[Clinical findings in vitamin D poisoning of swine].

Acute to subacute vitamin D toxicosis is described in two different fattening pig herds. A few days after admixing a new batch of vitamin/mineral-premix most of the fattening hogs developed anorexia, polydipsia and polyuria. Emesis was seen in some of the animals. About two weeks after consumption of the suspected feed two thirds of the pigs had a rough hair coat and a very poor weight gain. Because of the results of haematological and metabolic findings from blood and urine samples of diseased pigs a calcinosis and tubular nephrosis was diagnosed. Patho-histological findings of the euthanatized morbid pigs included severe chronic interstitial nephritis, glomerulonephritis and mineralisation of a number of organs. Some of the animals showed degeneration of the myocardium. Feeding two healthy pigs with the incriminated feed the disease was reproduced.

[Indirect detection of ovulation and fertilization in the dog by progesterone level testing].

In addition to already published data (Arbeiter et al., 1990) we could achieve important facts concerning the estimation of mating time of the bitch by measuring the progesterone (P4) level continuously and observing the clinical signs on a collective of 106 ambulant patients. We found that a P4-concentration of 5 ng/ml plasma and above is signaling ovulation or ova fertilisation; a sudden rise of the progesterone over 10 ng (Ovucheck), reflecting 19.2 ng/ml (Serozyme-Progesterone), turned out to be a diagnostic mark for determination of mating time. Copulation took place between the 9th and 19th day of heat. 89% of the controllable bitches (n = 79) conceive and birth occurred 61.4 days (mean) later. The litter size and the loss of dead born puppies (4.7%) was normal with regard to the pertinent races. Because of their easy handling the P4-test kits (Ovucheck; Serozyme-Progesterone) are of good use for the practitioner.

[Concentration of thyroglobulin, thyroxine and triiodothyronine in 3 different causes of death].

There are investigated the concentration of thyroglobulin, thyroxine and triiodthyronine in serum of heart blood from autopsies. The level of hormones depends from the reason of death. In comparison with values of healthy living individuals the postmortem concentrations of Tg, TT4 and TT3 are very different. Highered values of Tg were observed in cases of accident death, who died some days after the injuries. Thyroxin was higher in cases of hanging and TT3 in cases of hanging and unexpected natural death.

[Magnetic resonance rheography in comparison to Doppler ultrasonography in the assessment of mitral valve stenosis].

Ultrasound-Doppler-technique and heart catheterization today are used routinely for the evaluation and graduation of mitral valve stenosis. A new magnetic resonance-technique of intracardial blood-flow registration now allows the grading of mitral valve stenosis. Good reliability and practicability of the demonstrated method in comparison with the ultrasound-Doppler-technique is shown. Future development of an alternative, completing magnetic-resonance method for the evaluation of valvular heart disease and shunt diagnosis is expected.

[Copper poisoning of sheep after ad libitum supplementation of rations by a mineral feed].

Within 8 weeks 22 of 400 sheep died with symptoms of Cu intoxication. In the liver of 2 dead sheep Cu contents of 491 and 830 mg/kg dry matter were found. The concentration of copper in the concentrate and other feedstuffs (grass, hay, straw, kale, dried sugar beet pulp) could not explain the development of Cu-toxicosis. The mineral-vitamin-supplement contained 86.7 mg Cu/kg. Obviously the Cu intoxication of previously Cu loaded sheep was provoked by offering the mineral supplement ad libitum (with intervals of 3-4 days), the intake of this mixture reached values up to 400 g/animal and day. By renunciation of mineral supplements in the same flock 7 months later lambs died due to Cu deficiency (Cu content in the liver: 13-14 mg/kg dry matter).

[Behavior and prognostic value of QTc intervals in surface ECG in acute and chronic cerebral processes].

