Rebound responding following a single dose of drug using an amphetamine-vehicle-haloperidol drug discrimination.

The purpose of this research was to characterize drug-induced rebound cue states using a three-choice, agonist-vehicle-antagonist drug-discrimination procedure. Rats were trained to discriminate among 0.50 mg/kg amphetamine (AM), distilled water (DW), and 0.03 mg/kg haloperidol (HA) in a three-lever drug discrimination task. Time-dependent changes in cue state were assessed following single doses of AM (5 and 10 mg/kg), HA (1 mg/kg), and cocaine (30 and 40 mg/kg). Consistent with expectations derived from the results of a study that used a two-lever AM-HA discrimination task, single doses of AM produced rebound responding on the HA-appropriate lever that was dose-dependent and peaked at 24 h following administration. In addition, cocaine substituted for AM at 0.5-2 h post-injection and then produced HA-like rebound responding that peaked at 24-36 h post-administration. Contrary to expectations, however, rebound AM-like responding did not occur following HA administration. Perhaps two- and three-choice discrimination tasks differ in their ability to characterize qualitative aspects of the post-haloperidol cue state. Knowledge of the time course of drug-induced adaptive processes, measured as withdrawal in the present research, is necessary for a complete description of a drug's effects and is important in understanding the effects of repeated drug administration.

Evidence suggesting a point source exposure in an outbreak of bovine abortion due to neosporosis.

A Holstein dairy farm suffered an abortion outbreak due to neosporosis. Abortion losses were > 18%. Cows with the highest Neospora antibody titers were at the greatest risk of aborting. Mummified fetuses were found after the 43rd day of the outbreak. The epidemic curve was suggestive of a point source exposure, which is consistent with the hypothesis that Neospora can be spread by a definitive host.

D2-specific discriminative stimuli: parameters, blocking, and rebound.

This study characterizes the cue properties of quinpirole (LY 171555), a selective D2 agonist, and the blocking capabilities of spiperone, a selective D2 antagonist. After rats were trained to discriminate 0.025 mg/kg quinpirole from distilled water, a dose-response curve and time course of the quinpirole discriminative stimulus were determined. The effectiveness of three doses of spiperone in blocking the discriminative stimulus produced by 0.02 mg/kg quinpirole was then assessed. Finally, the time course of spiperone's blocking action was determined. Given the putative selective action of these drugs on D2 receptors and the parametric data presented here, it was predicted that following chronic treatment with spiperone, a rebound increase in quinpirole-appropriate responding would occur. Neither chronic treatment with spiperone nor chronic treatment with haloperidol produced the predicted changes. This result, however, may be confined to the specific dose and time parameters used.

Drug discrimination is a continuous rather than a quantal process following training on a VI-TO schedule of reinforcement.

Debate continues as to whether drug discrimination in animals is an inherently quantal or continuous process. This issue is important in determining the appropriate interpretation of results from drug discrimination studies designed to assess the nature of drug-induced interoceptive cues. The quantal approach holds that subjects perceive a drug cue in an all-or-none manner, while the continuous view proposes that when appropriate training and testing procedures are used, subjects can discriminate along a continuum of interoceptive cues. Data consistent with the quantal view have consistently been generated by animals trained to respond on schedules of reinforcement having an FR component. Since quantal responding is a characteristic of these schedules, results from drug discrimination studies using training schedules with FR components are of little value in empirically determining whether drug discrimination reflects a quantal or continuous process. Use of variable schedules of reinforcement might be more appropriate because the pattern of responding generated does not preclude results consistent with either of the competing views. Data from the following studies that trained subjects using VI schedules with a concurrent TO for incorrect lever responding were analyzed: Barrett et al. (1982): L-5-hydroxytryptophan versus saline; Smith (1990): diazepam versus pentylenetetrazol; Barrett et al. (1992): amphetamine versus haloperidol; Barrett and Steranka (1983): amphetamine versus haloperidol. In every case, when experimental conditions produced a group mean intermediate to that for the training drugs, the distribution of scores for individual animals was normally rather than bimodally distributed.

Relation among body condition score, serum urea nitrogen and cholesterol concentrations, and reproductive performance in high-producing Holstein dairy cows in early lactation.

