infertile marriages--were studied. No acrosome could be identified in the Papanicolaou stained smears of either patient. Electron microscopy study confirmed the absence of acrosome and post-acrosomal sheath. The physical and functional integrity of the sperm membrane was determined by vital staining technique (72% unstained) and hypoosmotic stress test (69% swollen). Acrosin assay showed a five-fold decrease, and proacrosin values were almost eight-fold lower than the normal values. Nonzymogen acrosin level was normal. Zone-free hamster oocytes were not penetrated by the acrosomeless sperm showing 46% progressive motility, but postcoital tests in both female spouses showed active sperm. The results would indicate that the sperm devoid of acrosome may be considered as a primary cause of male infertility.
A total of 50 Sprague-Dawley adult male rats were used to compare the effects of intratesticular injection of 0.2 ml of hyperosmolar solutions of glycerol, glucose and NaCl with or without simultaneous treatment with indomethacin or dexamethasone. The hyperosmolar solutions caused a characteristic focal lesion with destruction of central testicular tissue as early as 12 hr after injection, and an inflammatory leukocytic halo present throughout the first week after injection. Neither anti-inflammatory drug altered this lesion. We concluded that intratesticular sterilization by hyperosmolar solutions in not a viable means of male contraception.