Abnormal ECG-patterns in patients with acute and chronic cerebral processes are well known; the prognostic value, however, is still uncertain. Therefore, we examined 12 conventional ECG-leads retrospectively (at the day of admission to hospital) with respect to heart rate, occurrence of heart rhythm disturbances, and repolarization abnormalities in 131 patients (pts) with acute strokes and in 116 pts with cerebral tumors. Patients with atrial fibrillation or bundle branch blocks were excluded. In all pts the longest QTc-intervals (heart rate correction according to Bazett) were found in the precordial leads V2-V6 and Nehb D, A, I. Patients with strokes had the longest QTc-intervals: 418 +/- 43 ms (II) - 445 +/- 55 ms (V5). The differences in healthy persons (n = 25) and pts with cerebral tumors were significant (p less than or equal to 0.01). Between the standard and precordial leads the differences in this group were also significant. The QTc-values in non-survivors were significantly longer (429 +/- 45 ms in lead I, 458 +/- 45 ms in V4) than in survivors (409 +/- 35 ms in lead I, 436 +/- 52 ms in Nehb A). Initially, unconscious pts (n = 22) also revealed significantly longer QTc-intervals (485 +/- 58 ms, Nehb D) than conscious pts (440 +/- 45 ms, lead V5). 78.6% of pts with strokes had QTc-values in at least one lead longer than 420 ms, and in 64%, we registered QTc-intervals greater than 440 ms. Significant differences were found between non-survivors (n = 56) and survivors (n = 75).(ABSTRACT TRUNCATED AT 250 WORDS)

Enzymatic recognition of DNA modifications induced by singlet oxygen and photosensitizers.

DNA modifications induced either by photosensitization (illumination in the presence of methylene blue) or by chemically generated singlet oxygen (thermal decomposition of an 1,4-etheno-2,3-benzodioxin) are recognized and incised by repair endonucleases present in crude bacterial cell extracts. Only a small fraction of the incised modifications are sites of base loss (AP-sites) sensitive to exonuclease III, endonuclease IV from E. coli or to the UV-endonuclease from M. luteus. Cell extracts from E. coli strains overproducing or defective in endonuclease III recognize the modifications induced by illumination in the presence of methylene blue just as well as do those from wild-type E. coli strains. This indicates that dihydropyrimidine derivatives, which are characteristic of hydroxyl radical-induced DNA modifications, are absent. In contrast, most of the modifications induced are not recognized by a cell extract from a fpg strain defective in formamidopyrimidine-DNA glycosylase FPG protein). Furthermore, incision by a cell extract from an E. coli strain overproducing FPG protein takes place at much lower protein concentration than with the wild-type strain. Experiments with purified FPG protein confirm that this enzyme is responsible for the recognition of singlet oxygen-induced DNA base modifications.

Extracorporeal gas exchange in adult respiratory distress syndrome: associated morbidity and its surgical treatment.

Extracorporeal carbon dioxide removal (ECCO2-R) over a membrane lung is a new therapy for patients with adult respiratory distress syndrome (ARDS) who frequently suffer from lung complications caused by long-term artificial ventilation and who may require major thoracic surgery. This is a report of 76 patients with severe ARDS who were treated by ECCO2-R. Twenty-six of these 76 patients required thoracotomy: 19 for pneumothorax and pneumatocele, and seven for haemothorax, infected lung necrosis or oesophagotracheal fistula. Most pneumothoraces were bilateral. Ten of these 26 patients required reoperation, usually for extensive persisting alveolar air leaks. Sixteen (62 per cent) of the 26 patients who had a thoracotomy and 22 (44 per cent) of the 50 patients without surgery survived. These results demonstrate that performing a thoracotomy, if necessary, does not diminish the survival chance of high-risk patients with severe ARDS.

Unusual transcription termination of the ribosomal RNA genes in Ascaris lumbricoides.