Body condition scoring (using a 5-point scale with quarter-point divisions) was performed on 66 Holstein dairy cows that began their second or later lactation in August, September, or October 1988. Cows' body condition was scored beginning on postpartum day 4 (+/- 1) and subsequently at postpartum days (+/- 1) 18, 32, 46, 60, 73 and 87. Blood samples were obtained on the same dates. Reproductive health examinations were conducted by 1 of 2 veterinarians beginning at postpartum day 21. Reproductive performance was evaluated in relation to body condition score and serum urea nitrogen and cholesterol concentrations. Number of days to first recorded signs of estrus and first breeding were not related to body condition score at calving, amount of condition loss, cumulative 80-day milk yield, or 305-day fat corrected milk yield. Cows that calved with body condition score greater than or equal to 3.50 required more days to conceive. Cows losing greater than 0.75 points of condition had longer days of conception. Body condition score at calving and amount of condition lost were not related to services per conception or diagnosis of follicular cyst. Cumulative 80-day milk yield was not related to days to conception or services per conception. Cows that produced greater than or equal to the mean 305-day milk yield required more services and had longer days to conception than cows that produced less than the mean 305-day milk yield.(ABSTRACT TRUNCATED AT 250 WORDS)

Relation among body condition score, milk production, and serum urea nitrogen and cholesterol concentrations in high-producing Holstein dairy cows in early lactation.

Body condition scoring (using a 5-point scale with quarter-point divisions) was performed on 66 Holstein dairy cows that began their second or later lactation in August, September, or October 1988. Body condition was scored, beginning on postpartum day 4(+/- 1) and subsequently at postpartum days (+/- 1) 18, 32, 46, 60, 73, and 87. Blood samples were obtained on the same dates. Kilograms of milk produced per cow was measured daily. Body condition score and changes in body condition score were evaluated in relation to daily milk production, cumulative 80-day milk yield, and serum urea nitrogen and cholesterol concentrations. Average daily milk production during week 1 was indicative of cumulative 80-day production, but not of 305-day milk yields. Cows that calved with body condition score greater than or equal to 3.50 did not differ in average daily milk production, cumulative 80-day milk yield, or 305-day milk yield, compared with cows that calved with body condition score less than 3.50. Cows that calved with body condition score greater than or equal to 3.50 lost more condition than did cows that calved with body condition score less than 3.50. Body condition score at calving and amount of body condition loss interacted with the rate of change in daily milk production. Serum urea nitrogen concentration did not differ for cows grouped by cumulative 80-day milk production or for cows grouped by amount of condition loss. Serum cholesterol values were higher than previously reported values and increased directly with milk production. Serum cholesterol values were inversely related to condition loss but changes in cholesterol concentration were not related to condition loss.

Nonlinear pharmacokinetics and conversion to glucose of intravenous sodium propionate in dairy cattle.

Three doses of sodium propionate (.75, 1.5, and 3.0 mmol/kg) were administered intravenously to 6, 8, and 14 dairy cows. Using first order kinetic analysis, the apparent plasma half-life increased significantly with increasing propionate dose. The apparent increase of propionate half-life with increasing propionate dose was attributed to saturation of uptake and disposal mechanisms. Using the nonlinear mathematical model of Henri-Michaelis-Menten for propionate concentrations at 3.0 mmol/kg, propionate half-life was significantly shorter than that obtained with the first order kinetic model. The Michaelis constant was 4.0 mM, the maximal rate of concentration decrease was .55 mM/min, half-life was 4.8 min, and distribution volume was .37 L/kg. Plasma glucose concentrations increased following all doses of propionate. The maximal increase in glucose concentration occurred earliest for the lowest dose and latest for the highest dose and increased in magnitude with increasing propionate dose. The plasma glucose response to intravenous propionate has been suggested as a measure of liver function in ruminants. Of the three propionate doses tested, the 3.0 mmol/kg dose appeared to saturate the uptake and disposal mechanisms of healthy liver and should be the most satisfactory dose for observing the plasma glucose response to injected propionate.

Hepatic morphology and effects of intravenous injection of sodium propionate on plasma propionate and glucose in fed and fasted dairy cattle.

