T7 infection-dependent selective expression of cloned genes in P1-lysogenic Escherichia coli.

Expression systems based on the selective transcription of genes cloned behind a T7 promoter, by T7 RNA polymerase, display a non-negligible basal expression when the T7 RNA polymerase gene is present within the host organism before induction of the system. This is a problem, especially for cloning and controlled expression of genes toxic to the host organism. We have circumvented this problem by taking advantage of abortive T7 infection of E. coli (P1), in the course of which T7 RNA polymerase is synthesized but bacterial growth is not quantitatively impaired. We have tested this system with three reporter genes, the 6-phospho-beta-galactosidase gene of Staphylococcus aureus, the luciferase operon of Vibrio harveyi, and the rabbit beta-globin gene; we have found very low basal levels, while, upon T7 infection, transcription is at least as efficient as in other in vivo T7 RNA polymerase systems in use.

Dihydrolipoic acid activates oligomycin-sensitive thiol groups and increases ATP synthesis in mitochondria.

Investigations with dihydrolipoic acid in rat heart mitochondria and mitoplasts reveal an activation of ATP-synthase up to 45%, whereas ATPase activities decrease by 36%. In parallel with an increase in ATP synthesis oligomycin-sensitive mitochondrial -SH groups are activated at 2-4 nmol dihydrolipoic acid/mg protein. ATPase activation by the uncouplers carbonylcyanide-p-trifluoromethoxyphenylhydrazone and oleate is diminished by dihydrolipoic acid, and ATP synthesis depressed by oleate is partially restored. No such efficiency of dihydrolipoic acid is seen with palmitate-induced ATPase activation or decrease of ATP synthesis. This indicates different interference of oleate and palmitate with mitochondria. In addition to its known coenzymatic properties dihydrolipoic acid may act as a substitute for coenzyme A, thereby diminishing the uncoupling efficiency of oleate. Furthermore, dihydrolipoic acid is a very potent antioxidant, shifting the -SH-S-S- equilibrium in mitochondria to the reduced state and improving the energetic state of cells.

[Acute poisoning caused by the herbicide dichlorprop (preparation SYS 67 PROP)].

We report on the acute poisoning of a 5-year-old boy by the herbicide "dichlorprop". This preparation is described as an effective means of plant protection. The effects of the intoxication were established by changes observed in almost all of the child's organs. The predominant effect was haemolytic anemia. All the pathological parameters regressed as a result of symptomatic therapy. General conclusions are drawn about intoxication, especially by means of plant protection agents.

2,4-Dichlorophenoxyacetic Acid

[Conservative therapy of mandibular fractures in children with operatively secured splints].

In the treatment of mandibular fractures in children offering no adequate possibility for attaching splints to the teeth, we use prosthetic splints fixed with four circumferential wires. This monomaxillary fixation is sufficient to stabilize fractures of the mandibular anterior segment. Fractures situated distal to the canines require additional intermaxillary fixation, i.e. zygomaticomaxillary suspension of the mandible. When functional therapy is needed at the same time, we use an activator instead of the prosthetic splint. 59 patients treated in this way were reevaluated within a period of 10 years. The results of this reevaluation are reported.

[Neuromuscular diseases and respiratory insufficiency in newborn infants].

Myopathological examinations which served to gain information on the potential impact of neuromuscular disease on the syndrome of perinatal respiratory failure included 47 consecutively deceased premature infants and neonates. A definite muscular disease could not be safety diagnosed in any case. However, only a very small number of investigations and data on the initial (possibly prenatal) manifestations have been available up to now. We found a total of 11 cases with increased intrafibrillary fat deposits coupled with mitochondrial aggregations in three cases. In seven cases, we found a retarded maturation ranging from three to nine weeks. Focally manifest myopathic changes were recorded in three cases. In one case, a large-group atrophy was found which might indicate the existence of a defect in the region of the second motor neuron. Biochemical backup investigations are necessary to be able to assess the relatively high incidence of increased fat deposits. We consider myopathological examinations of premature infants who died of respiratory failure to be desirable because far more comprehensive studies are necessary to be able to assess the importance of perinatally occurring muscular changes for the diagnosis of neuromuscular diseases.

Stabilization of maxillary segments and dental arch after puberty, in cleft lip and palate cases.

To overcome the high tendency to relapse after orthodontic treatment in patients with cleft lip and palate, efficient stabilisation of the dental arch is essential. In patients with an alveolar defect, extended bridges are most suitable, because there is no need for further retention appliances, the functional and cosmetic results are acceptable and extensive prosthetic reconstruction can be avoided, regardless of the patients age.

[Psychiatric long-term hospitalization--and how does life go on?].

Problems of longterm psychiatric hospitalisation are represented at the example of the topical supply situation in the mental hospital Bezirksnervenklinik Haldensleben. By means of a shown catalogue of symptoms from longterm psychiatric hospitalised patients tasks of care and inferences are discussed under the aspect, that the living and therapy conditions respectively their structures of the psychical sick persons authenticately show reactions on the course of illness and attendance. A lot of graduated care-offers in form of a therapeutical network first of all open a social reintegration in regard of individual and reality. At the same time hence it follows the chance for modifications in the psychiatric hospital.

[Epidemiology of generalized periodontal atrophy].

1378 individuals of 15-44 years of age were examined for atrophic processes of the periodontia. 11% of the individuals showed typical inflammation - free gingival recession. Great importance must be attributed the McCall's festooms and the wedge shaped defects in cervical enamel, dentin or cement.

[Persistent nerve root compression symptoms following lumbar intervertebral disk operation caused by spinal neurofibrosarcoma].

Case history of a woman who was operated on a lumbar disc. After a mild improvement again radicular pain and neurological deterioration occurred and a second operation on a recurrence was performed. Intraoperatively a malignant tumour (neurofibrosarcoma) within the root sheath reaching into the pelvis was found. In spite of laparatomy and radiotherapy this patient died five months later.

[Comments on so-called "chronic" and "therapy refractory" depression].

The problems of so-called "therapy resistant or chronic" depression has been investigated on own patients of the year 1986, but simply one chronic example could be noticed. Configurations-frequence-analytic research didn't essentially result in significant statements. It is referred to that the frequent occurrence of the so-called "therapy resistant" depressions described by some authors, could be in connection with limited diagnosis inventories and insufficient to the personality orientated therapy regime.

[Aggressive behavior in the psychiatric clinic].

Self and other directed violence of male psychiatric patients have been registered. The diagnostic analysis revealed an unequivocal correlation to delusional diseases and cerebroorganic hamper. Few patients caused a high percentage of all registered aggressions. Due to limited possibilities of drug treatment, changes of organization of communication and of the socio-organisational framework are aspired for the future.