We studied termination of transcription of the ribosomal RNA genes in Ascaris lumbricoides, the first representative in the phylum of nemathelminthes analysed so far. RNase protection experiments in vivo reveal that the 3' end of the precursor rRNA coincides with the end of mature 26S rRNA. Promoter-containing miniplasmids are able to direct unique 3' end formation in vitro at a site identical to that observed in vivo, whereas deletion of these sequences abolishes 3' end formation throughout the entire spacer. A nuclear run-on experiment in vitro confirms the drop of polymerase I concentration down-stream of this site. The termination site for polymerase I transcription of the rDNA operon in A. lumbricoides is therefore unique, and located at the very end of the 26S rRNA gene.

[Thyroglobulin content of the blood in cases of hanging].

Thyroglobulin (Tg) was determined in serum of heart blood from the bodies of 24 people who had died by hanging. In contrast to cases of sudden death (Tg 22.7 +/- 13.5 ng/ml) the Tg values varied over a wide range (from 12 to more than 400 ng/ml). The highest concentrations were observed in cases of incomplete suspension, e.g. hanging in a sitting or semi-reclining posture. We conclude that the duration of the death agony has an influence on the Tg level in serum. Except of diseases of the thyroid gland (carcinoma, adenoma) a high level of Tg in serum may be a helpful adjunct in the diagnosis of mechanical forces to the neck region inflicted before death.

Beta-adrenergic receptors and adenylate cyclase activity in heart, muscle and adipose tissue of German Landrace pigs selectively bred for differences in backfat deposition.

The density of beta-adrenergic receptors was examined in heart, muscle (m. longissimus dorsi) and backfat tissue of German Landrace pigs using (-)-[3H]-dihydroalprenolol. The animals were selected for high (E(+)-line) or low (E(-)-line) activity of lipogenic enzymes in backfat and for low backfat thickness (U(-)-line) based on ultrasonic measurement. An unselected control (K0-line) also exists. The correlated selection response on stress susceptibility (measured by halothane reaction and by meat quality parameters) showed the following sequence of selection lines: U- greater than E- greater than E+ greater than K0. The strains used for examination descended from the 9th generation (heart and muscle tissue) and the 10th generation (heart, muscle and adipose tissue) of this selection experiment. No further selection took place between the 9th and 10th generation. Basal adenylate cyclase activity was determined as well as (-)-Isoproterenol + GTP or sodium fluoride (NaF) stimulated activity in heart and muscle of the 10th generation measuring [3H]-cAMP formation from [3H]-ATP. A good correlation (r greater than or equal to 0.59, n = 9) between beta-adrenergic receptor density and adenylate cyclase activity was found which pointed to an efficient receptor-effector system. The number of beta-adrenergic receptors and adenylate cyclase activity was higher in the U(-)-line in all investigated tissues compared to the other lines in the 9th and 10th generation. Most differences between these lines were also significant.(ABSTRACT TRUNCATED AT 250 WORDS)

[Combined surgical-antibiotic therapy of actinomycosis of the adnexa].

The majority of patients presenting with tubo-ovarian abscess caused by abdominopelvic actinomycosis are women using an IUD. Since the course of this disease is afebrile, and the diagnosis often established relatively late, it typically simulates a malignant tumour of the lower abdomen. Except for leukocytosis and an increased blood sedimentation rate, laboratory findings are within the normal range. The infection infiltrates neighboring structures, and hence a secondary involvement of the intestine is common. Two cases are presented, successfully treated by a combined operative-antibiotic approach.

Effects of urapidil, ketanserin and sodium nitroprusside on venous admixture and arterial oxygenation following coronary artery bypass grafting.

Thirty patients who developed arterial hypertension following coronary artery bypass grafting, despite sedation, were treated randomly with sodium nitroprusside (SNP), ketanserin or urapidil. All drugs significantly decreased arterial pressure. Two patients were withdrawn because hypertension failed to respond to ketanserin. Significant tachycardia was noted only in the SNP group. An increase in Q and significant decreases in systemic and pulmonary vascular resistances were seen in all groups. Following administration of SNP, (PaO2-PaO2) and Qs/Qt increased significantly, whereas PaO2 decreased significantly. Three patients were withdrawn from the SNP group because Qs/Qt was greater than 30%. (PAO2-PaO2) and Qs/Qt showed no significant changes following the administration of ketanserin or urapidil. These drugs may have advantages over SNP in the management of hypertension following coronary artery bypass surgery.