Sodium propionate (3 mmol/kg) was injected IV into 8 nonlactating dairy cows before and after 6 days (144 hours) of fasting. During fasting, long-chain fatty acids in plasma increased from 0.30 +/- 0.05 (SE) mM to 1.09 +/- 0.15 mM (P less than 0.05). Liver fat increased from 0.5 +/- 0.3% to 9.3 +/- 1.7% (P less than 0.05). Half-life of injected sodium propionate increased significantly (P less than 0.05) from 7.6 +/- 0.5 minutes to 10.1 +/- 1.0 minutes during fasting. Sulfobromophthalein half-life did not change significantly (3.8 +/- 0.79 minutes to 5.3 +/- 1.3 minutes). Increases in plasma glucose concentrations after propionate loading were significantly less during fasting than during feeding. Thus, the change in glucose concentration served as an indicator of hepatic conversion of propionate to glucose. Increases in glucose concentration of less than 2 mM at 30 minutes after propionate loading indicated that liver function was altered in nonlactating dairy cows.

Factors associated with neonatal lamb mortality.

Three factors were associated with lamb neonatal mortality: birthweight (P<0.003), number of lambs born per ewe (P<0.001) and lamb sex (P<0.32). Lamb birthweight had the greatest predictive power for survival during the neonatal period. The neonatal mortality rate was 14.3%. The age specific mortality for lambs one day old was 7.9% (P<0.05). Seventy-nine percent of the lambs that died, did so by the end of the fourth post-natal day. Starvation was associated with 58.3% (P < 0.05) of the lamb deaths.

Patterns of preweaning diarrhea in piglets on a central California ranch.

To determine risk factors for development of diarrhea in litters of preweaned piglets, data were analyzed from records of 4,397 litters of swine farrowed on a central California ranch 1978-1982. Sixty-four percent of litters were treated for diarrhea. Statistical analysis by multiple logistic regression showed that litters born to young sows (parity less than or equal to 2) were 1.7 times more likely to develop diarrhea before weaning than were litters born to older sows (parity greater than or equal to 3) and that litters born on one end of farrowing barns were 1.3 times more likely to develop diarrhea than were litters born in the other end. The association of diarrhea with other variables--dam breed, sire, gestation group, gestation length, size of litter, number of mummies, runts or stillbirths per litter, farrowing barn of birth or day of the week farrowed--was weak or nonexistent (P greater than 0.01).

Aspects of bacteriology and endocrinology of cows with pyometra and retained fetal membranes.

In an effort to clarify the pathogenesis of pyometra, 20 cows with retained fetal membranes and 20 without, but with contemporary calving dates were studied. They were palpated and their uteri were subjected to sample collections for bacteriologic cultural examinations twice weekly for 4 weeks. Blood samples were obtained each day and evaluated for serum progesterone concentration. Three cows without and 3 with retained fetal membranes developed pyometra during the study, resulting in 3 groups designated control (CON), cows with retained fetal membranes (RFM), and cows with pyometra (PYO). Bacterial isolations occurred less frequently in the CON group than in the PYO and RFM groups. Growth patterns of bacteria also varied between groups. Coliform and incidental bacteria disappeared from the uterus of the PYO group by the end of the 3rd week. In contrast, heavy growth of Corynebacterium pyogenes and gram-negative anaerobic bacteria developed during this same period in the PYO group. In cows with pyometra, the significant persistent pathogenic bacteria recovered were C pyogenes and gram-negative anaerobic bacteria, especially Fusobacterium necrophorum and Bacteroides melaninogenicus. Anaerobic bacteria were isolated simultaneously with C pyogenes in most cows of the PYO group, but less often in CON and RFM groups, and highest growth levels were present near the time of ovulation. Clinically, pyometra usually developed about 10 days after observation of concurrent ovulation and high growth levels of C pyogenes and gram-negative anaerobic bacteria. A hypothesis is presented for development of pyometra in the cow.

Serum progesterone concentration in pyometritic and normal postpartum dairy cows.

