[The neurochemical and behavioral indices of an acquired alcohol craving in rats under conditions of information loading].

The state of neurotransmitter systems was studied in the groups of Wistar rats discriminated by striving for alcohol and rejecting it after the information load (alimentary instrumental conditioning in a labyrinth). The specific activities of neurotransmitter metabolizing enzymes (MAO A and B, acetylcholinesterase, and acetylcholinetransferase) and the content of biogenic amines and their metabolites (serotonin, 5-hydroxyindoleacetic acid, noradrenaline, and dopamine) were measured in homogenates and subfractions of sensorimotor cortex and caudate nucleus. It was found out that the biochemical indices correlated with cognitive abilities of animals. Stress-resistant rats, which were capable for acquisition of the complex skill, refused alcohol after the information load and were characterized by activation of the brain neurotransmitter systems. The rats, which were unable to fulfill the cognitive task, began to abuse alcohol and were characterized by suppression of the neurotransmitter systems. It seems possible that the brain neurotransmitter metabolism adequately reflects the characteristics of the higher nervous activity of animals and their resistance to alcohol.

[The experimental development of the concept of O. S. Adrianov on the correlation of functional and neurochemical processes: regulatory peptides in mediator system dysfunction].

The article is devoted to commemoration of full member of Russian Academy of Medical Sciences, Oleg Andreevich Adrianov, who would have celebrated his 75-th anniversary in 1998. O. S. Adrianov, author of numerous works on physiology and morphology of central nervous system, in the recent years of his was studying the problem of the processes relationship at macro and micro levels of brain organization. Further to the concept created by O.S. Adrianov, data on action of two peptides: delta-sleep and tafcine, on behavior, neurophysiological and neurochemical processes have been consolidated. Experimental data were obtained for rabbits, cats, and dogs, both intact and in the state of pathology (psychomotoric excitement, bradykinesia, penicillin epilepsy). Impact of peptides on convergation processes is discussed: peptide of delta-sleep depresses reactions of brain structures to photo- and phono-stimulation, and activates the serotoninergic system in general; tafcine enforces the convergation processes and activates the dopaminergic system.

[Biochemical characteristics of subcellular components of the visual system following light deprivation].

A study was made of the influence of prolonged light deprivation (ten weeks) at early stages of the rabbit postnatal ontogenesis on the state of specific enzyme activity (of cytochromoxidase, AChEase, K-Na-ATPase, monoaminoxidase and ATPase) in the synaptosomes and cellular mitochondria of the visual cortex and superior colliculi. A specific and oppositely directed change has been found ro each enzyme and subcellular structures. Restoration of sensory impulse activity in two weeks was attended with normalization of the revealed metabolism changes, likewise dissimilar for different subcellular components.

[Effect of light deprivation on enzymic activity of synaptosomes and mitochondria of rabbit cortex visual region].

The specific activity of markers-enzymes in the subcellular fractions of the rabbit visual analyzer cortical end, the synaptosomes and mitochondria of nerve cells, changed under the effect of early long deprivation. For cytochromoxidase and Na+, K+-ATPase it lowers considerably in all subfractions, for monoaminoxidase and Mg2+-ATPase it rises mainly in synaptosomes; the activity of acetyl cholinesterase lowers per 1 g of tissue. In the light two weeks later a tendency is observed to normalization of the studied indexes. The specific activity of cytochrome oxidase (except for free mitochondria) and Na+, K+-ATPase reaches the control, that of monoaminoxidase also partially normalizes, but not competely; Mg2+ATPase in all the subfractions is more inhibited than in the control. This evidences for the effect of light deprivation on the activity of the enzymes associated with different cycles of metabolic processes, first of all, of oxidation and ion transport. These changes are reversible when visual impulsation is recovered. Disturbances in chemism at the subcellular level are specific for different enzymic systems and are not the same in certain subfractions of great hemispheres.

[The effect of tuftsin on various aspects of metabolism in the sensorimotor system of the animal brain].

Rates of synthesis and degradation of monoamines, acetyl choline and protein were distinctly altered in cortex-subcortex structures of the brain locomotory system after a single administration of tetrapeptide tuftsin Thr-Lys-Pro-Arg during the period 15-30 min of its pronounced effect on the animal motor activity. Correlation between the functional and morphological alterations in cellular and subcellular brain structures, caused by the peptide administration, is discussed.

[Changes in the functional parameters of the brain structures under the influence of enkephalin-like tetrapeptidamide].

In complex neurophysiological and cytobiochemical study single injections of tetrapeptide amide (TPA) caused a short-term analgetic effect which manifested itself in the absence of motor reactions and EEG changes of cortical and subcortical brain structures after painful stimulation of extremities. This effect was accompanied by changes of some indices of transmitter (monoamine oxidase) and protein metabolism in the cerebral hemispheres at cellular and subcellular levels. In 30-40 min after a TPA injection, EEG suppression and absence of EPs to light flashes were observed in cortical and subcortical structures. Simultaneously motor disorders developed. The observed EEG changes had an undulatory character: on the second day EEGs were restored and on the third day--suppressed once again. This period of TPA action was accompanied by varied changes of the investigated types of metabolism. The question of the necessity of systemic approach to the study of TPA action is discussed, as such an approach allows to reveal complex neurophysiological and fine biochemical relations in the reactions of brain structures and in animal behaviour.

[Effect of light deprivation on GABA metabolism in subcellular fractions of the rabbit visual system].

Specific activity of glutamate decarboxylase was decreased in subfractions of light and heavy synaptosomes, and gamma-aminobutyric acid (GABA) transaminase activity--also in subfractions of free mitochondria, isolated from visual zone of brain cortex and of anterior mesencephalon but not from locomotive region of brain cortex of the light-deprived rabbits. Decrease in the ratio GABA transaminase/glutamate decarboxylase indicated distinct inhibition of GABA degradation as compared with its synthesis due to absence of sensory impulsation within the early periods of postnatal ontogenesis of the animals.

[Bioelectric activity and metabolism of individual brain structures at different times after administration of synthetic tetrapeptidamide].

Complex study of the opioid tetrapeptide amid (TPA) effect on metabolism and neurophysiological processes of different cortical areas, caudate nucleus n. accumbens allowed to detect prolonged (several days) specific action on these structures. The influence was manifested in certain changes of transmitter metabolism, background bioelectric activity, as well as in changes of EEG reactions to adequate afferent signals. The duration of the detected changes significantly exceeded the duration of anti-pain TPA effect.
