[Mixed organ culture as new possibility for study of cellular immune phenomena according to Krebs].

A method for the detection of cellular tumor-versus-host relations is described, which consist in cultivating human tumor material together with autologous lymphocytes in one organ system and in determinating tumor and lymphocyte DNA synthesis separately before and after mixed organ culture. Advantages and disadvantages of this method are discussed. 58 human tumors were investigated (40 mammary, 13 gastric, 5 bronchial cancers). 37 tumors showed a sufficient in vitro growth. In about 50% of the mixed organ cultures, there was an increase in lymphocyte DNA synthesis compared to the controls. On the other hand, only in 9 cases a decrease in the tumor DNS synthesis could be observed under the influence of autologous lymphocytes. The investigations emphasize the individuality of the tumor-versus-host relations and lead to the consequence of a routine determination of the immune profile by a spectrum of several methods in tumor patients as a prognostic and therapeutic aid.

[Contribution to stability test of daunoblastin solutions (author's transl)].

It is known that drugs are not unlimitedly stable. Dilutions of the antineoplastic agent daunoblastin used in the clinic and experimental work, were examined, whether they could be stored for several days. By means of impulscytophotometry, spectral-photometric measurements, counting of surviving cells and estimation of DNA-synthesis after culturing pieces of tissue in the presence of daunoblastin it could be established that this drug, diluted even four days ago, still possesses sufficient activity.

Mixed organ culture as a tool for considering cellular tumor-host relationships in tumor patients.

A method is reported for considering tumour-host relationships, which consists in cultivation of human tumours jointly with autologous lymphocytes in an organ culture system and determining separately tumour and lymphocyte DNA synthesis before and after mixed culture. Some experiments explore variables of the new system. For mixed organ cultures the best conditions are: 4-day incubation, lymphocyte DNA ratio to tumour cell DNA less than 9:1 (2-10(6) lymphocytes/ml) and autologous serum. So far 41 tumours (20 mammary, 17 gastric, and 4 bronchial carcinomas) have been examined. 22 tumours showed satisfactory in vitro maintenance. In 50% of the mixed organ cultures, lymphocyte DNA synthesis proved to be higher than in lymphocyte control cultures. Blast counts in the cell smears are in good accord with results obtained by biochemical determination of DNA synthesis. 7 of the tumours studied have shown a pronounced reduction of DNA synthesis in the presence of lymphocytes. The present results underline the individuality of tumour-host relationships and suggest the necessity of determinations of the immune reactivity in cancer patients. Advantages and disadvantages of the method are discussed.

Preparation, characterization, therapeutic efficacy and toxicity of liposomes, containing the antitumour drug cis-dichlorodiamineplatinum(II).

Cis-dichlorodiamineplatinum(II) (cis-DDP) was encapsulated in reverse phase evaporation vesicles (REV, 0.6 mg cis-DDP/ml lipid solution) and multilayered liposomes (MLV, 0.3 mg cis-DDP/ml lipid solution) with different cholesterol content. The identity of cis-DDP in free and encapsulated form was checked by various techniques. Particle size, homogeneity of liposomes and distribution of cis-DDP in REVs were shown by electron microscopy. The examination of entrapped cis-DDP in REVs relating to buffer and serum stability, in vitro and in vivo antitumour activity and nephrotoxicity proved that all points are strongly influenced by the cholesterol (CH) content. Enclosed cis-DDP in phosphatidyl (PC)-REV has the same, and in PC: CH-REV, a lower effect in vitro and in vivo compared to treatment with the free drug. Irrespective of the application of tumour cells and substance (i.v., i.p.) in optimal therapeutic doses, an equal increase in life-span (ILS) was registered with the free drug and with PC-cis-DDP-REV, while cis-DDP in PC: CH-REV had a significantly reduced effectiveness. Liposomal encapsulation of cis-DDP also influenced body weight change and leucocyte counts. The blood urea nitrogen (BUN) level, as an indicator of renal toxicity, was only moderately increased after very high doses of cis-DDP (24 mg/kg) in PC: CH-REV.