Two studies were conducted to evaluate the concentration of serum progesterone in pyometritic cows and relate it to palpation of ovarian structures per rectum . In Trial 1, serum samples from 34 pyometritic cows were assayed for progesterone. Mean serum progesterone concentrations were 6.8 +/- 0.7 ng/ml. In Trial 2, each of 54 pyometritic cows was paired with a control cow on the basis of days post partum (18-50 days). Mean concentration of progesterone was 9.7 +/- 1.0 ng/ml for the pyometritic cows and 5.7 +/- 0.8 ng/ml in control cows (P<0.005). Progesterone concentration was greater (P<0.005) in both groups of cows with palpable corpora lutea (CLs). Ninety-six percent of the pyometritic cows had palpable CLs compared to 57% of the control group. Comparing serum progesterone only in cows with a palpable CL, the mean concentration was still greater (P<0.005) in the pyometritic group (10.6 +/- 1.0 ng/ml) than the control group (6.6 +/- 1.0 ng/ml). Compatability of rectal palpation findings and concentrations of serum progesterone were 92.6% for the pyometritic group and 72.2% for the control group. Progesterone concentration increased (P<0.05) by increased days post partum in Trial 2 (n=54) but not in Trial 1 (n=23). In both Trials 1 and 2, uterine size due to pyometra increased (P<0.05) with increased days post partum. No other associations were found.

Bovine cystic ovarian disease: hereditary relationships and case study.

Cystic ovarian disease (COD) was studied in a closed 300-cow dairy herd, using Dairy Herd Improvement Association and individual health records for a 7-year period (1974-1980). There were 2,112 calvings by 649 cows during the period. Cysts were found in 130 cows. Of the 649 cows that calved, 110 (16.9%) produced at least 1 daughter in which COD developed. Two bulls sired 17.6% of the daughters with COD, which was more than anticipated inasmuch as they sired only 11.6% of all daughters (P less than 0.05). These bulls were removed from the breeding program in an attempt to reduce code in the herd.

Clinical and clinicopathologic features of transport tetany of feedlot lambs.

Clinical and clinicopathologic features of transport tetany in 9 feedlot lambs were investigated. The lambs became recumbent within 10 days after arrival in the feedlot. The mean serum calcium concentration, mean magnesium concentration, and calcium:phosphorus ratio were significantly (P less than 0.01) less in affected lambs than in 6 unaffected lambs from the same feedlot. The mean serum phosphorus concentration was significantly (P less than 0.05) greater in the group of affected lambs. High activities of serum glutamic oxalacetic transaminase, creatine kinase, and lactate dehydrogenase in the serum of affected lambs indicated muscle necrosis, which was confirmed by histologic examination. Blood pH, HCO-3, and total carbon dioxide were significantly greater in affected lambs (P less than 0.01) than in unaffected lambs.

Border disease virus isolation from postpartum ewes.

The prevalence of border disease virus in a flock of Targhee sheep in southern Idaho was determined by serologic and virologic techniques. Of 249 ewes, 73 (29%) were seropositive to the antigenically related togavirus, bovine seropositive to the antigenically related togavirus, bovine viral diarrhea virus (BVDV). Concurrent testing of sera from 337 neonatal lambs indicated that 172 (51%) were seropositive for BVDV. It was observed that a high percentage (35%) of the BVDV-seropositive lambs were derived from seronegative ewes. Immunotolerance of the ewe was considered as the explanation for the lack of serologic relationship between seropositive lambs and their seronegative dams. Eleven cytopathogenic viruses were isolated from nasal and vaginal specimens obtained from 80 clinically healthy ewes within 24 hours of parturition. The viral isolates were demonstrated to be antigenically related to BVDV by direct immunofluorescence. Six of the 11 isolates were obtained from BVDV-seronegative ewes.

Relationship of neonatal mortality in lambs to serologic status of the ewe for Toxoplasma gondii.

The indirect hemagglutination method was used to determine the seroprevalence of Toxoplasma gondii in a random sample (250) of lambing ewes from a large (4,226 ewes) western range flock. Agglutination (greater than or equal to 2+) at the 1:64 serum dilution was considered a positive reaction; the seroprevalence was 20.8%. The neonatal mortality for lambs born to seropositive ewes was 30.7%, which was significantly greater than that for seronegative ewes (13.6%). The average age at death for lambs born to seropositive ewes was 24.8 hours, which was significantly less than that for lambs born to seronegative ewes (31.2 hours).

Serologic prevalence of ovine progressive pneumonia in a western range flock of sheep.

Of 254 sheep examined for antibodies to the virus of ovine progressive pneumonia, 162 (63.8%) were seropositive. The mean age of the seropositive ewes was 4.9 +/- 1.8 years, which was significantly greater than that for the seronegative ewes (4.1 +/- 2.3 years). The age-specific prevalence increased with ewe age, from 25% among yearling ewes to 85% among 7-year-old ewes. Seropositive ewes produced more pounds of live lamb at birth than did seronegative ewes.
