In vivo and in vitro synthesis of human rhinovirus type 2 ribonucleic acid.

HeLa cells infected with human rhinovirus type 2 synthesize a mixture of single-and double-stranded ribonucleic acid (RNA). The RNA synthesized by the membrane-bound RNA polymerase complex in vitro is also a mixture of single- and double-stranded RNA, whereas the deoxycholate-treated RNA polymerase complex synthesized only double-stranded RNA. Although twice as much cell-associated viral RNA is synthesized in vivo at 34 C than at 37 C, there is no difference in the rate of RNA synthesized in vitro at 34 C and 37 C by the polymerase complex. The RNA polymerase complex, after treatment with deoxycholate, sediments as a broad peak with an average sedimentation value of 120S.

Turnover of HeLa ribosomal RNA. The characterization of a class of RNA in HeLa cytoplasm derived from 28S RNA.

A class of RNA in HeLa cytoplasm with a sedimentation value of 22S is described. This RNA is a true cytoplasmic component and is not an artifact of cell rupture or of the method of RNA preparation. The 22S RNA sediments in a sucrose gradient with those ribosomal structures containing 28S ribosomal RNA (60S and 74S particles and polyribosomes). It has a base composition and methyl content similar to those of 28S RNA. The kinetics of formation of 22S suggest that it is not a direct product of transcription but is derived in vivo from "old" molecules of 28S RNA.

Transfer RNA: A comparison by gel electrophoresis of the tRNA in HeLa cytoplasm, HeLa mitochondrial fraction, and E. coli.

A class of transfer RNA separable from the majority of transfer RNA by gel electrophoresis has been observed in E. coli and in the mitochondrial fraction of HeLa cells but not in HeLa cytoplasm. The size of this class of transfer RNA, as judged by its mobility on polyacrylamide gel, is intermediate between the major part of the transfer RNA and 5S RNA. It exists in part associated with the ribosomes, and its amino acid acceptor activity is different from the major class of transfer RNA.