Effects of (22S,23S)-Homobrassinolide and Related Compounds on Membrane Potential and Transport of Egeria Leaf Cells.

(22S,23S)-Homobrassinolide was tested for its effect on the electric cell potential, proton extrusion, ferricyanide reduction, and amino acid and sucrose uptake of leaves of Egeria densa Planchon. In the light, (22S,23S)-homobrassinolide and its derivative, 2alpha-3alpha-dihydroxy-5alpha-stigmast-22-en-6-one, were similar to each other and similar to fusicoccin in causing hyperpolarization and proton extrusion, whereas stigmasterol was less effective. In darkness, the three sterols showed comparable effects. (22S,23S)-Homobrassinolide slightly stimulated ferricyanide reduction and promoted uptake of sucrose and alpha-aminoisobutyric acid. The results are compatible with a stimulation of an electrogenic proton pump mechanism at the plasmalemma by (22S,23S)-homobrassinolide.

Photobiological studies with dioxetanes in isolated DNA, bacteria, and mammalian cells.

1,2-Dioxetanes, efficient chemical sources of triplet excited carbonyl compounds, were observed to be genotoxic in isolated DNA, bacteria, and cultured mammalian cells. In superhelical DNA of bacteriophage PM2, various alkyl- and hydroxyalkyl-substituted dioxetanes (1) induced predominantly endonuclease-sensitive base modifications and only few single strand breaks. With a specific endonuclease a small fraction of the base modifications was identified as pyrimidine dimers. The psoralen dioxetane (2a) or PsD bound photochemically to calf thymus DNA at the alpha-pyrone ring of psoralen (fluorescence measurements). Photobinding was also observed when calf thymus DNA was incubated with psoralen and 3-hydroxymethyl-3,4,4-trimethyl-1,2-dioxetane. In Syrian hamster embryo fibroblasts and HL-60 cells, dioxetanes induced DNA single strand breaks. The alkyl- and hydroxyalkyl-substituted dioxetanes 1 and 2 were efficiently inactivated by cysteine, glutathione, ascorbic acid, tocopherol, NADH and FADH2. While dioxetanes 1 and 2 were not mutagenic in Salmonella typhimurium strain TA100, benzofuran dioxetanes 3 exhibited substantial effects. Further data imply that presumably a mutagenic intermediate with a lifetime of a few minutes is produced from the benzofuran dioxetane.

[Results of early education of developmentally delayed infants and young children by parents with a clinical rehabilitation program].

Early registration of disturbances in the development of infants was based on neurological examination. Since 1984 exists the possibility of psychological evaluation of infants and children in early childhood in the Department of Paediatrics of the University of Rostock. Parents of children suffering from disturbances of development receive additionally to the concept of physiotherapy an early intervention program to furtherance of children. In this way best conditions are prepared in development and evaluation of admission in special institutions (special kindergarten, day-nursery).

Prognostic significance of different QT-intervals in the body surface ECG in patients with acute myocardial infarction and in patients with acute or chronic cerebral processes.

In 73 patients after AMI, 131 patients with cerebral stroke and in 116 patients with cerebral tumours the QTc-intervals in different ECG leads were measured. In about 30% of pts with AMI there were prolonged (greater than or equal to 440 ms) QTc-values. Patients with AMI suffering from VT or non-survivors had the longest QTc-values. Also in patients with cerebral stroke and prolonged QT-values (greater than or equal to 440 ms) the risk of dying was significantly higher than in patients that survived. The prolongation of QTc in patients with cerebral tumours are not correlated with prognosis but with the severity of neurological disturbances.
